Tumor-Derived Extracellular Vesicle Proteomics Enables Precise Ovarian Cancer Liquid Biopsy


ABSTRACT: Ovarian cancer (OC) remains one of the most lethal gynecological malignancies, and effective non-invasive biomarkers for early diagnosis, postoperative monitoring, and prognosis assessment are still limited. Serum CA125, the current standard biomarker for OC, has limited diagnostic accuracy, particularly in early-stage disease. Cancer-derived extracellular vesicles (EVs) carry tumor-specific proteomic information and represent promising biomarkers; however, their sensitive and specific isolation from complex biological fluids remains challenging. In this project, we developed BP-EVlent, a novel approach that combines polyethylene glycol (PEG)-based precipitation with CA125 immunoaffinity capture to precisely isolate plasma CA125-positive EVs from patients with OC.
