The expression of a conditional Ssn6HA-ts strain was compared against the expression in a WT strain as described before (Xue et al, 2004), except that cells were shifted to 36 deg 1 h prior to RNA extraction. Project description:Here, we report the high-throughput profiling of histone modification (H3K9me2) in fission yeast Schizosaccharomyces pombe. We generated genome-wide H3K9me2 maps of fission yeast mutants in swo1-26 (temperature sensitive, ts) cells at 25℃ and 37℃. We find that H3K9me2 enrichment at heterochromatin regions, especially at the mating-type locus and subtelomeres, is compromised, suggesting heterochromatin assembly defects.
Project description:Expression profiles of polg mutant cells reveal that many genes encoding proteins involved in cell wall biogenesis and stress response are induced, suggesting that polg mutant cells attempt to maintain growth potential and undergo extensive oxidative metabolism. Conversely, many genes encoding proteins involved in ribosome biogenesis and respiration are repressed, indicating that cells depleted of mtDNA are adapted to grow slowly in absence of mitochondrial function.
