Untargeted LC–MS metabolomics analysis of sorghum root exudates under drought stress

ABSTRACT:

This study presents an untargeted metabolomics analysis of root exudates from sorghum (Sorghum bicolor) under control and severe drought conditions. Samples were collected from experimental sites in Shanxi, China. The study aims to investigate metabolic changes in sorghum root exudates in response to drought stress.

Project description:In our previous work, we found that the root exudates of sgn3 myb36 promoted the colonization of CHA0 on roots. Through LC-MS, we identified a large amount of glutamine (Gln) in the root exudates of sgn3 myb36. Therefore, we aim to use RNA-seq to uncover whether the root exudates of sgn3 myb36 and Gln have the same regulatory effects on CHA0. By conducting differential analysis with the CK (CHA0 treated with wild-type root exudates), we hope to identify the specific regulatory mechanisms of sgn3 myb36 and Gln on CHA0.

Rhizophagus irregularis gene regulation in response to rice root exudates

Project description:Purpose: The recent publication of the fungal mutualist R. irregularis genome facilitated transcriptomic studies. We here adress the gene regulation of R. irregularis in response to root exudates from rice wild-type and osnope1 (no perception candidate - mutant unable to host arbuscular mycorrhizal fungi) Methods: Spores of R. irregularis were treated with root exudates and collected at 1 hour, 24 hours and 7 days after addition. To monitor fungal gene regulation, control conditions were also prepared at T0, 1h, 24h and 7d. mRNA were sequenced by HiSeq Illumina. Reads were mapped on the Rhizophagus irregularis genome assembly (Gloin1 - Tisserant et al., PNAS, 2013) using CLCworkbench suite. Results: -At 1h, a set of 92 fungal genes were found up-regulated in response to wt root exudates (92), not to osnope1 root exudates, many of them being involved in cell signaling. -At 24h and 7d, numerous genes putatively involved in primary metabolism were up-regulated in response to wt root exudates, not in response to osnope1 root exudates -Several vital genes involved in cell development are repressed in response to osnope1 RE compared to wt RE. Conclusions: these results argue for a high metabolic activity induced by wt root exudates, not by osnope1 root exudates.

The spatial differences of root apex contribute to improved drought tolerance as induced by drought priming in wheat

Project description:Drought priming is a promising approach to improve drought tolerance, and root apex plays important role in coping with drought adversities in plants. It has been observed that less inhibition of root growth under drought stress was found in paralleling with the enhanced drought tolerance induced by drought priming in wheat. However, the contribution and mechanisms of root apex to the improved tolerance induced by drought priming remains unknown. Here, the transcriptome and proteome of three different zones along the root axis under drought stress were investigated. The two zones distal from the root apex showed more sensitive to drought priming and the later drought stress than the zone proximal to the root apex, as exemplified by the principal component analysis of the different expressed genes (DEGs) among treatments. The DEGs which decreased by drought stress, while higher expressed in primed plants than non-primed plants, might play critical roles in enhanced root growth in primed plants. These genes were mainly involved in the pathways related to cell membrane sensing, plant hormone signaling, stress defense and cell more modification. Moreover, the plant hormone pathway showed positive correlated between transcriptome and proteome analysis, and genes network analysis hinted that ABA receptor and aquaporin could be potential markers in regulation of priming induced root growth and drought stress tolerance.

Proteomic analysis of Pennisetum glaucum under drought stress

Project description:Pearl millet [Pennisetum glaucum (L.) R.Br] is the fifth most important cereal crop next to rice, wheat, maize, and sorghum. It is cultivated especially by small holder farmers in arid and semi-arid regions because of its drought resistance. However, the molecular mechanisms during drought stress in Pennisetum remain elusive. In the present study we have used a shotgun proteomics approach (GEL-LC-Orbitrap-MS) for identification and quantification of proteins from different tissues (root, seed and leaf) under drought and control conditions. Plants were grown in a tube system to survey root growth under drought stress. The water content was measured in the upper and the lower part of the tube using soil moisture sensors. Under drought stress root elongation was observed. Measurement of stomatal conductance showed a clear response to drought stress. For proteomics measurements root, leaf and seed tissues were harvested. In total 2281 proteins were identified, 1095 in root, 1299 in seed, and 1208 in leaf in both stress and control conditions.

We then performed differential gene expression analysis, identifying changes in transcriptome profiles between each treatment (Tomato, Maize) and the Control as reference conditions, and between the two treatments.

Small RNAs of sorghum in response to drought stress

Project description:We sequenced three small-RNA (sRNA) libraries constructed from leaves of sorghum subjected to three different treatments, well-watered (CK), mild drought (DR1) and severe drought (DR2). These findings will be useful for research on drought resistance and provide insights into the mechanisms of drought adaptation and resistance in sorghum.
