Collagen induced arthritis plasma proteomic analysis

ABSTRACT: The aim of the project is to analyze the plasma proteomic profile of an animal model of arthritis (Collagen Induced Arthritis; CIA) in response to different treatments. For this, plasma samples from mice under five different treatments were analyzed (N=6 per group): Vehicle, CIA, CIA + Δ9-THCA-A (20 mg / kg), CIA + Δ9-THCA-A (20 mg / kg) + T0070907 (5 mg / kg) and CIA + Δ9-THCA-A (20 mg / kg) + SR141716A (5 mg / kg).

Docosahexaenoic and Arachidonic Free Fatty Acids Exert Different Antitumor Activities on Colorectal Cancer Cells

Project description:Two PUFA, docosahexaenoic (DHA, 22: 6n3) and arachidonic acids (ARA, 20: 4n6), as well as their derivatives such as eicosanoids, regulate different activities, which include changes in receptor signaling, the composition of rafts, cell metabolism, and membrane structures. These also modify the function of transcription factors and their target genes, a key step essential for the function of FA-activated signaling. This work is focused to determine the antitumor actions of these compounds linked to the regulation of gene transcription on HT-29 colorectal cancer. For this, we performed antiproliferative antitumour assay, LDH breakage test, caspase-3 production, and proteome changes were assessed by SWATH-MS quantitative proteomics followed by pathway analysis in order to find out which molecular mechanisms were being affected. In all cases tested, DHA exercised antitumor actions to a higher extent than ARA, acting mainly by means of down-regulating most of the proteasome system particles, while ARA presented a heavy effect on all the six DNA replication helicase particles but did not affect proteasome. The results indicated that both DHA and ARA stopped colorectal cancer growth and proliferation, thus they represent the ideal candidates as chemopreventive agents.

Breaking the mucin barrier: a new affinity chromatography-mass spectrometry approach to unveil potential cell markers and pathways altered in Pseudomyxoma peritonei

Project description:Pseudomyxoma peritonei (PMP) is a rare, malignant tumour with unknown underlying molecular mechanisms characterised by a progressive accumulation of mucin and secreting tumour cells within the abdomen and pelvis. Cytoreductive surgery (CRS) combined with hyperthermic intraperitoneal chemotherapy (HIPEC) is the only therapeutic option currently available for its treatment and recurrence with a fatal outcome is common despite its use. The lack of information available about the molecular mechanisms underlying this condition is mainly due to the physicochemical characteristics of mucin, whose main purpose is to protect epithelial cells, which makes it inaccessible to a proteomic analysis. In this paper we describe the first protocol available to break this barrier and offer a chance to isolate proteins from soft and hard mucinous tumour tissues. This approach is based on the depletion of glycoproteins, immunoglobins, and albumins by affinity liquid chromatography, which makes the mucin available for label free quantification by mass spectrometry. As in the case of non mucinous tissues, our protocol yields a mix of total proteins and has allowed us to establish the first protein profile in mucinous tumours. In addition, our protein-protein interaction networks and pathway enrichment analyses revealed MUC13 and TFF1 as potential therapeutic targets in this disease.

SWATH differential abundance proteomics and cellular assays show in vitro anticancer activity of arachidonic and docosahexaenoic acid monoacylglycerols in HT-29 colorectal cancer cell line

Project description:Colorectal cancer (CRC) is one of the most common and mortal types of cancer. There is increasing evidence that some polyunsaturated fatty acids (PUFA) exercise specific inhibitory actions on cancer cells through different mechanisms, as a previous study on the effect on CRC cells of two PUFA free fatty acids (FFA), docosahexaenoic acid (DHA, 22:6n3) and arachidonic acid (ARA, 20:4n6)-FFA, shown. Here we have used the same study design and technology to investigate the actions of DHA and ARA-monoacylglycerols (MAG), and we have compared the results with the previous study of the corresponding FFA. Cell assays revealed that ARA- and DHA-MAG exercised dose- and time-dependent antiproliferative actions, with DHA-MAG acting on cancer cells more efficiently than ARA-MAG. SWATH-MS massive quantitative proteomics, validated by parallel reaction monitoring and followed by pathway analysis, revealed that DHA-MAG had a massive effect in the proteasome complex, while ARA-MAG main effect was related to DNA replication. Prostaglandin synthesis also resulted inhibited by DHA-MAG. Results clearly demonstrated the ability of MAGs to induce cell death in colon cancer cells and suggested a direct relationship between chemical structure and effect.

Project description:Colorectal cancer is one of the most common and widespread disease in the world and the third type of cancer causing a high mortality rate. There is increasing evidence that some polyunsaturated fatty acids (PUFAs) are involved in the reduction of cancer risk and progression. Recent studies showed that sn-2 monoacylglycerols (MAG) exercise specific inhibitory actions on cancer cells through different mechanisms. However, the anticancer effect of PUFA-based MAGs on colorectal cancer has yet to be assessed. In this work we investigated the actions of two PUFAs, docosahexaenoic (DHA, 22: 6n3) and arachidonic acids (ARA, 20:4n6), both as MAG, on colon cancer human (HT-29) cell line. We performed the MTT test, LDH, and caspase-3 assays, while global proteome changes were assessed by SWATH-MS quantitative proteomics followed by pathway analysis in order to find out which molecular mechanisms were being affected. It has been proven that ARA- and DHA-MAG exercises dose- and time-dependent antiproliferative actions. In all cases, DHA-MAG acts on cancer cells more efficiently than ARA-MAG. Results clearly demonstrate the ability of MAGs to induce cell death in colon cancer cells and suggest a direct relationship between their chemical structure and their potency. Therefore, sn-2 MAG are suitable candidate for the production of new functional ingredients.

SWATH proteomic profiling of prostate cancer cells identifies NUSAP1 as a biomarker of response to Galiellalactone

Project description:Galiellalactone (GL) is a fungal metabolite that presents antitumor and anti-inflammatory activities in vitro and in vivo. Previous studies have shown that GL targets NF-KB and STAT3 pathways and induces G2/M cell cycle arrest in androgen-insensitive prostate cancer cells. In this study, we show that GL-induced cell cycle arrest is independent of the NF-KB and STAT3 pathways in DU145 and PC-3 cells, and also that GL did not affect cell cycling in androgen-sensitive prostate cancer cell such as LNCaP and 22Rv1 cells. In addition, we showed confluence resistance to GL in DU145 cells. Using a SWATH proteomic approach we identified the down-regulation of Nucleolar and spindle associated protein 1 (NUSAP1) under DU145 confluence and in LNCaP cells. Also, the inhibition of NUSAP1 by siRNAs induced resistance to GL in DU145 cells, suggesting that NUSAP1 may be a target for GL and could be useful as biomarker for responsiveness of the antitumor activity of GL.

Proteomic profiling in mouse kidneys post-UUO

Project description:Increased export of transglutaminase-2 (TG2) by tubular epithelial cells (TECs) into the surrounding interstitium modifies the extracellular homeostatic balance leading to fibrotic membrane expansion. Although silencing of extracellular TG2 ameliorates progressive kidney scarring in animal models of chronic kidney disease, the pathway through which TG2 is secreted from TECs and contributes to disease progression has not been elucidated. In this study, we developed a global proteomic approach to identify binding partners of TG2 responsible for TG2 externalization in kidneys subjected to unilateral ureteric obstruction, using TG2-knockout kidneys as negative controls. We report a robust and unbiased analysis of the membrane interactome of TG2 in fibrotic kidneys relative to the entire proteome post-UUO detected by SWATH-mass spectrometry.
