Metabolomic profiling of goat testicular tissue under graded uranium exposure

ABSTRACT:

This study presents metabolomic profiles of goat testicular tissue obtained under controlled exposure conditions. An untargeted metabolomics approach was applied to characterize global metabolic alterations associated with environmental contaminants. The dataset provides information on small-molecule metabolites and supports investigations into metabolic responses in reproductive tissues under stress conditions.

Gut microbiota-mediated histidine deficiency drives testicular ferroptosis induced by bisphenol F exposure

Project description:<p>Bisphenol F (BPF), a widespread environmental contaminant and a major substitute for the restricted bisphenol A (BPA), has raised increasing concerns regarding its potential male reproductive health risks, yet its underlying mechanisms remain poorly understood. This study investigates the mechanisms underlying BPF-induced testicular damage, focusing on the interplay among gut microbiota (GM) dysbiosis, histidine metabolism disruption, and ferroptosis. Using a mouse model exposed to BPF (50, 100, and 200 mg/kg/day) for 28 days, we observed significant testicular pathology, including seminiferous tubule atrophy, vacuolation, and blood-testis barrier (BTB) impairment. Metagenomic and metabolomic analyses revealed GM dysbiosis and suppressed intestinal histidine metabolism, accompanied by decreased abundance of beneficial taxa (e.g., Bacteroides, Ligilactobacillus) and increased potential pathobionts (e.g., Akkermansia, Mucispirillum). BPF exposure also inhibited the expression of the histidine transporter LAT1, reducing histidine uptake in testes. Crucially, BPF triggered testicular ferroptosis, characterized by mitochondrial damage, iron accumulation, lipid peroxidation, and downregulation of the ferroptosis regulatory system xCT-GSH-GPX4 axis. In vitro experiments using mouse Sertoli cells (mSCs) confirmed BPF-induced ferroptosis, which was mitigated by the exogenous histidine supplementation. Histidine administration in vivo ameliorated testicular damage, restored BTB integrity, and reversed ferroptotic markers. Our findings unveil a novel “GM-histidine-testis” axis as a key mechanistic pathway for BPF-induced reproductive toxicity, providing critical insights into the health risks of emerging environmental pollutants and suggesting potential dietary intervention strategies.</p>

Integrated study of the circRNA, lncRNA and mRNA networks in testicular heat exposure

Project description:The World Health Organization has recognized testicular function with temperature dependence. Testicular heat exposure caused by occupational factors, lifestyle and clinical disease, etc., can lead to different degrees of reproductive obstacles. The aim of this study is to reveal the transcriptional regulatory network and their potential crucial roles in testicular heat exposure. After passing quality control, the high throughput sequencing data of mouse testicular tissue for scrotum heat exposure and control group were carried out various analyses including differentially expressed transcriptome exploration, Gene Ontology (GO), Kyoto Encyclopedia of Genes and Genomes (KEGG) and network regulation. Differential transcriptome expression analysis revealed that 172 circRNA (91 upregulation and 81 downregulation), 465 lncRNA (96 upregulation and 369 downregulation) and 2721 mRNA (2003 upregulation and 718 downregulation) were identified as significant differential expression in the mouse testicular tissue of heat exposure compared with control group using the cut-off of log2fold change >= 0.585, p value <= 0.05 and q value <= 1.00. Through GO term analysis, differentially expressed lncRNA and mRNA may play important functions in negative regulation of cellular process [GO:0048523], meiotic cell cycle [GO:0051321], cytoplasm [GO:0005737], membrane raft [GO:0045121], amide binding [GO:0033218], extracellular matrix binding [GO: 0050840]. KEGG pathway analysis of differentially expressed lncRNA and mRNA can be classified into MAPK signaling [mmu04010], purine metabolism [mmu00230], homologous recombination [mmu03440], RNA transport [mmu03013], nod-like receptor signaling pathway [mmu04621]. This research provides high throughput sequencing data of transcriptome in testicular heat exposure model and lays the foundation for further study on the circRNA and lncRNA in male reproductive diseases related to elevated testicular temperature.

Altered gene expression profiles in testis developmental exposed to endocrine disruptors

Project description:Environmental reproductive health focuses on the effect of exposure to contaminants considered as endocrine disruptors. Developmental testis is considered as target of these compounds affecting testicular functions in adults and suspected implications in tumor etiology. Comparative analysis of gene expression in mouse testis exposed to five disruptors, three different dosages and three accumulative developmental stages shown defined signature profiles of gene deregulation for MEHP (monoethyl phthalate) and zearalenone (a phytoestrogen) and different to 17β-estradiol exposure. The effects are even detected in postpuberal male offspring from premating exposed mothers. Oxidative stress response, protein ubiquitination and oxidative phosphorylation are the most representative pathways affected.

This atlas was built upon 241K nuclei across the worm lifespan at day 1, 6, 12, and 14 (100%, 99%, 61%, and 14% survival rates, respectively). The worms were aged under normal physiological conditions, without interfering with their reproductive processes.

Vitamin C mitigates potassium perchlorate exposure-induced disruption of spermatogenesis in medaka

Project description:Potassium perchlorate (KClO4), widely used in industrial and military applications, is an emerging environmental contaminant known to disrupt thyroid function. However, its potential impact on male reproductive health remains underexplored. In this study, we investigated the testicular toxicity induced by chronic KClO4 exposure over one spermatogenic cycle of medaka (Oryzias latipes) and evaluated the ameliorative effects of vitamin C. Adult male medaka were treated with 0.01 mg/L, 10 mg/L KClO4 and 10 mg/L KClO4 plus vitamin C (3 mg/mL) for 21 days. Fertilization percentage, histological examination, and transcriptomic profiling of the testis were performed. KClO4 exposure decreased fertilization success caused disorganization of seminiferous tubules, and dysregulated spermatogenic genes in the testis. Transcriptomic analysis revealed substantial dysregulation of genes involved in cadherin and tubulin binding, chromatin remodeling, oxidative stress response, and germ cell development. Co-administration of vitamin C mitigated these effects by restoring testicular morphology, restoring fertilization rates, and partially reversing gene expression changes disrupted by potassium perchlorate exposure. The present findings suggest that vitamin C provides protective effects against perchlorate-induced testicular toxicity and highlight the need for further exploration of antioxidant-based interventions to safeguard reproductive health from perchlorate exposure.

Effect of TCDD exposure in adult female and male mice on the expression of miRNA in the ovaries and testes and associated reproductive functions

Project description:The current study demonstrates for the first time that exposure of adult male and female mice to TCDD results in significant alterations in a large number of miRNAs in the ovaries and the testes. These findings suggest that environmental contaminants such as TCDD may alter reproductive functions through alterations in the expression of miRNAs. Future studies will focus on whether such miRNA changes also occur in the germ cells which would explain how TCDD exerts transgenerational effects on the reproductive system by regulating epigenetic pathways involving miRNA

Embryonic exposure to aluminium chloride blocks the onset of spermatogenesis through disturbing the dynamics of testicular tight junctions via upregulating Slc25a5 in offspring

Project description:Studies reveal neurotoxicity, hepatotoxicity, and developmental and reproductive toxicity in mice exposed to aluminium (Al). However, relatively few researches are conducted to clarify the mechanism of the impact of embryonic exposure to aluminium chloride (AlCl3) on the development of the reproductive system in male offspring. Here, pregnant mice were fed AlCl3 by gavage from day 12.5 of gestation. The results showed that embryonic AlCl3 exposure disrupted testicular development and spermatogenesis by damaging the testicular structure, reducing sperm counts, and altering the expression of the tight junction (TJ) proteins between Sertoli cells (SCs). Further in vitro studies showed that AlCl3 treatment stabilized the TJ proteins Occludin and ZO-1 expression by inhibiting the ERK signaling pathway to upregulate the Slc25a5 protein, which prompted ATP production to disrupt cytoskeletal protein homeostasis. Therefore, our study provided a new mechanism for AlCl3 exposure to interfere with testicular development and spermatogenesis and implied that Slc25a5 could be a target of AlCl3 affecting cell metabolism.

Evaluation of the testicular toxicity of prenatal exposure to bisphenol A and phthalate based on microarray analysis

Project description:In the present study, the effects of prenatal exposure to bisphenol A (BPA) and diethyl hexyl phthalate (DEHP) on male reproductive system of offspring were examined by a tissue-specific microarray and gene annotation with Medical Subject Headings (MeSH) indicative of testicular components. The aim of this study was to investigate the validity of microarray analysis with MeSH as a method for obtaining and evaluating an overview of testicular toxicity mechanisms in maternally exposed specimens. Pregnant mice were treated with BPA or DEHP by subcutaneous administration on days 7 and 14 of pregnancy. Tissue and blood samples were collected from 6-week-old male offspring. Testes were subjected to gene expression analysis using quantitative RT-PCR and cDNA microarray (testis2). To interpret the microarray data, genes were classified using MeSH related to the functions of testis and sperm. As a result, MeSH categories related to androgen of dysregulated genes and Sertoli cells of downregulated genes were only enriched in BPA-treated groups. These results were consistent with the observations in the sperm properties and histological examination of testes. This article concludes that 1) microarray analysis of expression changes of genes associated with testicular function and spermatogenetic process, and 2) a method using MeSH to extract common terms among dysregulated genes, are useful for predicting an overview of the testicular and reproductive toxicity of prenatal exposure to chemical substances.
