Project description:Saline is a key environmental parameter in aquaculture. Research on acute salinity stress can help farmers determine the optimal salinity level to reduce stress responses in fish larvae, improve survival rates, and increase production efficiencies. It can also aid in the selection or breeding of species suited to environments with specific salinities. However, the molecular mechanisms underlying the negative effects of high salinity on Pseudobagras ussuriensis and the regulatory mechanisms underlying its tolerance remain unclear. In this study, Pseudobagras ussuriensis was exposed to 10 g/L NaCl for 96 h, and the physiological and biochemical changes in the gill and kidney tissues were continuously monitored. Transcriptomic and metabolomic analyses of the gill and kidney tissues were conducted at 24 h to elucidate the molecular adaptation mechanisms to salt stress. A total of 2,554 differentially expressed genes (DEGs) were identified in gill and 1,066 DEGs in the kidney tissue, respectively. Compared with the control group, 826 genes were upregulated and 1,728 were downregulated in the gill tissue under acute salinity stress at 24 h, whereas 508 genes were upregulated and 558 were downregulated in the kidney tissue. These DEGs were involved in oxidative stress, energy metabolism, ion transport, and immune responses. Metabolomic analysis showed that compared to the control group, 85 differential metabolites (DMs) were identified in the gill tissue, with 49 upregulated and 36 downregulated, whereas 433 DMs were identified in the kidney tissue, with 252 upregulated and 181 downregulated. Notably, the levels of phosphatidylcholine (PC) and phosphatidylethanolamine (PE) in the kidney and phosphatidic acid (PA) and PC in the gills were significantly increased, while sphingomyelin (SM) decreased. Integrated analysis of metabolomics and transcriptomics through the KGML network map revealed the downregulation of ALDH, ALDH7A1, AOX, and HADHA in the gill, and ENPP1_3, CD203, L-glutamine (downregulated), and Succinic Acid (upregulated) in the kidney. This study combined metabolomics, transcriptomics, and physiological-biochemical analyses to provide new insights into the molecular mechanisms of oxidative stress, energy metabolism, and immune regulation in gill and kidney tissues under acute salinity stress.

Project description:In this study, RNA-Seq was used to reveal the differences of molecular pathways in hepatopancreas of O. niloticus adapated to water with salinity of 8 or 16 practical salinity (psu), respectively, with fish at freshwater as the control,. Significantly changed pathways were mainly related to lipid metabolism, glucose utilization, protein consumption, osmotic regulation, signal transduction and immunology. Based on the tendencies from freshwater to 8 or 16 psu, the differentially expressed gene unions were categorized into eight unique models, which were further classified into three categories which were constant-change (either keep increasing or decreasing), change-then-stable and stable-then-change. In constant-change category, steroid biosynthesis, steroid hormone biosynthesis, fat digestion and absorption, complement and coagulation cascades were extremely significantly affected by ambient salinity (P < 0.01), indicating that these pathways play pivotal roles in molecular response to salinity acclimation from freshwater to saline water in O. niloticus, and should be the main research focus in the future. In change-then-stable category, ribosome, oxidative phosphorylation, peroxisome proliferator-activated receptors (PPAR) signaling pathway, fat digestion and absorption changed significantly with ambient increasing salinity (P < 0.01), showing these pathways were sensitive to environmental salinity variation, but had a response threshold, and would stop changing once salinity exceeds the threshold. In stable-then-change category, protein export, protein processing in endoplasmic reticulum, tight junction, thyroid hormone synthesis, antigen processing and presentation, glycolysis/gluconeogenesis and glycosaminoglycan biosynthesis - keratan sulfate were the top changed pathways (P < 0.01), suggesting that these pathways were not sensitive to salinity variation, but these pathways will respond significantly under salinity exceeding a certain level. The pathways and genes reported in this study laid on a solid foundation for future studies in understanding the underlying mechanism for salinity adaptation of freshwater fish.

Project description:Ambient salinity is one of the crucial abiotic factors that poses substantial impacts on fish growth, development and reproduction. Greater amberjack (Seriola dumerili) is of high economic value because, and its reproduction and survival are sensitive to water salinity. To better understand the molecular adaptive mechanism to salinity fluctuations in greater amberjack, we performed comparative transcriptome analysis for gill and kidney between the optimum salinity (30 ppt, CK) and undesired regimes (10 and 40 ppt). For the gill, the skeletal development was provoked upon either hypo- or hyper-salinity stimuli, and the development of pronephros, as well as vascular endothelial cells and cortisol-mediated mitochondria-rich cell, was activated in response to the salinity alterations in kidney. These enhancements may encourage the maintenance of the gill and kidney structures and alleviate the salinity-induced damage. Ion channels NKCC1 and CFTR and the transporters for ammonium and other substances were highly upregulated in the gills and kidney, respectively, which act important roles in the osmoregulation of greater amberjack. More important, undesirable alterations of ambient salinity were found to pose adverse impacts on the immune function of greater amberjack, which may increase the risk of pathogen infection and reduce the security and yield of aquaculture of greater amberjack. In addition, deviation from the optimum salinity condition may result in undesirable uptake and accumulation of environmental toxins in greater amberjack, which attracts further attention to the food safety. Collectively, these novel findings advance our knowledge on adaptative mechanisms to ambient salinity oscillations in greater amberjack and provide a theoretical guidance for the optimal breeding mode for the aquaculture of greater amberjack.

Project description:Melatonin plays a potential role in multiple plant developmental processes and stress response. However, there are no reports regarding exogenous melatonin promoting rice seed germination under salinity and nor about the underlying molecular mechanisms at genome-wide. Here, we revealed that exogenous application of melatonin conferred roles in promoting rice seed germination under salinity. The putative molecular mechanisms of exogenous melatonin in promoting rice seed germination under high salinity were further investigated through metabolomic and transcriptomic analyses. The results state clearly that the phytohormone contents were reprogrammed, the activities of SOD, CAT, POD were enhanced, and the total antioxidant capacity was activated under salinity by exogenous melatonin. Additionally, melatonin-pre-treated seeds exhibited higher concentrations of glycosides than non-treated seeds under salinity. Furthermore, exogenous melatonin alleviated the accumulation of fatty acids induced by salinity. Genome-wide transcriptomic profiling identified 7160 transcripts that were differentially expressed in NaCl, MT100 and control. Pathway and GO term enrichment analysis revealed that genes involved in the response to oxidative stress, hormone metabolism, heme building, mitochondrion, tricarboxylic acid transformation were altered after melatonin pre-treatment under salinity. This study provides the first evidence of the protective roles of exogenous melatonin in increasing rice seed germination under salt stress, mainly via activation of antioxidants and modulation of metabolic homeostasis.

Project description:To understand the sensitivity of larval lice to changing environmental parameters we applied a 38K oligo microarray and characterized transcriptome responses after 24 hour exposures to reduced salinity (30-10 parts per thousand (ppt)) or varied temperature (16-4M-BM-:C). To further characterize the effects of salinity, we profiled expression changes over a range of seawater with single increment differences (30-25 ppt). Three separate projects. Low resolution salinity (aka wide range or LR_SAL) contains 2 duplicate experiments, each with pools of lice (~500 copepodid lice per beaker; n=3 beakers per condition), incubated for 24 hr at 10M-BM-:C diluted to 30, 25, 20, or 10 parts per thousand (ppt). Temperature study conducted as above, but with a constant salinity of 30 ppt and changed temperature of 4, 10, or 16M-BM-:C. High resolution salinity conducted as above, but with no experiment replication, but with biological repication of n=6 beakers per condition, and with a range of 25, 26, 27, 28, 29, 30 ppt salinity. Please note that each experiment was normalized separately.
