Project description:The objective of this research is to compare the differentially expressed genes in the vascular tissues of DAPK1 wild-type (WT) and DAPK1 knockout (KO) mice after being induced with or without Ang II. WT (n=10) mice were randomly divided into two groups: WT+NS group and WT+Ang II group (n=5 for each group). KO (n=10) mice were also randomly divided into two groups: KO+NS group and KO+Ang II group (n=5 for each group). Then the vascular tissues were used to identify differentially expressed genes among different groups.

Expression data of E13.5 placenta from intra-cytoplasmic sperm injection(ICSI)-F2 offspring

Project description:Various behavioural abnormalities and congenital anomalies were observed in the F2 offspring of ICSI (Kanatsu-Shinohara et al., 2023, J. Clin.). Invest. 133(22): e170140). The data presented here aim to clarify the cause of these abnormalities. Gene expression in the ICSI-F2 placenta was investigated using RNA sequencing (RNA-seq). ICSI was performed using C57BL/6 epididymal sperm and oocytes. Male ICSI-F2 offspring was mated with wild-type C57BL/6 female, and placentae recovered from pregnant females at E13.5 were examined for gene expression. Each group contains five biological replicates.

Transcription profiling of human xenografted radioresistant and radiosensitive tumors before and after irradiation

Project description:Nu61, a radiation-resistant human tumor xenograft, was selected from a parental radiosensitive tumor SCC-61 by eight serial cycles of passage in athymic nude mice and in vivo irradiation. Obtained tumors were profiled using Affymetrix U133A arrays. Most abundant gene pattern associated with radioresistant phenotype was presented by IFN-inducible, Stat1-dependent pathway Experiment Overall Design: Obtained tumors were established as xenografts in nude mice. RNA were purified from each xenograft, normalized by concentration and used for hybridization with each array. Three arrays were used for each group (un-treated nu61 and SCC61 and irradiated nu61 and SCC61 5 and 24 hours post-IR). Each group contained from 5 animals; tumors were randomized by size and three tumors from each group were used for profiling

Lung gene expression after long-term styrene inhalation in three strains of C57BL/6 mice, and CD-1 mice

Project description:Lung gene expression after long-term (26 weeks, 52 weeks, 78 weeks & 104 weeks) styrene inhalation exposure at a single concentration (120ppm) in three strains of C57BL/6 mice -- wild-type (WT), CYP2F2 knockout (KO), and CYP2F1 humanized (TG) mice, and CD-1 mice. These data examine transcriptomic changes in lung tissue after long-term styrene inhalation in male C57Bl/6 and CD-1 mice. Styrene causes increased lung tumors in mice, but not in rats. Mouse lung tumors were found mostly at the conclusion of a life-time (104 weeks for males) exposure study and most were benign. Styrene is largely negative in genotoxicity assays. Styrene metabolism by CYP2F2 produced a different metabolite pattern in mouse lung than in liver or in rats or humans. The purpose of this study was to use genomic analyses to further investigate potential modes of action (MoA) of styrene in mice after long-term exposure to styrene. Mice strains exposed were C57BL/6 wild-type (WT), CYP2F2 knockout (-/-; KO) and CYP2F21 humanized transgenic (2F2-KO + 2F1,2A13,2B6-transgenic, TG), and CD-1 male mice using 120 ppm styrene at 6 hr/day 5 days/wk for 26, 52, 78 and 104 weeks. Lungs were analyzed by Affymetrix whole genome microarrays for each strain relative to sample time-specific vehicle controls for each strain.

Total RNA sequencing of mouse LSK cells in E14.5 fetal liver

Project description:The study is to obtain a transcriptome-wide map of prenatal murine stem and progenitor cells. VavCre+ mice were used for conditional knockout of Ddx41. Wild-type, Ddx41 heterozygous (Ddx41+/-) and Ddx41 knockout (Ddx41-/-) LSK cells from embryonic 14.5 day fetal liver were harvested (three samples from each group) for RNA. Gene expression analysis and alternative splicing analysis were performed. RNA-seq data were confirmed by RT-qPCR and regular PCR followed by gel quantification. A global change of gene expression and substantial differential splicing events were observed in KO samples compared to WT and het.

RNA-seq analysis was performed to examine the differential express pattern of the two type of xenografts.

Project description:Tumor-draining lymph nodes (TdLNs) were collected from wild-type (WT) and tenascin-C knockout (TNC-KO) C57BL/6 mice (N = 3 per group), each bearing 4NQO-induced tongue tumors. Both male and female mice were included. Beginning at 12 weeks of age, mice were exposed to 4NQO via drinking water for 16 weeks. Tumors were then subjected to a single 2 Gy irradiation. TdLNs were harvested six weeks after irradiation, and total RNA was isolated for bulk RNA-seq analysis.
