Project description:This study aims to identify the potential protein targets of Ginkgolic Acid in human hepatocellular carcinoma HCCLM3 cells using the Drug Affinity Responsive Target Stability (DARTS) assay combined with LC-MS/MS proteomics. Whole cell lysates from Ginkgolic Acid-treated and vehicle control (DMSO) HCCLM3 cells were subjected to limited proteolysis with proteinase K. The digested protein samples were separated by SDS-PAGE, and differential protein bands were excised, digested in-gel, and analyzed by mass spectrometry. The raw data will be used to identify proteins whose stability is altered upon Ginkgolic Acid binding, providing insights into its anti-tumor mechanisms.

Project description:Here we have compared the impact of loss the protein synthesis initiation factor eIF4e in non samll cell lung cancer cells. Expression of eIF4E was targeted by an eIF4E siRNA or by expressiojn of eIF4E-dTAG construct allowing degradation of eIF4e-dTAG with the heterobifunctional dTAGv-1 small molecule. Proteomes were profiled following 72 hr exposure of the four H1299 eIF4E-dTAG clones (C2-2, C3-1, N2-1 and N3-2) to 0.5 microM dTAGV-1 or H1299 parent cells transfected with 1 microg eIF4E siRNA (siTOOLS, Planegg, Germany) using Lipofectamine 3000 transfection reagent (Fisher Scientific, cat#L3000008).

Project description:DNA integrity is constantly challenged by both endogenous and exogenous damaging agents, resulting in various forms of damage. Failure to repair DNA accurately leads to genomic instability, a hallmark of cancer. Distinct pathways exist to repair different types of DNA damage. Double-strand breaks (DSBs) represent particularly severe form of damage, due to physical separation of DNA strands. Repair of DSBs requires activity of RNA Polymerase II (RNAPII) and generation of Damage-associated transcripts (DARTs). Here we show that the RNA m5C-methyltransferase NSUN2 localizes to DSBs in transcription-dependent manner, where it binds to and methylates DARTs. Depletion of NSUN2 results in an accumulation of nascent DARTs around DSBs, specifically of the de novo primary DARTs. Furthermore, we detected an RNA-dependent interaction between NSUN2 and DICER, which was stimulated by DNA damage. NSUN2 activity promoted Dicer cleavage of DARTs associated R-loops, which is required for efficient DNA repair. We report a previously unrecognized role of the RNA m5C-methyltransferase NSUN2 within RNA-dependent DNA damage response, highlighting its function as a DICER chaperone for the clearance of non-canonical substrates such as DARTs, thereby contributing to genomic integrity.

Project description:While minimizing lung cancer metastasis can improve overall survival, few models exist where subpopulations with stable high and low metastatic potential can be easily and reliably isolated. Our previous work characterized two stable subpopulations, termed Leaders and Followers, from the H1299 Non-Small Cell Lung Cancer model. H1299 Leaders and Followers have different transcriptional profiles, biomarkers, genomic methylation patterns, metabolic states, and morphologies.Of note, H1299 Leaders express high levels of Jagged1 (JAG1) and Myosin 10 (MYO10) and drive collective invasion and metastasis. To determine how JAG1 and MYO10 contribute to Leader cell biology and lung cancer metastasis, we utilized CRISPR-Cas9 to generate edited pools of H1299 cells targeting either JAG1 or MYO10. Fluorescence-activated cell sorting using a Leader-Follower biomarker, IL13R2, was used to generate single cell derived candidate clones. Candidate clones were screened using western blot for Leader and Follower cell biomarkers (JAG1, MYO10, HTATIP2 and IL13R2) followed by Sanger sequencing of candidate clones was also conducted to confirm target gene status. Using this method, 2 JAG1 knockout, 2 MYO10 knockout, 1 MYO10 knockdown and, 2 wildtype MYO10 JAG1 H1299 single-cell derived clones were generated.

Project description:In order to detect the difference of tumorigenicity induced by different cell lines in nude mice. We have employed whole genome microarray expression profiling as a discovery platform to identify the difference of genes expression. 10 week old 44 thymus-free nude mice were randomly divided into four groups, control group, A549 group, H1299 group and A549+H1299 group. 0.2ml saline suspension containing 5×10^6 cells were implanted subcutaneously in the flanks of each nude mouse in the control group. A549+H1299 group means 0.2ml saline suspension containing 2.5×10^6 A549 cells and 2.5×10^6 H1299 cells. After 30 days, Tumor tissue in each nude mouse Tumor tissue was taken out and detected with Agilent Human 4x44K Gene Expression Microarrays.
