Project description:Ubiquitination is the key mechanism that mediates protein stability and degradation, and the interference of ubiquitination is a promising approach to improve the therapeutic effects in cancer therapy. Here, we demonstrate that targeting inhibition of deubiquitinase OTUB1 could increase the ubiquitylation of PD-L1 and GPX4, which makes tumor cells vulnerable to the ferroptosis and immunotherapy. The therapeutic vehicles are designed through the encapsulation of metal-polyphenol network nanocomplexes (GA/Fe), doxorubicin (DOX), and siRNA targeting OTUB1 (siOTUB1) within a modified tumor cell membrane (SFD@M). Targeting delivery of SFD@M driven by tumor membrane efficiently decreases the expression of PD-L1 and GPX4 in tumor tissues. Without the protection of GPX4, GA/Fe complexes in SFD@M induce strong ferroptosis in tumor cells and initiate anti-tumor immune responses. Low expression of PD-L1 in tumor tissues due to excess ubiquitination maintain the activation of CD8+ T cells. In addition, the overexpression of PD-1 on edited tumor cell membrane regulates the function of Tregs and MDSCs, and further enhanced the anti-tumor effect of CD8+ T cells. SFD@M demonstrates efficient inhibition of tumor growth in orthotopic tumor models and cell line-derived xenograft (CDX) models in humanized NOG mice. This study employs the ubiquitination process in tumor cells to develop a biomimetic strategy that enhances ferroptosis and augments the immunotherapeutic efficacy in cancer treatment.

Project description:Glutathione peroxidase 4 (GPX4) utilizes glutathione (GSH) to detoxify lipid peroxidation and
plays an essential role in inhibiting ferroptosis. As a selenoprotein, GPX4 protein synthesis is
highly inefficient and energetically costly. How cells coordinate GPX4 synthesis with nutrient
availability remains unclear. In this study, integrated proteomic and functional analyses reveal
that SLC7A11-mediated cystine uptake promotes not only GSH synthesis but also GPX4 protein synthesis. Mechanistically, cyst(e)ine activates mechanistic/mammalian target of rapamycin complex 1 (mTORC1) and promotes GPX4 protein synthesis at least partly through the mTORC1-4E-BP signaling axis. Pharmacologic inhibition of mTORC1 decreases GPX4 protein levels, sensitizes cancer cells to ferroptosis, and synergizes with ferroptosis inducers (FINs) to suppress patient-derived xenograft tumor growth in vivo. Together, our results reveal a hitherto unrecognized regulatory mechanism to coordinate GPX4 protein synthesis with cyst(e)ine availability and suggest to use the combination of mTORC1 inhibitors and FINs in cancer treatment.

Project description:Cystine-xCT transporter-Glutathione (GSH)-GPX4 axis is the canonical pathway to protect against ferroptosis. While not required for ferroptosis-inducing compounds (FINs) targeting GPX4, FINs targeting the xCT transporter require mitochondria and its lipid peroxidation to trigger ferroptosis. However, the mechanism leading to the distinct difference between these FINs is still unknown. Given that cysteine is also required for coenzyme A (CoA) biosynthesis, here, we showed that CoA supplementation specifically prevents ferroptosis induced by xCT inhibitors but not GPX4 inhibitors.

Project description:GPX4-dependent ferroptosis has emerged as a therapeutic strategy for cancer treatment. Here, we demonstrated that protein kinase A (PKA) participates in the regulation of ferroptosis by controlling the m6A modification of GPX4 in an ALKBH5-dependent manner. Notably, we identified ALKBH5, an m6A demethylase, as a novel target of PKA, which drives phosphorylation-dependent degradation of ALKBH5 protein. Moreover, the deletion of ALKBH5 represses ferroptotic cell death by maintaining GPX4 m6A modification and stability. Thus, by regulating ALKBH5-dependent GPX4 stability, PKA acts as a key regulator of ferroptosis. Our study unveils the involvement of PKA in m6A modification, which could control GPX4-dependent ferroptosis and tumor progression.
