To develop an improved rodent system for the study of chemical warfare nerve agents, a serum carboxylesterase knock out mouse model was made (Es-1 knock out). This study utilized mRNA analysis to evaluate the effects of knocking out the serum carboxylesterase in a mouse model.

Project description:Transcriptomic analysis of mosquito larvae exposed to LC50 of Cry11Aa toxin from Bti at different times was conducted to determine defense response pathways in order to better understand the toxin's mode of action and identify possible cellular targets to enhance toxin activity. 4 replicates of 5 intoxication time points and 3 replicates of 2 control timepoints were sequenced on an Illumina platform, analyzed for differential expression with two packages. Project description:To promote infections, pathogens exploit host cell machineries including structural elements of the plasma membrane. Studying these interactions and identifying molecular players is an ideal way to gain insights into the fundamental biology of the host cell. Here, we used the anthrax toxin to screen a library of 1500 regulatory, cell surface, and membrane trafficking genes for their involvement in the intoxication process. We found that ER-Golgi localized proteins TMED2 and TMED10 are required for toxin oligomerization at the plasma-membrane of human cells, an essential step dependant on localization to cholesterol-rich lipid nanodomains. Biochemical, morphological and mechanistic analyses showed that TMED2 and TMED10 are essential components of a complex that operates the exchange of both cholesterol and ceramides at ER-Golgi membrane contact sites. Overall, this study of anthrax intoxication led to the discovery that lipid compositional remodelling at ER-Golgi interfaces fully controls the formation of functional membrane nanodomains at the cell surface.

Project description:Opioid abuse poses significant risk to individuals in the United States and epigenetic changes are a leading potential biomarker of opioid abuse. Current evidence, however, is mostly limited to candidate gene analysis in whole blood. Here, we provide Illumina HumanMethylationEPIC array data generated in dorsolateral prefrontal cortex tissue from 153 deceased invidiuals: 72 who died of acute opioid intoxication, 53 psychiatric controls, and 28 normal controls. Using these data, we conducted an epigenome-wide association study and identified one CpG site within a gene (NTN1) that may be related to opioid use. We also detected accelerated PhenoAge in opioid samples compared to control samples.

Project description:Cellular immunotherapies are rapidly gaining clinical importance, yet predictive platforms for modeling their mode of action are lacking. Here, we unravel the behavioral and underlying molecular mechanisms that govern breast cancer targeting by an emerging cancer metabolome-sensing immunotherapy: T cells engineered to express a V9/V2 T cell receptor (TEGs). Using a dynamic immuno-organoid 3D imaging-transcriptomics platform, BEHAV3D, we first demonstrate that TEGs kill organoids of multiple breast cancer subtypes, yet with varying sensitivity between individual patient-derived organoids (PDOs) and even among individual PDOs derived from the same patient. Furthermore, live-tracking of over 120,000 TEGs revealed a diverse behavioral landscape and identified a ‘super-engager’ cluster with serial killing capability. Inference of single-cell behavior with transcriptomics unraveled gene signatures linked to specific behaviors. The most notable gene signature identified ‘super engager’ killer TEGs and contained multiple genes previously unstudied in T cells. Furthermore, guided by a dynamic type 1 interferon (IFN-I) signaling module specifically induced by the highest TEG-sensitive organoid culture, we show that IFN-I can prime resistant organoids for¬ TEG-mediated killing. Thus, BEHAV3D characterizes behavioral-phenotypic heterogeneity of cellular immunotherapies and holds promise for improving solid tumor-targeting in a patient-specific manner.

Project description:The impact of alcohol abuse on AD is poorly understood. Here we show that the onset of neurocognitive impairment in a mouse model of AD is hastened by repeated alcohol intoxication through exposure to alcohol vapor and we provide a comprehensive gene expression dataset of the prefrontal cortex by single nucleus RNA sequencing of 113,242 cells. Several genes previously associated with AD in humans by GWAS were differentially regulated in specific neuronal populations. Gene expression patterns of AD mice with a history of alcohol intoxication were more similar to the gene expression signatures of older AD mice with more advanced disease and cognitive impairment than to those of younger AD mice with prodromic disease, suggesting that alcohol promotes transcriptional changes consistent with AD progression. Our gene expression dataset at the single cell level provides a unique resource for investigations of the molecular bases of the detrimental role of excessive alcohol intake in AD.
