Secretome proteomics of Galdieria sulphuraria cultured on glucose, maltose and starch hydrolysate

ABSTRACT: We profiled the extracellular proteome (secretome) of the acidophilic microalga Galdieria sulphuraria (strain SAG 108.79) cultivated under three carbon sources: glucose, maltose and a starch-derived hydrolysate. The dataset contains 15 raw runs (5 biological replicates per condition) together with search/quantification results and experimental-design (SDRF) metadata, enabling re-analysis and cross-condition comparison of secreted proteins.

Project description:Micractinium conductrix SAG 241.80 secretes sugars (especially maltose) at pH 5.7 while very little sugar is found at pH 7.6. Another algae, Chlorella sorokiniana does not secrete sugars at either pH. We evaluated the transcriptional profiles of both species of algae at pH 5.7 and 7.6 to make comparisons in the regulation of potential genes involved in these pathways.

Aloin Protects Against UVB-Induced Apoptosis

Project description:This study comprehensively investigates the photoprotective effects of aloin against UVB-induced damage in HaCaT cells, elucidating its antioxidant capacity and its role in preventing cellular apoptosis. The proteomic effect of 50 μg/mL aloin on HaCaT cell damage induced by UVB was evaluated through comparative protein expression. Compared to control cells, UVB irradiation significantly altered protein expression in HaCaT cells, with 64 proteins upregulated and 236 proteins downregulated. In cells treated with both UVB and aloin, 79 proteins were upregulated and 84 proteins were downregulated compared to the UVB-only group. Proteomic analysis showed that aloin modulated the expression of proteins involved in critical signaling pathways, including PI3K-Akt, p53, TGF-β and pathways in cancer, promoting cell survival. Aloin upregulated proteins associated with cell cycle regula-tion and antioxidant responses, such as CCND3, GSTM4, GNA12, SKIL, YWHAZ, and PKN3 while downregulating pro-apoptotic protein FOXO3.

ZDHHC11-mediated palmitoylation alleviates chondrocyte senescence and serves as a therapeutic target for osteoarthritis

Project description:Osteoarthritis (OA) is a chronic, degenerative whole-joint disorder that interferes with quality of life in older individuals. Here, we report a high expression of ZDHHC11 in articular chondrocytes, which is downregulated in the degenerated cartilage of aged mice and OA patients. ZDHHC11 prevents chondrocyte senescence and fosters cartilage anabolism, culminating in an improved OA phenotype. Mice with Zdhhc11 deletion (Zdhhc11fl/fl) exacerbate OA progression in a destabilized medial meniscus (DMM) model. Specifically, we identify ZDHHC11 as a key palmitoyltransferase, the depletion of which leads to GNB2-dependent E3 ubiquitin ligase-mediated proteasomal degradation of APOD.

Prolonged Maltose-Limited Cultivation of Saccharomyces cerevisiae

Project description:Prolonged cultivation (>25 generations) of Saccharomyces cerevisiae in aerobic, maltose-limited chemostat cultures led to profound physiological changes. Maltose hypersensitivity was observed when cells from prolonged cultivations were suddenly exposed to excess maltose. This substrate hypersensitivity was evident from massive cell lysis and loss of viability. During prolonged cultivation at a fixed specific growth rate, the affinity for the growth-limiting nutrient (i.e., maltose) increased, as evident from a decreasing residual maltose concentration. Furthermore, the capacity of maltose-dependent proton uptake increased up to 2.5-fold during prolonged cultivation. Genome-wide transcriptome analysis showed that the increased maltose transport capacity was not primarily due to increased transcript levels of maltose-permease genes upon prolonged cultivation. We propose that selection for improved substrate affinity (ratio of maximum substrate consumption rate and substrate saturation constant) in maltose-limited cultures leads to selection for cells with an increased capacity for maltose uptake. At the same time, the accumulative nature of maltose-proton symport in S. cerevisiae leads to unrestricted uptake when maltose-adapted cells are exposed to a substrate excess. These changes were retained after isolation of individual cell lines from the chemostat cultures and nonselective cultivation, indicating that mutations were involved. The observed trade-off between substrate affinity and substrate tolerance may be relevant for metabolic engineering and strain selection for utilization of substrates that are taken up by proton symport. SE50/110 and the mutant ΔacrA were each cultivated in triplicates in minimal medium supplemented with maltose or glucose as single carbon source. RNA samples from the biological replicates were taken from the middle of the growth phase of both strains in each maltose and glucose minimal medium, respectively. RNA was isolated and the three replicates were combined for each strain and condition. For each cultivation condition, the data from two arrays (dye swap) were combined to make statistically reliable conclusions.

Comparison of the Streptococcus pneumoniae D39 ccpA Mutant compared to D39 wild-type

Project description:The maltose regulon (mal regulon) has previously been shown to consist of the mal gene cluster (malQP, malXCD and malAR operons) in Streptococcus pneumoniae. In this study, we have further elucidated the complete mal regulon in S. pneumoniae D39 using microarray analyses and β-galactosidase assays. In addition to the mal gene cluster, the complete mal regulon of S. pneumoniae D39 consists of a pullulanase (PulA), a glucosidase (DexB), a glucokinase (RokB), a PTS component (PtsG) and an amylase (AmyA2). Our microarray studies and β-galactosidase assays further showed that the LacI-family transcriptional regulator MalR represses the expression of the mal regulon in the absence of maltose. Furthermore, the role of the pleiotropic transcriptional regulator CcpA in the regulation of the mal regulon in the presence of maltose was also explored. Our microarray analysis with a ΔccpA strain showed that CcpA only represses the expression of the malXCD operon and the pulA gene in the presence of maltose.
