Nucleus-Translocated GCLM Promotes Chemoresistance in Colorectal Cancer through a Moonlighting Function

Project description:Emerging evidence indicates that metabolic enzymes perform moonlighting functions during tumor progression, including the modulation of chemoresistance. However, the underlying mechanisms of these functions remain elusive. In this study, utilizing a metabolic CRISPR-Cas9 knockout library screen, we observed that loss of Glutamate-cysteine ligase modifier subunit (GCLM), a rate-limiting enzyme in glutathione biosynthesis, noticeably heightens the sensitivity of colorectal cancer (CRC) cells to platinum-based chemotherapy. Mechanistically, we unveil a noncanonical mechanism through which nuclear GCLM competitively interacts with NF-kappa-B-repressing factor (NKRF), a known inhibitor of NF-κB signaling, to promote NF-κB activity and subsequently facilitate chemoresistance. In response to platinum drug treatment, P38 MAPK phosphorylates GCLM at T17, resulting in its recognition by importin a5 and subsequent nuclear translocation. Furthermore, elevated expression of nuclear GCLM correlates with unfavorable prognosis and poor benefit from standard chemotherapy. Overall, our work shed light on the essential nonmetabolic role and posttranslational regulatory mechanism of GCLM in enhancing NF-κB activity and subsequent chemoresistance.

A combined omics approach to elucidate the molecular interplay behind a beneficial Arabidopsis-Caulobacter interaction

Project description:Plants rely on complex signaling networks to regulate growth and to interact with their environment, including beneficial microbes such as plant growth–promoting rhizobacteria (PGPR). Among these microbes, Caulobacter RHG1 has been shown to enhance plant growth and is recognized by Arabidopsis roots via specific receptor-like kinases (RLKs), but the molecular mechanisms underlying the plant growth-promotion effect remain largely unknown. To investigate these mechanisms, we used an integrative approach combining proteomics and phosphoproteomics to explore how RHG1 stimulates growth in Arabidopsis thaliana. Our phosphoproteome analysis revealed that RHG1 triggers a range of signaling events in the root, involving two RLKs (LysM RLK1-INTERACTING KINASE 1 [LIK1] and an uncharacterized RLK, AT5G49770). Notably, via the proteome analysis, we revealed that upon RHG1 treatment, defense proteins are downregulated early. Later on, developmental processes, such as those related to ribosome biogenesis and maturation, are involved in the growth-promotion effect and might lead to enhanced cell division and/or activity. These findings suggest that RHG1 is perceived at the root-level and this, via phosphorylation-based signaling pathways, triggers early protein-based effects that are characterized by timely modulation of defense and developmental processes, which eventually lead to plant growth promotion. Our data, combined with that of previous reports, points to the involvement of multiple signaling pathways, suggesting that RHG1 activates a broad network of regulatory mechanisms that extend beyond a single pathway. Understanding the mechanisms behind such beneficial plant-microbe interactions can pave the way for more effective commercialization of PGPR-based agricultural products, and our approach offers valuable insights for future research in this field.

Rise in KRT17 sensitizes platinum-resistant ovarian cancer organoids to targeted therapies

Project description:Variable responses to platinum chemotherapy and the development of resistant disease drive high mortality in high-grade serous ovarian cancer (HGSOC). Here, we developed the organoid drug resistance assay (ODR-test) to quantify the patient-specific efficacy of platinum treatment and generated post-platinum lines as an in vitro model of platinum-resistant ovarian cancer (PROC). We observed a gradual increase in resistance, mirroring the clinical outcomes of patients. Transcriptional and proteomic analyses of post-platinum phenotypes revealed changes in cell adhesion and differentiation as adaptive mechanisms irrespective of the basal resistance level. We found that Keratin 17 (KRT17) is a mediator of developing resistance and validated its function by CRISPR/Cas9 and overexpression. Additionally, we determined KRT17 (K-score) as a significant negative prognostic factor for survival based on analysis of a tumor microarray (N=377) of patients with advanced HGSOC. In organoids, increased KRT17 levels enhanced sensitivity to PI3K/Akt inhibitors Alpelisib and Afuresertib, highlighting the potential of KRT17 as a stratification biomarker for targeted therapies. These findings quantify patient-specific platinum response, identify PROC via KRT17 as a biomarker, and propose alternative targeted therapy for this challenging group of patients.

Project description:Fasciola hepatica represents a socioeconomically important species of parasitic fluke which causes significant economic losses in the livestock industry. During an indirect life cycle, immature eggs are passed out of the host in the stool and embyonate in the external environment until the release of miracidium which invades a freshwater snail - intermediate host. By utilization of RNA-seq technique (Illumina NextSeq 500), differentially expressed genes were analysed in F. hepatica eggs during the maturation process in three time points: 0 days (freshly laid eggs by adult F. hepatica individuals) and after incubation for 5 and 10 days.
