Untargeted Metabolomic Profiling of Left Ventricular Myocardium in a Mouse Model of Diabetic Cardiomyopathy

ABSTRACT: This study characterizes metabolic alterations in the left ventricular myocardium of a high-fat diet and streptozotocin-induced mouse model of diabetic cardiomyopathy. Untargeted LC–MS/MS metabolomics was performed using a Vanquish UHPLC system coupled to an Orbitrap Exploris 120 mass spectrometer. Data were processed using XCMS and analyzed by principal component analysis, OPLS-DA, differential metabolite screening, hierarchical clustering, and KEGG- and MetaboAnalyst-based pathway analysis. Pooled quality-control samples were used throughout the analytical sequence to monitor system stability.
