Project description:We report a global survey of viral small RNAs (vsmRNAs) from >200 Aedes aegypti samples to identify many mosquito viruses that actively infect this prominent arboviral vector. Ae. aegypti viruses in the Americas were abundant, with some displaying geographical boundaries. Viruses infecting Asian Ae. aegypti were similar to those in the Americas and revealed the first wild example of dengue vsmRNAs. African Ae. aegypti displayed vsmRNAs from viruses unique to these African strains. Academic lab colonies generally lacked viruses, yet two commercial strains were deeply infected by a tombus-like virus that is related to plant viruses. Comparing matched viral long RNAs to vsmRNAs revealed viral transcripts evading the mosquito RNA interference (RNAi) pathway. By infecting mosquito cells with Ae. aegypti homogenates, we generated stably infected cell lines which produced vsmRNAs that were comparable to native mosquito vsmRNA patterns. Lastly, we demonstrate that these stably infected mosquito cells producing vsmRNAs can exert gene silencing of reporters bearing viral sequence segments, providing a potential explanation for how Ae. aegypti can tolerate the persistence of viral infections. This vsmRNA genomics approach in Ae. aegypti can add to existing vector surveillance approaches by discovering new viruses that persist in mosquito populations.

Project description:Under physiological conditions, extracellular vesicles (EVs) are present simultaneously in the extracellular compartment together with cytokines. Thus, we hypothesized that EVs in combination with cytokines induce different responses of monocyte cells compared to EVs or cytokines alone. Human monocyte U937 cells were incubated with EV-containing or EV-free CCRF human T-cell supernatant, with or without the addition of TNF. U937 cells cultured in EV-free supernatant, supernatant containing CCRF t-cell derived EVs, TNF or both. Each treatment option was measured in 3 replicates.

Project description:With the goal to identify miRNAs and other small RNAs involved in EV-mediated oligodendrocyte-neuron communication, we performed deep sequencing (RNA-seq) of small RNAs derived from EVs isolated from culture supernatants of primary mouse oligodendrocytes by differential centrifugation. Oligodendrocytes were cultured and EVs harvested under serum-free conditions in presence of B27 supplement. We found that RNA isolated from EVs harvested from cells under serum-replacement conditions contains miRNA contaminants carried into the sample by defined media components.

Project description:Aedes aegypti is a major vector for dengue, chikungunya and yellow fever. Though salivary gland plays a crucial role in transmission of virus and vector life cycle, there is no global and unbiased published report on salivary gland proteome of Ae. aegypti. In this study, we have carried out mass spectrometry based proteomic analysis of salivary gland from adult female Ae. aegypti mosquitoes. By fractionating the proteins isolated from salivary gland on SDS-PAGE and analyzing the in-gel digested bands on high resolution mass spectrometry, we identified 1,205 proteins. This is by far the largest catalogue of salivary gland proteome of Ae. aegypti. These proteins were then assigned molecular functions and biological processes. Several immunity related pathways were found to be enriched in salivary gland. In addition, subset of proteins was predicted to secretory in nature and may play an important role during blood feeding. This study provides a useful resource of proteins expressed in salivary gland of Ae. aegypti female mosquitoes and will aid in biomedical research focused on development of transmission blocking vaccine.

Project description:The mosquito Aedes aegypti serves as the primary vector for dengue virus (DENV), a human pathogen responsible for infecting hundreds of millions of people globally each year. Natural populations of Ae. aegypti show significant variability in their ability to become infected with DENV, which is largely governed by mosquito genetic factors. Previous studies suggest that genes encoding digestive enzymes, such as trypsins, and canonical antiviral immune genes may contribute to this variation 5-10. However, the specific causal gene variants underlying DENV susceptibility in Ae. aegypti are unknown. Here, we identify a gene encoding a cytochrome P450 (CYP4G15) whose genetic variants drive differences in DENV susceptibility in a natural Ae. aegypti population. CYP4G15 is transiently overexpressed in the midguts of DENV-resistant mosquitoes early in infection. Knockdown of CYP4G15 expression increases DENV susceptibility, while transgenic overexpression enhances resistance. A naturally occurring CYP4G15 variant with an 18-base-pair deletion in its promoter shows reduced expression and confers higher DENV susceptibility. The identification of CYP4G15 as a factor underlying DENV susceptibility in Ae. aegypti is unexpected because cytochrome P450 enzymes are typically associated with metabolism. Our finding challenges the long-standing focus on canonical immune pathways and opens new avenues for understanding antiviral defense and DENV transmission in mosquitoes.

Project description:Aedes mosquitoes transmit pathogenic arthropod-borne (arbo) viruses, putting nearly half the world’s population at risk. Blocking virus replication in mosquitoes rather than in humans serves as a promising approach to prevent arbovirus transmission, which requires in-depth knowledge of mosquito immunity. By integrating multi-omics data, we identified that heat shock factor 1 (Hsf1) regulates eight small heat shock protein (sHsp) genes within one topological associated domain. This Hsf1-sHsp cascade acts as an early response against chikungunya virus (CHIKV) infection and shows pan-antiviral activity in three vector mosquitoes, Aedes aegypti, Aedes albopictus, and Anopheles gambiae. We then assessed the baseline expression of sHsp genes in different tissues of female Ae. aegypti using RNA-seq, and we observed a highly dynamic expression pattern of sHsp genes that varied dramatically across different tissues. Interestingly, sHsp genes were expressed at low levels in two main barrier tissues, the midgut and salivary glands, compared to other tissues such as the crop. Importantly, activation of Hsf1 led to a reduced CHIKV infection rate in adult Ae. aegypti mosquitoes, demonstrating Hsf1 as a promising target for the development of novel intervention strategies to limit arbovirus transmission by mosquitoes.

Project description:Chromosomal inversions play a fundamental role in evolution and have been shown to be responsible for the epidemiologically important traits in malaria mosquitoes. However, they have never been characterized in the major vector of arboviruses Aedes aegypti because of the poor structure of its polytene chromosomes. In this study, we applied a Hi-C proximity ligation approach to identify chromosomal inversions in 23 recently collected strains of Ae. aegypti from its worldwide distribution, two old laboratory colonies, and Ae. mascarensis.
