Project description:This study was designed to investigate gene expression in kidneys of adult Sabra rats (SBH/y and SBN/y rat strains) with two indwelling kidneys or after uni-ninephrectomy, seeking those genes that are differentially expressed between the two strains, and between animals with one or two kidneys. SBH/y after uninephrectomy develop proteinuria to a much greater extent than SBN/y. The study was performed as part of an overall effort to detect the genes that are associated with the pathophysiology of proteinuria. Keywords: Analysis of genes that are differentially expressed in the kidneys, contrasting between an animal strain that tends to develop proteinuria which is amplified by uninephrectomy and another strain that is relatively resistant to the development of proteinuria The experiment was performed in male animals after weaning. Four groups were studied: SBN/y with 2 kidneys (ham uni-nephrectomy), SBN/y after uni-nephrectomy, SBH/y with 2 kidneys (sham uninephrectomy) and SBH/y after uni-nephrectomy. Animals were provided rat chow ad libitum. At age 4 months, 3 months after uninephrectomy or sham operation, animals were sacrificed under ether anesthesia, killed by exsanguination and the kidneys were rapidly removed and snap frozen with liquid nitrogen. The kidneys were stored at -800C until RNA was extracted for the differentiale xpression experiment.

After 4 additional days (during which blood pressure measurements were obtained), they were injected daily subcut. with either saline (1ml/kg), adrenocorticotropic hormone (ACTH, 0.2mg/kg) or dexamethasone (DEX, 0.02mg/kg) for 8 days before the right kidney was removed. ACTH and DEX treatment caused hypertension in the rats as illustrated by increased systolic blood pressure. The gene expression patterns of the kidneys were analysed using Affymetrix arrays. Experiment Overall Design: The left kidney of each rat acts as a control for the treated right kidney. Two rats each were treated with either saline, ACTH or DEX.

Project description:Puberty unmasks or accelerates nephropathies, including the nephropathy of diabetes mellitus (DM). A number of cellular systems implicated in the kidney disease of DM interweave, forming an interdependent functional web. We performed focused microarray analysis to test the hypothesis that one or more genes in the transforming growth factor beta (TGF-β) signaling system would be differentially regulated in male rats depending on the age of onset of DM. Experiment Overall Design: Male rat littermates began the 6-week-protocol at 4 or 14 weeks of age with injection of streptozocin, 65 mg/kg iv, or equal volume of vehicle. 3 days later insulin palmitate or palmitate vehicle pellets were implanted once hyperglycemia was confirmed. Rats received ad lib food and water for 6 weeks and maintained blood glucose levels 350-350 mg/dl in diabetic groups. Kidneys were removed under isoflurane anesthesia. The cortex was rapidly dissected from the medulla and snap-frozen. Cortex was stored at -80 until all rats had completed the protocol. RNA was isolated from renal cortex and the transcriptome analyzed using gene chips with more than 30,000 transcripts. Age-specific effects of DM were demonstrated for 1,760 transcripts. Analysis then focused on 89 genes involved in the TGF-β signaling pathway.

Project description:Promoter methylation analysis of hypothalamc DNA from female rats at different juvenile developmental reproductive stages. Results provide insight into the role of the hypothalamus in controlling the onset of puberty. SD rats were housed (4/cage) in a controlled environment and euthanized at different ages (Early Juvenile: 21 days, Late Juvenile: 28 days, Late Proestus (the day of first ovulation): 31 days. Rats were anesthetized and brains were rapidly removed. The hypothalamus was dissected away from the rest of the brain and flash frozen. Total RNA and DNA was isolated from each sample using AllPrep DNA/RNA Mini Kit-Qiagen (Valencia, CA).

Project description:Hybridization of one kidney of cortisol treated fish vs. one kidney of control fish. Kidneys were collected from untreated juvenile sea bream (n=4) and from fish, which received for 72h a coconut-oil implant containing 10mg/Kg (fish wet weight) (n=4) cortisol. Experiments were carried out at the University of the Algarve, Portugal in accordance with National legislation for the welfare of animals. Experiments were conducted in two 125 l cylindriconical tanks supplied with a continuous through-flow of oxygenated seawater at 20+1 °C using juvenile sea bream (25 g + 3 g) adapted for 1 week to the experimental conditions. One tank contained 8 untreated fish (control) and the other tank 8 cortisol treated fish and the end of experiments fish were removed form tanks, decapitated and the kidneys rapidly removed and place in RNAlater (Qiagen) at –20 °C. No mortality occurred in the control tank but 2 fish died in the cortisol treated tank. Keywords: other
