Iron regulatory pathways differentially expressed during Madurella mycetomatis grain development in Galleria mellonella

ABSTRACT: Mycetoma is a chronic granulomatous infection of the subcutaneous tissue, most often caused by the fungal pathogen Madurella mycetomatis. Characteristic of the infection is the formation of grains. However, knowledge of the function and formation of the grain is limited. To map the processes leading to M. mycetomatis grain formation, we used a Galleria mellonella larvae infection model and transcriptomic profiling. G. mellonella larvae were infected with M. mycetomatis and after 4h, 24h, 72h and 168h post-inoculation, RNA was extracted from larval content and sequenced. Differential Gene Expression analysis revealed that 3,498 G. mellonella and 136 M. mycetomatis genes were differentially expressed during infection. Among these genes, genes related to iron transport were highly expressed by both G. mellonella (transferrin and ferritin) and M. mycetomatis (SidA, SidD and SidI). LC-MS/MS analysis of M. mycetomatis cultured under iron-limiting conditions revealed the presence of SidA and SidD orthologs and concurrent RP-HPLC and LC-MS identified a singly charged, putative siderophore in culture supernatant. M. mycetomatis could also obtain iron from holoferritin. The identification of the importance of iron acquisition during grain formation can be exploited as a potential novel diagnostic and therapeutic strategy for mycetoma.

Project description:Polymicrobial infections are intrinsically less susceptible to antimicrobial therapy, are more invasive and can induce enhanced inflammatory responses leading to negative clinical outcomes. The importance of studying the interaction between microbes themselves and their interactions with their host in combination has only been realised in recent years. However, there is a lack of simple, effective and low cost in vivo models to study these interactions with the host immune response. This study details the responses of Galleria mellonella larvae to disseminated combination infection by C. albicans and S. aureus and demonstrates the efficacy of antimicrobial chemotherapy on treating co-infected larvae. Doses of C. albicans (1 × 105 larva-1) and S. aureus (1 × 104 larva-1) which were non-lethal in mono-infection, when combined significantly lowered larval survival at (70 ± 3.33% [p < 0.01]), 48 (10 ± 3.33% [p < 0.0001]) and 72h (5 ± 6.66% [p < 0.0001]) h relative to larvae receiving S. aureus 2 × 104 larva-1 mono-infection. Co-infected larvae with S. aureus 2 × 104 larva-1 resulted in an increase in the density of circulating hemocytes relative to S. aureus mono-infected larvae. Co-infected larvae displayed a significantly higher overall S. aureus c.f.u larva-1 compared to larvae only infected with S. aureus. Co-infection resulted in dissemination throughout the host and the appearance of large nodules. Proteomic analysis of co-infected larval hemolymph revealed specific immune responses to bacterial and fungal infection by the increased abundance of a range of antimicrobial peptides, peptidoglycan, lipopolysaccharide and β-glucan recognition proteins and proteins associated with tissue invasion and nodule formation. The in vitro and in vivo efficacy of empirical antimicrobial drugs (amphotericin B & meropenem) was examined against C. albicans and S. aureus was also examined. meropenem was more effective than amphotericin B but a combination was most effective at improving survival of co-infected larvae. The effect of antimicrobial therapy on symptoms associated with co-infection was also assessed using Cryo-imaging. The processes of polymicrobial co-infection in G. mellonella are compared to those in mice models and larvae provide an easy to use and ethically acceptable in vivo system to assess polymicrobial interactions with their host.

Project description:The increasing problem of antifungal drug resistance is a serious concern, making it necessary to find new treatment options. In this study, we evaluated the antifungal activity and molecular effects of biogenic silver nanoparticles (AgNPs) synthesized using the culture filtrate of Epicoccum nigrum against Fusarium keratoplasticum, a fungal genera with high resistance to antifungal drugs. The AgNPs showed strong antifungal activity, with a MIC50 of 1.79 μg/mL, leading to 92.85% inhibition of fungal growth at the highest concentration tested (5.92 μg/mL). To understand how AgNPs affect F. keratoplasticum, we performed label-free quantitative proteomic mass spectrometric (LFQ-MS) analysis. We identified 853 proteins, 52 of which were significantly altered in abundance after AgNPs treatment. Principal component analysis (PCA) and hierarchical clustering showed a clear difference between the proteomes of control and AgNP-treated samples. Several proteins related to oxidative stress response, mitochondrial function, and riboflavin biosynthesis were affected. Proteins involved in riboflavin biosynthesis and electron transport decreased in abundance, suggesting that AgNPs disrupt fungal metabolism and energy production. On the other hand, proteins related to oxidative stress response and heat shock increased in abundance, showing that the fungus is under strong stress when exposed to AgNPs. To check the toxicity and antifungal activity of AgNPs in vivo, we used the Galleria mellonella larval model. At a dose of 2.58 mg/kg, AgNPs were not highly toxic, with 90% of the larvae surviving after seven days. Hemocyte density increased after AgNPs treatment, showing that there was some immune activation, but it returned to normal levels after 48 hours. Also, proteomic analysis of G. mellonella hemolymph after AgNPs exposure showed only small changes in protein abundance, mostly in immune response and metabolism, confirming that AgNPs do not cause serious damage to the host. The antifungal effect of AgNPs was also tested in G. mellonella larvae infected with F. keratoplasticum. Infection with 10⁵ conidia/mL caused a 90% mortality rate by day seven in untreated larvae. However, AgNP-treated larvae had a fivefold increase in survival, with 50% surviving by day seven, proving that AgNPs can help fight fungal infections in vivo. These results confirm that biogenic AgNPs are effective antifungal agents that act through oxidative stress, metabolic disruption, and mitochondrial damage in F. keratoplasticum. The combination of proteomic analysis and in vivo experiments provides strong evidence that AgNPs are effective and safe in insect model. Future studies should focus on testing long-term toxicity, and exploring their use in medicine and agriculture to help combat the increasing problem of antifungal resistance.
