Across the time course, differentially expressed genes involved in terpenoid backbone biosynthesis were significantly enriched, while JAZ genes involved in jasmonic acid pathway and TGA genes involved in salicylic acid pathway were significantly enriched at later time point after treatment.
Project description:Smilax glabra Roxb, a traditional Chinese herb, has been widely used for folk medicine. Previous studies have found that it has various pharmacological activities, such as cytotoxic, anti-inflammation, anti-oxidant, hepatoprotective, and cardiovascular system protective activities. However, its roles in adipogenesis are poorly understood. We hypothesized that Smilax glabra Roxb and its main component may play important roles in regulating adipocyte diffrentiation and function. To test this hypothesis, we performed RNA-Seq on 3T3-L1 adipocytes treated with and without pure total flavonoids from Smilax glabra Roxb.

Project description:We used Affymetrix 1.0 ST arrays to compare the transcriptome of 4 weeks‐day‐old plants sprayed with 250uM of MeJA or mock (control). For the experiments we used wild-type (Col-0), phyB-9 and jaz10 single mutants and jaz10phyb double mutant. Samples were harvested after 3hours of induction with MeJA or mock. We performed a first analysis with the transcriptomes of wild‐type (Col-0) to identify those group of genes that are up‐ and down‐regulated by MeJA treatment (2 fold-changes). We founded 719 genes whose expression was statistically significantly different between the two treatments. To gain knowledge into the regulation of phyB over these set of 719 genes, we contrasted their expression levels against phyB transcriptome. By cluster analysis, we identified five major groups of genes, where in two of them the transcript levels increased in presence of MeJA (clusters 1 and 2) and three whose expression decrease after MeJA treatment (Cluster 3, 4 and 5).
Project description:Sugarcane one-eyed seed sets (genotype SP80-3280, CTC, Brazil) were planted in 200 ml plastic cups containing a commercial planting mix (Plantmax, Eucatex) and cultivated for 20 days under greenhouse conditions. The plantlets were subsequently transferred to a growth chamber at 26°C on a 16 h/8 h light/dark cycle with a photon flux density of 70 µE.m-2.s-1 to acclimate. Plantlets were then sprayed with a 100 µmol.L-1 MeJA solution (Bedoukian Research Inc., Danbury, CT), whereas control plantlets were treated with distilled water. Leaves were collected 0, 1, 6and 12 h after exposure to MeJA and immediately frozen in liquid nitrogen. Six plantlets were used for each time point. Project description:Using this microarray data, we identified 19898 probes (717 upregulated and 1512 downregulated in the mock- and MeJA-treated leaf samples at ‘1wk’ and ‘2wk’ stages; 17669 with differential expression in these samples), compared with the mock-treated sample at ‘0wk’ stage. This work aims to identify the genes related to MeJA-induced senescence of tobacco whole-plant, and found several genes differentially expressed between the mock- and MeJA-treated samples at the same stage.
