Characterization of the nasal microbiome and metabolome in children with Henoch-Schonlein purpura and children with Henoch-Schonlein purpura nephritis

ABSTRACT:

Three groups of eligible subjects were included, including 20 cases in the HSP group, 20 cases in the HSPN group, and 20 cases in the healthy control (HC) group. Nasal lavage samples from each group were subjected to 16SrRNA gene sequencing and metabolomic analyses to screen differential flora and differential metabolites, and Spearman correlation analysis was done.

Exploring the mechanism of Jia Wei Xing Bi Gel nasal drops on Children with Allergic Rhinitis based on Transcriptomics and Network Pharmacology

Project description:Objective: This study combines transcriptomics and network pharmacology to explore the mechanism of action of Jia Wei Xing Bi Gel nasal drops in treating allergic rhinitis (AR) in children. Methods: A total of 56 AR children were randomly divided into Jia Wei Xing Bi Gel nasal drops(JWXBGND) group and mometasone furoate nasal spray (MFGS) group, with 28 cases in each group, and a healthy control group of 20 cases. After 4 weeks of treatment, efficacy was evaluated, and RNA-seq, database mining, and bioinformatics analysis methods were employed. Results: In terms of clinical efficacy, the TCM group showed better improvement in nasal itching, ocular symptoms, non-nasal ocular symptoms, and behavioral issues compared to the western medicine group, with a more significant decrease in TCM syndrome scores; transcriptomic analysis revealed 2776 differentially expressed genes when comparing AR children with healthy children, and 441 differential genes were identified between JWXBGND group and MFGS group; combined analysis of transcriptomics and network pharmacology identified 195 active components and 493 targets of action for Jiawei Xingbi Gel, with core components including ferulic acid, fexofenadine, and curcumin A, and core targets involving TNF, IL-6, IL-1β, AKT1, ALB, SRC, etc.; KEGG pathway analysis mainly enriched in caffeine metabolism, linoleic acid metabolism, Th17 cell differentiation, HIF-1 signaling pathway, etc.; molecular docking results indicated that key active components of Jia Wei Xing Bi Gel nasal drops bind stably to targets such as ALB, AKT1, SRC, TP53, etc. Conclusion: Jia Wei Xing Bi Gel nasal drops can effectively alleviate AR symptoms, and its mechanism may be related to the regulation of inflammatory signaling pathways through multiple components and multiple targets

Project description:To enhance granulocyte recovery in scRNA-seq, we employed nasal lavage samples from children with asthma, leveraging the 10x Genomics Flex platform combined with a customized data processing pipeline. Nasal lavage samples were processed without prior manipulation to avoid technical artifacts such as lysis or stimulation. Granulocyte recovery was optimized using fixation to preserve cell quality and advanced computational techniques to separate low RNA content cells from background noise.

Project description:To elucidate the potential role of commensal viruses in the etiology of AR, we performed a metagenomic analysis of nasal lavage fluid (NLF) to identify commensal viruses in the nose of mice colonizing in the specific pathogen-free (SPF) circumstances (Vehicle group). Glycoproteomics revealed novel N-glycosylation biomarkers for early diagnosis of lung adenocarcinoma cancers

Project description:Circulating biomarkers play important roles in diagnosis of malignant tumors. N-glycosylation is an important post-translation patter and obviously affect biological behaviors of malignant tumor cells. However, the role of N-glycosylation sites in early diagnosis of tumors still remains further investigation. In this study, plasma from 20 lung adenocarcinoma (LUAD), which were all classified as stage I, as well as 20 normal controls (NL) were labeled and screened by mass spectrometry (MS). Total 39 differential N-glycosylation sites were detected in LUAD, 17 were up-regulated and 22 were down-regulated. In all differential sites, ITGB3-680 showed highest potential in LUAD which showed 99.2% AUC, 95.0% SP and 95.0% SN. Besides, APOB-1523 (AUC: 89.0%, SP: 95.0%, SN: 70.0%), APOB-2982 (AUC: 86.8%, SP: 95.0%, SN: 45.0%) and LPAL2-101 (AUC: 81.1%, SP: 95.0%, SN: 47.4%) also acted as candidate biomarkers in LUAD. Combination analysis was then performed by random forest model, all samples were divided into training group (16 cases) and testing group (4 cases) and conducted by feature selection, machine learning, integrated model of classifier and model evaluation. And the results indicated that combination of differential sites could reach 100% AUC in both training and testing group. Taken together, our study revealed multiple N-glycosylation sites which could be applied as candidate biomarkers for early diagnosis diagnosis of LUAD.
