Gut Microbiota-Proline Axis Drives HFpEF Through Fibroblast Ferroptosis

ABSTRACT:

Heart failure with preserved ejection fraction (HFpEF) is a complex cardiometabolic disorder closely associated with systemic metabolic dysregulation. In this study, plasma samples were collected from C57BL/6J mice subjected to a high-fat diet–induced HFpEF model, with or without intervention, and analyzed using untargeted metabolomics to characterize disease-associated metabolic alterations. This dataset aims to provide insights into plasma metabolic signatures and mechanisms underlying HFpEF.

Project description:In this project we explore the cellular heterogeneity of a mouse model of heart failure with preserved ejection fraction (HFpEF) involving a two-hit model of feeding a high fat diet (HFD) along with L-NAME administration. Healthy adult male mice (C57BL/6J inbred) were fed either a normal chow diet or HFD/L-NAME for 10 weeks or 15 weeks before performing sequencing experiments. Both cardiomyocytes (CMs) and total interstitial population (TIP) were captured using a protocol to jointly capture and sequence single-nuclei (for cardiomyocytes) and single-cells (for TIP) using the 10x Genomics Chromium system.

Canagliflozin regulates ferroptosis to ameliorate heart failure in rats with preserved ejection fraction

Project description:Chronic heart failure (CHF), with whether reduced ejection fraction (HFrEF) or preserved ejection fraction (HFpEF), is the end-stage of various cardiovascular diseases and severely affects the patients’ lifespan. There has been no effective drugs to improve clinical outcomes for a long time. Sine the recent years, as a hypoglycemic drug, sodium-glucose cotransporter 2 inhibitor (SGLT2i) has aroused great attention due to it’s cardiovascular benefits. However, the underlying mechanisms remain unclear, particularly regarding ferroptosis, a newly defined mechanism of iron-dependent non-apoptotic cell death in heart failure (HF). Here, we discovered and demonstrated that ferroptosis was a key mechanism in rat model of high-salt diet-induced HF, characterized by iron overloading and lipid peroxidation, which was blocked by canagliflozin administration. These findings highlight that targeting ferroptosis serves as a cardioprotective strategy for HF prevention and canagliflozin might display cardiovascular benefits through ferroptosis mitigation.

Transcriptomic Profiling of L-Proline Induced Ferroptosis Reveals PPARγ-Mediated Regulatory Networks in HFpEF Progression

Project description:This study investigates the molecular mechanisms by which L-proline contributes to diabetic heart failure with preserved ejection fraction (HFpEF) through the induction of ferroptotic stress under metabolic conditions. We found that L-proline exacerbates ferroptosis-related oxidative and lipid peroxidation stress in cardiac tissue within a metabolically driven HFpEF context. Mechanistically, L-proline markedly suppressed PPARγ signaling, a key regulator of lipid metabolism, redox homeostasis, and ferroptosis resistance. Downregulation of PPARγ was accompanied by upregulation of pro-ferroptotic mediators, including ACSL4 and TFR1, indicating enhanced iron-dependent lipid peroxidation and ferroptotic susceptibility. These findings support a mechanistic link between amino acid metabolic remodeling and fibroblast-centered cardiac vulnerability in diabetic HFpEF, highlighting the role of the proline–PPARγ–ferroptosis axis in promoting myocardial fibrosis and diastolic dysfunction under chronic metabolic stress.
