Serum Metabolomics Reveals Stage-Dependent Metabolic Transitions During CIDR-PGF2alpha-Induced Estrus in Kazak Ewes

ABSTRACT: This study investigated serum metabolic transitions during CIDR-PGF2alpha-induced estrus synchronization in Kazak ewes using UHPLC-MS/MS untargeted metabolomics. Eighteen Kazak ewes were assigned to three reproductive stages: luteal phase (LP), luteolysis phase (LDP), and estrus phase (OES), with six animals per group. Serum samples were analyzed by UHPLC-MS/MS on an Orbitrap Exploris 120 instrument using HILIC chromatography (BEH Amide column) in both positive and negative ionization modes. A total of 1,861 metabolites were annotated. Key biomarkers identified include choline, betaine aldehyde, sphingosine, 3-hydroxybutyrylcarnitine, and pregnanediol 3-O-glucuronide, reflecting stage-dependent shifts in phospholipid metabolism, energy utilization, and steroid hormone pathways.
