Spatial proteomics based on laser capture micro-dissection (LCM) for investigating the impact of CD168+ TAM- coated tumor region (CCTR) on CD8+ T cells Function in colon cancer tissues

ABSTRACT: Multiplex immunofluorescence staining for CD11b, CD168, CD8, and Pan-CK was performed on three colon cancer specimens to identify CCTR regions. Subsequently, CD8+ T cells located at varying distances from the CCTR were subjected to single-cell resolution laser capture micro-dissection (LCM) and spatial proteomics analysis. These cells were categorized into three groups: the CCTR group (≤50 μm, n=393), the ad-CCTR group (>50 μm and ≤200 μm, n=375), and the non-CCTR group (>200 μm, n=383).

To measure extension longevity effect of soraprazan and identify the mechanism

Project description:Lipofuscin is known as intracellular waste substance produced by damaged mitochondrial incompleted decomposition. And it is a biomarker of aging. As aging, accumulation of the lipofuscin in retinal pigment epithelium induces blindness called Age-related macular degeneration. Soraprazan is developed to reduce the lipofuscin production and cure the age-related macular degeneration. Soraprazan reducing the age biomarker lipofuscin can be suggested to have effect of longevity extension. C.elegans were treated on NGM agar plate containing 0, 50, 100, 200 μM soraprazan and showed the lipofuscin reduction and extension of longevity. To identify the mechanism, 0 and 200 μM treated worms’ RNA expression was analyzed through microarray.

Project description:For RNA-seq, new cuttings from 3-week-old in vivo plants were cultured on the solid Woody Plant Medium (WPM) containing 20 g/L sucrose and 3 g/L Gelrite (Duchefa). After 5 days of culture, the root primordium formed cuttings were transferred to modified liquid WPM containing 10 μM or 200 μM KH2PO4, and harvested the basal ends of stem cuttings containing root primordium with its surrounding tissues using 11# scalpel after 2h, 4h, 8h, 12h and 24h culture. Samples were collected from two biological replicates (50 cuttings each).

Transcriptome analysis of endometrium cells in Equus asinus

Project description:Deoxynivalenol (DON) and zearalenone (ZEA) are the most common mycotoxins, which are damage to the reproductive system of human and animal. Endometrium cells perform vital roles of female reproduction. In the present study, transcriptomic was conducted on different dosage (0 μM, 10 μM and 30 μM) of DON and ZEA treatment donkey endometrium cells. Transcriptomic analysis showed 7386 differentially expressed genes (DEGs) were identified between DON 10 μM treatment group and control group, with 3655 up regulated and 3731 down; 8694 DEGs were identified between DON 30 μM treatment group and control group, with 4532 up regulated and 4162 down; 1768 DEGs were identified between ZEA 10 μM treatment group and control group, with 763 up regulated and 1005 down; and 3311 DEGs were identified between ZEA 30 μM treatment group and control group, with 1501 up regulated and 1810 down. In DEGs, Caspase-1, GSDMD-NT, ESR 1, PRDX 4, NOX 1, MSH 6 and CD 44, were expressed changed in DON and ZEA treatment group, which related to pyroptosis, hormone secretion, oxidative stress and cancerous. It is unprecedented that DON induced cell death by pyroptosis. This study provides valuable genetic resources for understanding the mechanisms and pathways of the toxin effect of DON and ZEA for Equus asinus’ endometrium cells.

Transcriptome analysis of Sertoli cells in Equus asinus

Project description:Deoxynivalenol (DON) and zearalenone (ZEA) are the most common mycotoxins, which are damaging to the reproductive system of humans and animals. Sertoli cells perform vital roles of male reproduction include composition of the blood testis barrier, support and release sperm. In the present study, transcriptomic analysis was conducted on different dosages (0 μM, 10 μM and 30 μM) of DON and ZEA treated donkey Sertoli cells. Transcriptomic analysis showed 6551 differentially expressed genes (DEGs) were identified between DON 10 μM treatment group and control group, with 3300 up regulated and 3251 down; 8605 DEGs were identified between DON 30 μM treatment group and control group, with 4841 up regulated and 3764 down; 803 DEGs were identified between ZEA 10 μM treatment group and control group, with 391 up regulated and 412 down; and 5947 DEGs were identified between ZEA 30 μM treatment group and control group, with 2816 up regulated and 3131 down. In DEGs, Caspase-1, GSDMD-NT, ESR 1, AR, PRDX 4, NOX 1, MSH 6 and CDK 1, were expressed highly in DON and ZEA treatment group, which related to pyroptosis, hormone secretion, oxidative stress and cancerous. It is unprecedented that DON induced cell death by pyroptosis. This study provides valuable genetic resources for understanding the mechanisms and pathways of the toxin effect of DON and ZEA for Equus asinus' Sertoli cells.

Uptake of polystyrene nanoplastics by cumulus-oocyte complexes impairs oocyte and embryo development

Project description:Nanoplastics (NPs), plastic particles smaller than 1 μm, have gained increasing attention due to their potential health effects. Experimental studies in animals have shown that NPs can pass biological barriers, reach the female reproductive tract, and enter the ovary. Their presence in the ovary might have profound consequences for reproductive endpoints, such as oocyte development. This study aimed to investigate the uptake and associated effects of polystyrene (PS) NPs on oocyte and embryo development using a bovine in vitro fertilization (IVF) model. Bovine cumulus-oocyte complexes (COCs) were exposed to pristine PS-NPs (50 nm or 200 nm) at different doses during in vitro maturation (23 hours), followed by IVF and embryo culture. RNA sequencing was performed to investigate mechanisms involved in possible effects on oocyte maturation. Confocal microscopy confirmed the internalization of 50 nm and 200 nm PS-NPs into cumulus cells, however, only 50 nm particles were able to transfer into oocytes. Additionally, exposure to 50 nm PS-NPs at 3 μg/mL significantly delayed oocyte nuclear maturation and early embryo development. At this dose, RNA sequencing did not identify differentially expressed genes in the COCs, however, gene set enrichment analyses did indicate effects on mitochondrial processes. Nevertheless, subsequent analysis on mitochondrial activity remained unaffected.
