Project description:This study aimed to identify the protein composition of mucus in Crassostrea gigas (Pacific oyster) and analyze the proteomic changes during Vibrio ZJ-51 infection. Mucus samples were collected from control (C1–C3) and infected oysters (T1–T3). Using LC-MS/MS, over 4000 proteins were identified from oyster mucus. Differential expression analysis revealed significant proteomic alterations in response to Vibrio infection, providing insights into the host-pathogen interaction and potential defense mechanisms. This study contributes to the understanding of oyster mucus composition and its response to bacterial challenge.

Project description:Vibrio natriegens strain ATCC 14048 is a salt marsh isolate with a notably rapid growth rate that has garnered considerable interest for biotechnological applications. We used systems-level tools (i.e. metabolomics, transcriptomics, proteomics) to characterize its physiological response to different salinities and temperatures that are relevant not only to its natural environment but also to planned scalable culturing processes. We found organic osmolyte synthesis and membrane transporters were most responsive to changes in salinity. The osmolytes glutamate, glutamine and ectoine responded to salinity across temperature treatments. However, when media was supplemented with choline, glycine betaine appeared to replace ectoine. These results provide a baseline data set for metabolic activity under a variety of conditions that will influence future basic and applied V. natriegens research.

Project description:Parasite biology, by its very nature, cannot be understood without integrating it with that of the host, nor can the host response be adequately explained without considering the activity of the parasite. However, due to experimental limitations, molecular studies of parasite-host systems have been predominantly one-sided investigations focusing on either of the partners. Here we conduct a joint dual RNA-seq time course analysis of filarial parasite and host mosquito to better understand the parasite processes underlying development in, and interaction with, the host tissue from the establishment of infection to the emergence of infective-stage larva. Using the Brugia malayi-Aedes aegypti system, we report the parasite gene transcription dynamics, which exhibit a highly ordered developmental program consisting of a series of cyclical and state-transitioning temporal patterns. And, we contextualize these parasite data in relation to the concurrent dynamics of the host transcriptome. Comparative analyses using uninfected tissues and different host strains reveal the influence of parasite development on the host gene transcription as well as the influence of host environment on the parasite gene transcription. Furthermore, we critically evaluate the life-cycle transcriptome of B. malayi by comparing developmental stages in the mosquito relative to those in the mammalian host, providing insight into gene expression changes underpinning the mosquito-borne parasitic lifestyle of this heteroxenous parasite. Time-course mRNA profiles of filarial parasite Brugia malayi and host mosqutio Aedes aegypti were generated by deep sequencing using Illumina GAIIx.

Project description:Pathogenic bacteria can rapidly respond to stress environments, such as exposure to reactive oxygen species (ROS). The thiol (-SH) groups of cysteine residues in many proteins serve as redox-sensitive switches, providing triggers for ROS-mediated signaling events. In this study, we profiled the reversible thiol oxidation during ROS exposure of the proteome of Vibrio cholerae, a Gram-negative human pathogen that causes cholera. We identified posttranslational modifications of two cysteine residues of ArcA, a response regulator that is known to be phosphorylated under oxygen limiting conditions and regulates global carbon oxidation pathways. We showed that although ROS exposure abolished ArcA phosphorylation, it induced the formation of an intramolecular disulfide that promoted ArcA-ArcA interaction. Thiol oxidation of ArcA led to sustained ArcA activity and ROS resistance. We further demonstrated that V. cholerae ArcA cysteine residues were oxidized in cholera patient diarrheal stools, and that ArcA thiol oxidation is crucial for V. cholerae in vitro ROS resistance, colonization of ROS-rich gut niches, and environmental survival. Moreover, in other enteric pathogens such as Salmonella enterica, the cysteine residues in ArcA orthologs are conserved and thiol oxidation of ArcA plays important roles in ROS resistance both in vitro and in host cells. These results suggest that in enteric pathogens, as a response to ROS insults, thiol oxidation of ArcA is able to functionally mimic phosphorylation and retain ArcA activity, allowing for a balance in the expression of stress-related and pathogenesis-related genetic programs.

Project description:The shrimp aquaculture industry is vulnerable to large losses due to acute hepatopancreatic necrosis disease (AHPND), caused by the bacterium Vibrio parahaemolyticus. The mechanism by which the pathogen causes disease, and the host immune response, is not completely understood. The shrimp hepatopancreas is a multi-functional organ with roles in digestion, immunity, molting and reproduction. Therefore, we set out to characterize the cells of the hepatopancreas and the host response to Vibrio parahaemolyticus infection at single-cell resolution. The hepatopancreas from three individual shrimp were processed to create a single-cell transcriptomic atlas. Then the hepatopancreas from three Vibrio parahaemolyticus infected and two mocked treated shrimp were sampled for infection study. All single-cell libraries were generated using the 10X Genomics platform and sequenced on an Illumina sequencer. Data were aligned to the Litopenaeus vannamei reference genome using Cell Ranger. Seurat and clusterProfiler were used for downstream analyses. Cells of the hepatopancreas were characterized and the transcriptomic response to AHPND-causing V. parahaemolyticus was examined. Data will inform further functional studies and has the potential to aid in the development of novel preventative measures or treatments.

Project description:Type VI secretion systems (T6SS) are widely distributed among Vibrio species, yet their roles in the coexistence of toxigenic and non- toxigenic strains remain unclear. Here, we report a novel orphan T6SS effector-immunity module, TseVs-TsiVs, primarily harbored by non- toxigenic Vibrio cholerae. TseVs exhibits robust vibriocidal activity, specifically targeting susceptible Vibrios (lacking TsiVs). TseVs forms dual-membrane, ion-selective pores that collapse Na⁺/K⁺ homeostasis, resulting in membrane depolarization and ATP depletion. Remarkably, non-Vibrio bacteria evade TseVs through proton motive force (PMF)-dependent resilience, uncovering a previously unrecognized immunity-independent defense strategy. Furthermore, tseVs+ non- toxigenic V. cholerae strains are globally distributed and have dominated in recent decades, highlighting TseVs’s ecological significance in Vibrio population dynamics. By linking TseVs’s bioenergetic assassination to Vibrio population shifts, we demonstrate how T6SS effectors shape microbial genetic diversity. Our findings suggest that TseVs represents a promising model for precision antimicrobial strategies, minimizing collateral damage to commensal microbiota.

Project description:Although many members of the genus Vibrio are known to inhabit the marine photic zone, an understanding of the influence of light on the molecular physiology of Vibrio spp. has largely been neglected. To begin to characterize the photophysiology of one such Vibrio sp. (Vibrio campbellii ATCC strain BAA-1116) we used microarray-based expression profiling to compare the transcriptomes of illuminated versus dark cell cultures. Specficially, we compared the transcriptomes of wild type V. campbellii (STR) cells that were cultured in M9 minimal salts medium plus glucose under two conditions: (i) after 24 hours of continuous dark and (ii) after a 12 hour dark:12 hour light cycle (white light illumination at 54 µmol photons s-1 m-2). The results revealed a large photostimulon (differential expression of ~20% of the V. campbellii genome; adjusted p value < 0.0001) that surprisingly included ~75% of the type III secretion system (T3SS) genes which were found to be 1.6 – 5.4X more abundant in illuminated cultures. These findings, which were confirmed by quantitative reverse transcription PCR and quantitative membrane proteomics, strongly suggest that the photostimulon of strain BAA-1116 includes the T3SS.
