Non-targeted metabolomic profiling reveals Flo8-associated metabolic alterations in Candida glabrata

ABSTRACT: Flo8 is a transcriptional regulator involved in multiple physiological processes in Candida glabrata. To investigate the metabolic changes associated with Flo8, non-targeted metabolomic profiling was performed using the reference strain and a FLO8 deletion mutant. Intracellular metabolites were extracted from cells cultured under the same conditions and analyzed by liquid chromatography–mass spectrometry. Comparative metabolomic analysis revealed extensive alterations in metabolic pathways related to vitamin B6 metabolism, amino acid metabolism and glutathione metabolism. In particular, the FLO8 deletion mutant exhibited altered levels of pyridoxal 5′-phosphate and its related vitamers, together with changes in glutathione-associated metabolites. This dataset provides a global view of Flo8-dependent metabolic regulation and supports further investigation of the relationship between PLP homeostasis, mitochondrial function, and iron metabolism in C. glabrata.
