Excitatory neurons SIRT6 mediates chronic stress‑induced depression in mice

ABSTRACT: We used acetyl-lysine antibody-based affinity enrichment followed by mass spectrometry to identify acetylation changes caused by SIRT6 overexpression. We co-overexpressed Flag-OVOL2 and pCMV-SIRT6 in N2a cells. At 96 h after transfection, we treated cells with TSA for 12 h, collected proteins and digested proteins into peptides, enriched acetylated peptides using an anti-acetyl-lysine antibody, and then performed LC-MS/MS.

Quantitative acetylome analysis of Fusarium oxysporum

The collected cellular proteins were extracted, digested, and labeled by different TMTs. Project description:Lysine acetylation is a dynamic, reversible post-translational modification that is known to play an important role in regulating the activity of many key enzymes in bacteria. Acetylproteome studies have been performed on some bacteria. However, until now there have been no data on the Actinomycetes, which are the major producers of therapeutic antibiotics. In this study, we investigated the first acetylproteome of the erythromycin-producing Actinomycete Saccharopolyspora erythraea using a high-resolution mass spectrometry-based proteomics approach. Using immune-affinity isolation of acetyl-peptides with an anti-acetyllysine antibody followed by nano-UPLC/MS/MS analysis, we identified 664 unique lysine acetylated sites on 363 proteins.

Lysine acetolome of breast cancer-derived small extracellular vesicles reveals specific acetylation pattern for metabolic enzymes

Project description:Cancer-derived small extracellular vesicles have been proposed as promising potential biomarkers for diagnosis and prognosis of breast cancer (BC). We performed a proteomic study of lysine acetylation of breast cancer-derived small extracellular vesicles (sEVs) to understand the potential role of the aberrant acetylated proteins in the metastasis and progression of BC. Three cell lines were used as models for this study: MCF10A (non-metastatic), MCF7 (estrogen and progesterone receptor positive, metastatic) and MDA-MB-231 (triple-negative, highly metastatic). For a com-prehensive protein acetylation analysis of the sEVs derived from each cell line, acetylated peptides were enriched using the anti-acetyl-lysine antibody, followed by LC-MS/MS analysis. In total, 118 lysine acetylated peptides of which 22, 58 and 82 have been identified in MCF10A, MCF7 and MDA-MB-231 cell lines, respectively. These acetylated peptides were mapped to 60 distinct pro-teins and mainly identified proteins involved in metabolic pathways. Among those identified acetylated proteins presented in cancer-derived sEVs (MCF7, MDA-MB-231) are proteins associ-ated with glycolysis pathway, annexins and histones. Five enzymes from the glycolytic pathway, present only in sEVs isolated from cancer-derived sEVs cell lines, were validated. These include aldolase (ALDOA), glyceraldehyde-3-phosphate dehydrogenase (GAPDH), phosphoglycerate kinase (PGK 1), enolase (ENO) and pyruvate kinase M1/2 (PKM). For three of these enzymes, ALDOA, PGK 1 and ENO, the specific enzymatic activity was significantly higher in MDA-MB-231 when compared with MCF10A-derived sEVs. This study reveals that sEVs contain acetylated glycolytic metabolic enzymes that could be interesting potential candidates for early BC diagnostics.

Global acetylomics of Campylobacter jejuni shows lysine acetylation regulates CadF adhesin processing and fibronectin binding during in vivo-like growth

Project description:Campylobacter jejuni is the leading cause of acute bacterial gastroenteritis in the developed world. Despite the prevalence of infection, the pathogenesis of C. jejuni remains poorly understood. Lysine acetylation (KAc) is a reversible post-translational modification (PTM) that can alter the protein structure / function paradigm, however specific roles for KAc are largely undefined in bacteria. Acetyl-lysine immunoprecipitation and liquid chromatography coupled to tandem mass spectrometry (LC-MS/MS) identified 5567 acetylated lysines on 1026 C. jejuni proteins (~63% of the predicted C. jejuni proteome). Functional enrichment confirmed acetylated proteins are involved in metabolism, transcription and translation, stress responses and chemotaxis. KAc was identified on proteins from all subcellular locations, including outer membrane (OM) and extracellular proteins. Label-based LC-MS/MS identified acetylated proteins and modified sites associated with growth in 0.1% sodium deoxycholate (DOC, a component of gut bile salts). A total of 3571 acetylated peptides were quantified and 761 (from 409 proteins) were differentially abundant following growth in DOC. Changes in KAc involved multiple pathways suggesting a dynamic role for this PTM in bile resistance. As observed in other bacteria, we show that KAc is primarily non-enzymatically mediated via acetyl-phosphate; however, the sole deacetylase CobB also contributes to a global elevation of this modification in DOC. We observed several multiply acetylated OM proteins and altered DOC abundance of acetylated peptides in the major fibronectin (Fn)-binding adhesin CadF. We show that KAc influences CadF Fn binding and prevalence of processed lower mass variants. This study provides the first system-wide analysis of the C. jejuni lysine acetylome and contributes to our understanding of KAc as an emerging PTM in bacteria.

Role of MYC acetylated lysine residues in cell transformation and gene-selective regulation [Rat1A]

Project description:The functions of MYC acetylation have remained unclear. In this study we identified the major lysine residues of MYC that are acetylated by p300 and GCN5 in cancer cell lines and established the requirment of three specific acetyl-lysine (AcK) residues for MYC transformation of Rat1a fibroblasts and MCF10 mammary epithelial cells. We further uncovered gene-selective regulatory functions and conserved genetic programs and biolofical processes regulated by MYC AcK residues in both cell types.
