Protein acetylation profiling of Klebsiella pneumoniae wild type and acyT knockout mutant by DIA-based proteomics

ABSTRACT: This dataset contains raw LC–MS/MS data acquired by data-independent acquisition (DIA) from Klebsiella pneumoniae wild type (WT) and acyT knockout (ΔacyT) strains. The study aimed to identify proteins and lysine sites whose acetylation depends on the acyT gene. A total of six biological samples were analyzed (three replicates per genotype).

Coexistence of Klebsiella pneumonia biofilms with Candida albicans in an acute pneumonia

Project description:Coexistence of polymicrobial organisms acted as biofilm communities has become more and more prominent that these interactions lead to an increased resistance against host immune responses and antimicrobial agents, especially bacterial-fungal interactions. Klebsiella pneumoniae and Candida albicans are one of the interesting candidates acted as interkingdom biofilms. K. pneumoniae and C. albicans (KP+CA) presented the interkingdom biofilms with synergistic interaction through molecular analysis and confocal fluorescent imaging with z-stack thickness in vitro experiments. Both of an acute pneumonia model and in vitro neutrophil responses demonstrated that K. pneumoniae infection performed the highest severity when compared to mixed K. pneumoniae and C. albicans and alone C. albicans infections through various biomarkers and the responsiveness of neutrophils. Consequently, this study indicated that polymicrobial with bacterial-Candida infection may less severe than single-bacterial infection.

The mutants analyzed in this study are further described in Huang T.W., Stapleton J.C., Chang H.Y., Tsai S.F., Palsson B.O., Charusanti P. Capsule removal via lambda-Red knockout system perturbs biofilm formation and fimbriae extression in Klesiella pneumoniae MGH 78578 (manuscript submission) A six chip study using total RNA recovered from three separate wild-type cultures and three separate cultures of a capsule deltion mutant of Klebsiella pneumoniae MGH 78578. The capsule gene cluster (KPN_02493 to KPN_02515) was entirely removed in the capsule deletion mutant. Each chip measures the expression level of 5,305 genes from Klebsiella pneumoniae MGH 78578 and the associated five plasmids (pKPN3, pKPN4, pKPN5, pKPN6 and pKPN7) with 50-mer oligo tiling array with 30-mer spacer.

The resistance response of Klebsiella pneumoniae against the antimicrobial peptide PaDBS1R1

Project description:Klebsiella pneumoniae is an antibiotic-resistant bacteria associated with severe infections, which has led to the search for new antimicrobial drugs to face these infections. Antimicrobial peptides (AMPs) are antimicrobials that exert anti-K. pneumoniae activity. Consequently, AMPs have been explored as a therapeutic option. However, similarly to other antimicrobials, K. pneumoniae can develop resistance against AMPs, although it is less frequent. Therefore, understanding the resistance mechanisms developed by K. pneumoniae against AMPs could aid in the design and development of more effective AMPs. This study aimed to identify via a label-free quantitative proteomic approach the resistance mechanisms involved in the resistance response of K. pneumoniae against the AMP PaDBS1R1.

Host Immune Response to Clinical Hypervirulent Klebsiella pneumoniae Pulmonary Infections via Transcriptome Analysis

Project description:Klebsiella pneumoniae is an arising threat to human health. However, host immune responses in response to this bacterium remain to be elucidated. The goal of this study was to identify the dominant host immune responses associated with Klebsiella pneumoniae pulmonary infection. Pulmonary mRNA profiles of 6-8-weeks-old BALB/c mice infected with/without Klebsiella pneumoniae were generated by deep sequencing using Illumina Novaseq 6000. qRT–PCR validation was performed using SYBR Green assays. Using KEGG (Kyoto Encyclopedia of Genes and Genomes) analysis, we identified several immune associated pathways, including complement and coagulation cascades, Toll-like receptor signaling pathway, Rap1 signaling pathway, chemokine signaling pathway, TNF signaling pathway, phagosome and NOD-like receptor signaling pathway, were involved in Klebsiella pneumoniae pulmonary infection. Using ICEPOP (Immune CEll POPulation) analysis, we found that several cell types were involved in the host immune response to Klebsiella pneumoniae pulmonary infection, including dendritic cells, macrophages, monocytes, NK (natural killer) cells, stromal cells. Further, IL-17 chemokines were significantly increased during Klebsiella pneumoniae infection. This study provided evidence for further studying the pathogenic mechanism of Klebsiella pneumoniae pneumonia infection.
