Serum Metabolomics in the Rat Model of Recurrent Pelvic Pain

Project description:<p>This study aimed to investigate systemic metabolic alterations underlying pain-depression comorbidity in the Recurrent Pelvic Pain (RPP) rat model using untargeted serum metabolomics (UPLC-Q-Orbitrap-MS). Comparative analysis revealed distinct metabolic profiles, identifying 234 differential metabolites enriched in steroids, fatty acyls, and carboxylic acids. KEGG analysis highlighted reprogramming of amino acid metabolism (e.g., phenylalanine, tyrosine, and tryptophan biosynthesis) and arachidonic acid metabolism. Integrative target prediction identified 10 key overlapping targets (e.g., TRPV1, OPRM1, PTGS2) associated with inflammatory response, fatty acid metabolism, and synaptic signaling. These targets were further linked to chronic pain and hyperalgesia. The metabolomic profile supports progressive nociceptive sensitization and reveals altered neurotransmitter biosynthesis pathways, providing insight into mechanisms of pain-depression comorbidity and potential therapeutic targets.</p>

Recurrent triple-negative breast cancer from cysteine deprivation loses tumorigenicity via downregulation of the CST4 signaling

Project description:Triple-negative breast cancer (TNBC) is an aggressive type of breast cancer with a high risk of recurrence following treatment. Inhibition of cysteine uptake by erastin effectively induces mesenchymal TNBC undergoing prompt ferroptosis. However, a small residual population of cancer cells keeps resistant and survived during the erastin treatments, which might mimic tumor relapse in patients after therapy. We enriched and established the erastin-resistant/recurrent TNBC cell models by multi-cycle challenges of erastin. Identified epigenetic compounds could abolish such resistance in recurrent cells. Intriguingly, recurrent TNBC failed to grow in anchorage-independent conditions, implying a loss of tumorigenicity. By analyzing transcriptomic profiling, recurrent TNBC displayed highly transcriptomic alterations and attenuated signaling processes, including K-Ras signaling. Knockout of CST4, a member of the cystatin gene family strongly suppressed in recurrent cells, significantly reduced the tumorigenic potential and K-Ras signaling. Our findings suggested that targeted cysteine therapy could be a valid treatment for TNBC with a low possibility of tumorigenicity formation.

A methylome-based score to predict response to regorafenib in recurrent glioblastoma

Project description:Following encouraging results of the REGOMA Phase 2 trial, regorafenib is increasingly used for the treatment of recurrent glioblastoma, IDH-wildtype. Identifying predictive factors for response to regorafenib is of paramount importance. DNA methylation profiling is considered the gold standard for diagnosing and classifying central nervous system tumors. We explored the predictive value of methylation profiling in recurrent glioblastoma patients treated with regorafenib, highlighting the potential of DNA methylation profiling in guiding therapeutic strategies and warrant further validation in clinical settings.

A dataset describing a suite of novel antibody reagents for the RAS signaling network

Project description:RAS genes are frequently mutated in cancer and have for decades eluded effective therapeutic attack. The National Cancer Institute’s RAS Initiative has a focus on understanding pathways and discovering therapies for RAS-driven cancers. Part of these efforts is the generation of novel reagents to enable the quantification of RAS network proteins. Here we present a dataset describing the development, validation (following consensus principles developed by the broader research community), and distribution of 104 monoclonal antibodies (mAbs) enabling detection of 27 phosphopeptides and 69 unmodified peptides from 20 proteins in the RAS network. The dataset characterizes the utility of the antibodies in a variety of applications, including Western blotting, immunoprecipitation, protein array, immunohistochemistry, and targeted mass spectrometry. All antibodies and characterization data are publicly available through the CPTAC Antibody Portal, Panorama Public Repository, and/or PRIDE databases. These reagents will aid researchers in discerning pathways and measuring expression changes in the RAS signaling network.
