Repertoire and abundance of secreted virulence factors shape the pathogenic capacity of Pseudomonas syringae pv. aptata

Project description:Pseudomonas syringae pv. aptata is a member of the sugar beet pathobiome and the causative agent of leaf spot disease. Like many pathogenic bacteria, P. syringae relies on the secretion of toxins, which manipulate host-pathogen interactions, to establish and maintain an infection. This study analyzes the secretome of six pathogenic P. syringae pv. aptata strains with different defined virulence capacities in order to identify common and strain-specific features, and correlate the secretome with disease outcome. All strains show a high type III secretion system (T3SS) and type VI secretion system (T6SS) activity under apoplast-like conditions mimicking the infection. Surprisingly, we found that low pathogenic strains show a higher secretion of most T3SS substrates (19 of the 23 detected effectors and accessory harpin proteins), whereas a distinct subgroup of four effectors is exclusively secreted in medium and high pathogenic strains. Similarly, we detected two T6SS secretion patterns: while one set of proteins was highly secreted in all strains, another subset consisting of known T6SS substrates and previously uncharacterized proteins with a highly similar secretion pattern was exclusively secreted in medium and high virulence strains. Taken together, our data shows that P. syringae pathogenicity is correlated with the repertoire and fine-tuning of effector secretion and indicates distinct strategies for establishing virulence of P. syringae pv. aptata in plants.

ClpV3 affects pathogenicity in Pseudomonas aeruginosa

Project description:ClpV3 is a cytoplasmic AAA+ ATPase protein and is an essential component of H3-T6SS in Pseudomonas aeruginosa. Here, we report that an H3-T6SS deletion mutant PAO1(ΔclpV3) significantly affected the virulence-related phenotypes including pyocyanin production, biofilm formation, proteolytic activity and motilities. Most interestingly, the expression of T3SS genes was markedly affected, indicating a strong link between H3-T6SS and T3SS. RNA-Sequencing was performed to globally identify the genes differentially expressed when H3-T6SS was inactivated and the results obtained could be well correlated to the observed phenotypes.

Novel insights into the genetic basis and proteotranscriptomic elucidation of virulence attenuation in Pseudomonas syringae pv. actinidiae variants

Project description:Pseudomonas syringae pv. actinidiae (Psa) is the causal agent of kiwifruit bacterial canker, but the factors affecting its pathogenicity in natural settings remain poorly explored. In this study, we isolated two Psa strains, G126 and G282 from infected kiwifruit orchards in Guizhou Province of China. Both isolates, categorized as Psa-biovar 3, were confirmed through Psa3-specific primers and phylogenomic analysis. Pathogenicity assays on kiwifruit cultivar ‘Hongyang’ leaves and branches showed significantly reduced numbers of necrotic spots and reduced lesions sizes upon infection with G282 compared to the G1 positive control strain, while G126 showed nonpathogenic phenotype. Additionally, both strains failed to induce a hypersensitive response in nonhost Nicotiana tabacum plants, and exhibited significantly reduced promoter activity of the hrpR/S, hrpL, and hrpA genes, which are crucial for the type III secretion system (T3SS). Genomic sequencing revealed that the T3SS of G126 was defective due to a single-nucleotide polymorphism in the hrpR gene, while G282 was entirely deficient in the type VI secretion system (T6SS), which potentially regulates the expression of T3SS genes. Transcriptomic analysis showed widespread alterations in key aspects of the secretion system, protein transport, and signal transduction, further supporting the phenotype characteristics exhibited by the strains. This study enhances our understanding of the genetic basis of nonpathogenic and partially pathogenic Psa isolates, highlighting the functional interdependencies between T3SS and T6SS.

Pseudomonas syringae Virulence Regulatory Network construction

Project description:Pseudomonas syringae, a Gram-negative plant pathogen, infects more than 50 crops with its type III secretion system (T3SS) and causes severe economic losses around the world. Although the mechanisms of virulence-associated regulators of P. syringae T3SS have been studied for decades, the crosstalk and network underlying these regulators are still elusive. Previously, we have individually studied a group of T3SS regulators, including AefR, HrpS, and RhpRS. In the present study, we found 4 new T3SS regulator genes (envZ, ompR, tsiS and phoQ) via transposon-mediated mutagenesis. Two-component systems EnvZ and TsiS natively regulate T3SS. In order to uncover the crosstalk between 16 virulence-associated regulators, (including AefR, AlgU, CvsR, GacA, HrpL, HrpR, HrpS, MgrA, OmpR, PhoP, PilR, PsrA, RhpR, RpoN, TsiR and Vfr) in P. syringae, we mapped an intricate network named PSVnet (Pseudomonas syringae Virulence Regulatory Network) by combining differentially expression genes in RNA-seq and binding loci in ChIP-seq of all regulators.

A serial sample processing strategy with improved performance for in-depth quantitative analysis of type III secretion events in Pseudomonas aeruginosa

Project description:Bacterial pathogens have the ability to secrete effector proteins to promote host cell invasion. The efficient analysis of the secretomes in close to wildtype cell systems is important to study the mechanisms of bacterial secretion efficiently at different levels. In particular the secretome analysis of bacteria that rely on rich media for optimal secretion may hamper analysis via modern sample preparation shotgun proteomics workflows due to higher degree of sample impurities. This is may be a reason for the low number of quantitative secretome investigations in such cells. We assessed the efficiency of different workflows and their amenability for secretome analysis including precipitation, SP3 and a combined, serial workflow. Using the model organism Pseudomonas aeruginosa, we found that, the combined TCA-SP3 strategy outperforms the other tested methods on all levels and proofed most efficient in the recovery of proteins related to the type III secretion system (T3SS), including all known effector proteins and secretion machinery components. Moreover, the serial workflows resulted in improved quantification accuracy using MS1 label-free quantification. Finally by monitoring the compositional changes of secretome samples over time we observed strong increase of the fraction made up by T3SS related proteins with parallel decrease of proteins not linked to T3SS. Our study shows how combining sample preparation procedures can lead to orthogonality in depleting impurities that result in better chromatographic peptide separation, and more efficient MS detection with improved quantification parameters.

Project description:Purpose: The outcome of host–pathogen interactions is thought to reflect the offensive and defensive capabilities of both players. When plants interact with Pseudomonas syringae, several well-characterized virulence factors contribute to early bacterial pathogenicity, including the type III secretion system (T3SS), which must be activated by signals from the plant and environment to allow the secretion of virulence effectors. The manner in which these signals regulate T3SS activity is still unclear. Conlusion: the analysis revealed that the perception of plant signals from kiwifruit or tomato extracts anticipates T3SS expression in P. syringae pv. actinidiae compared to apoplast-like conditions
