Proteome Changes of Diary Calve Rumen Epithelium from Birth to Postweaning

Project description:Rumen epithelium plays a central role in absorbing, transporting, and metabolizing of short-chain fatty acids. For diary calve, the growth of rumen papillae greatly enhances the rumen surface area to absorb nutrients. However, the molecular mechanism underlying diary calve rumen postnatal development remains rarely understood. Here, we firstly performed a shotgun approach and bioinformatics analyses were used to investigate and compare proteomic profiles of Holstein calve rumen epithelium on day 0, 30, 60 and 90 of age. Then,a total of 4372 proteins were identified, in which we found 852, 342, 164 and 95 differentially expressed proteins (DEPs) between D0 and D30, between D30 and D60, between D60 and D90, respectively. Finally, Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) analyses were performed to provide a comprehensive proteomic landscape of diary calve rumen development at tissue level.To conclude, our data indicated that keratinocyte differentiation, mitochondrion formation, the establishment of urea transport and innate immune system play central roles during rumen epithelium development. BH4 presents an important role in rumen epithelial keratinization. The biological processes of BH4 biosynthesis and molecular function of NADP binding participate in mitochondrial cristae formation. The proposed datasets provide a useful basis for future studies to better comprehend diary calve rumen epithelial development.

Quantitative differences in rumen epithelium proteins in lambs fed wheat, perennial wheat or perennial wheat plus lucerne.

Project description:The rumen is a major the site of digestion and absorption of energy predominantly in the form of short chain fatty acids as well as amino acids and simple carbohydrates. The value of crops such as perennial wheat (PW) for grain and grazing compared to conventional wheat (W), or the addition of lucerne to PW (PWL) is still being determined. The research sought to determine if these diets representing high potassium (K): low sodium (Na) ratio (W, PW and PWL) was associated with changes in the membrane bound proteins that transport nutrients in the rumen epithelium (RE). Information about these proteins and the influence of different diets is scarce due to difficulties developing a procedure to examine them using quantitative proteomics. The research focuses on proteins extracted using an enzymatic procedure (Bond et al. 2019) from the rumen epithelium layer of the sheep’s’ forestomach. A membrane protein enriched fraction was then extracted and prepared for TMT-MS. The comprehensive assignment of proteins (>4000) to cellular location and quantification of differences in protein abundance will progress the annotation and understanding of protein functions in the ruminant genomes.

Effect of sodium butyrate on rumen epithelial cell proliferation

Project description:Promoting rumen development is closely related to the health and efficient growth of ruminants. We investigated the effect of sodium butyrate (SB) on rumen epithelium (RE), and whether the YAP1/TAZ-dependent alteration was involved in the RE developmental process induced by sodium butyrate. In the SB-treated cells, theYAP1/TAZ-dependent changes were not observed. SB increased the expression of genes involved in short-chain fatty acid (SCFA) metabolism, while YAP1/TAZ did not. Thus, SB mediated RE development was not associated with YAP1/TAZ.

Project description:Emerging data has highlighted the importance of short-chain fatty acids (SCFAs), particularly butyrate, in regulating ruminal homeostasis in vivo isolated epithelial cells. However, little is known about other SCFAs like acetate or propionate, and the interaction between rumen microbes and epithelial immunity are rarely reported. Here, we firstly combined infusion of three SCFAs, to study their different roles in ruminal development, antioxidant capacity, barrier functions, and immunity, as well as cross-talk with ruminal microbiome (16S rRNA sequencing data of rumen digesta) and derived transcriptome (RNA-Seq) and metabolism using an in vivo goat model.

Microbiota-host crosstalk mechanism in the newborn and adult rumen at single-cell resolution

Project description:As the unique organ, rumen plays vital roles in providing products for humans, however, the underlying cell composition and interactions with epithelium-attached microbes remain largely unknown. Herein, we performed an integrated analysis in single-cell transcriptome, epithelial microbiome, and metabolome of rumen tissues to explore the differences of microbiota-host crosstalk between newborn and adult cattle models. We found that fewer epithelial cell subtypes and more abundant immune cells (e.g., Th17 cells) in the rumen tissue of adult cattle. Metabolism-related functions and oxidation-reduction process were significantly upregulated in the adult rumen epithelial cell subtypes. The epithelial Desulfovibrio was significantly enriched in the adult cattle. To further clarify the role of Desulfovibrio in host’s oxidation-reduction process, we performed metabolomics analysis of rumen tissues and found that Desulfovibrio showed a high co-occurrence probability with the pyridoxal in the adult cattle compared with newborn ones. The adult rumen epithelial cell subtypes also showed stronger ability of pyridoxal binding. These indicates that Desulfovibrio and pyridoxal likely play important roles in maintaining redox balance in adult rumen. The integrated analysis provides novel insights into the understanding of rumen function and facilitate the future precision improvement of rumen function and milk/meat production in cattle.

Single-cell RNA analyses of cattle ruminal epithelial cells

Project description:Comprehensive analyses of tissues at single-cell level will benefit our understanding of genetic bases for complex traits. Here we present an initial effort of single-cell transcriptomic analyses of cattle ruminal epithelial cells during the rumen development. We obtained 5064 and 1372 cells from Holstein ruminal epithelial cells before and after weaning, respectively. We reported 6 cell types across their temporal and spatial distributions, which were partially correlated with rumen epithelium layer’s structures and functions. We also reported a distinct sets of cell markers for these cell types, for example, CRA1, HMMR, MKI67, and EZH2 for the dividing epithelial cells and the TGFB pathway and the keratin gene family for keratinized epithelial cells. Our proposed a cell lineage model may contribute to the understanding of cattle rumen epithelial proliferation and development.
