Project description:Brown adipocytes (BAs) are a potential therapeutic cell source for the treatment of metabolic disease such as type 2 diabetes. In this report, human pluripotent stem cells (hPSCs) are subject to directed differentiation to brown dipocytes through a paraxial mesoderm intermediate at high-efficiency. RNA-Seq and ATAC-seq was performed to characterized hPSCs derived paraxial mesoderm and brown adipocytes generated in this study.

Paraxial mesoderm organoids model development of human somites

Project description:During the development of the vertebrate embryo, segmented structures called somites are periodically formed from the presomitic mesoderm (PSM), and give rise to the vertebral column. While somite formation has been studied in several animal models, it is less clear how well this process is conserved in humans. Recent progress has made it possible to study aspects of human paraxial mesoderm development such as the human segmentation clock in vitro using human pluripotent stem cells (hPSCs), however, somite formation has not been observed in these monolayer cultures. Here, we describe the generation of human paraxial mesoderm (PM) organoids from hPSCs (termed Somitoids), which recapitulate the molecular, morphological and functional features of paraxial mesoderm development, including formation of somite-like structures in vitro. Using a quantitative image-based screen, we identify critical parameters such as initial cell number and signaling modulations that reproducibly yielded somite formation in our organoid system. In addition, using single-cell RNA sequencing and 3D imaging, we show that PM organoids both transcriptionally and morphologically resemble their in vivo counterparts and can be differentiated into somite derivatives. Our organoid system is reproducible and scalable, allowing for the systematic and quantitative analysis of human spinal cord development and disease in vitro.

A Single-Cell Transcriptomic Atlas Identifies Hierarchical Skeletal Progenitors and Zonal Patterning in Mouse Mandibular Cartilage

Project description:Skeletal and synovial joint development involves distinct skeletal stem/progenitor populations. However, in neural crest-derived temporomandibular joint (TMJ), skeletal stem/progenitor lineage dynamics and ossification programs remain poorly defined. Here, we performed single-cell RNA sequencing (scRNA-seq) on mouse mandibular cartilage at embryonic day 16.5 (E16.5), postnatal day 0 (P0), and 13 weeks to construct a high-resolution atlas of skeletal stem/progenitor populations. The outer perichondrium lining the mandibular cartilage contained Lgr5⁺Pthlh⁺ cells expressing Dlx5, suggesting intramembranous ossification. In contrast, a previously uncharacterized Crabp1⁺ subpopulation within the inner perichondrium expressed Nrg1, Lsamp, Tac1, and Igfbp5, consistent with early cartilage progenitors biased toward endochondral ossification. These results reveal a spatial division of ossification modes with both intramembranous and endochondral growth and clarify the transcriptional regulation underlying TMJ development. Our findings provide a framework for understanding neural crest–derived skeletal stem/progenitor cell regulation and may inform cartilage regenerative strategies.

Project description:We previously showed that two major cellular components of the human heart, cardiac endothelial cells and cardiomyocytes, could be derived simultaneously from human induced pluripotent stem cells (hiPSCs). In vivo studies in mice have shown these lineages derive from a common MESP1+ mesoderm progenitor. Although this has been investigated using whole transcriptome sequencing, the transcriptional control and dynamics of their segregation from cardiac mesoderm are not yet fully understood. In addition, the same transgenic approach cannot be used in human. Here, we used bulk and single cell RNA sequencing (RNAseq) to investigate EC and cardiomyocytes co-differentiation in hiPSC. We confirmed segregation of these two cardiac lineages from common cardiac mesoderm precursors but more importantly, revealed a critical role of transient expression of the transcription factor ETV2 for endothelial fate specification and showed unexpectedly that functional cardiomyocytes could also originate from ETV2+ progenitors. This dataset contain 10X-based scRNA-seq data human induced pluripotent stem cell derived endothelial cells and 3D cardac microtissues, as well as bulk RNA-seq data of endothelial cells derived from cardiac and paraxial mesoderm.

Global transcriptional characterization of differentiating non-human primate (cynomolgus) iPAX7 iPS cells and comparison with their human counterpart

Project description:Pluripotent stem (PS) cells enable the scalable production of tissue-specific derivatives with therapeutic potential for various clinical applications, including muscular dystrophies. Given the similarity to human counterparts, the non-human primate (NHP) is an ideal preclinical model to evaluate several questions, including delivery, biodistribution and immune response. While the generation of human induced PS (iPS) cell-derived myogenic progenitors is well established, there has been no data for NHP counterparts, probably due to the lack of an efficient system to differentiate NHP iPS cells towards the skeletal muscle lineage. Here we report the generation of three independent Macaca fascicularis iPS cell lines and their myogenic differentiation using PAX7 conditional expression. Whole transcriptome analysis confirmed the successful sequential induction of mesoderm, paraxial mesoderm, and myogenic lineages. NHP myogenic progenitors efficiently gave rise to myotubes under appropriate in vitro differentiation conditions and engrafted in vivo into TA muscles of NSG and FKRP-NSG mice. Lastly, we explored the pre-clinical potential of these NHP myogenic progenitors in a single wild-type NHP recipient, demonstrating engraftment and characterizing the interaction with the host immune response. These studies establish an NHP model system in which to study iPS cell-derived myogenic progenitors.

The prenatal frontal and parietal bones are distinct organs.

Project description:The skull vault is composed of frontal and parietal bones that are connected by flexible sutures that protect the growing brain. Intramembranous ossification in the prenatal skull vault starts by mid-gestation in the mouse, and sutures must remain flexible for normal growth and development. Therefore, the balance of bone formation and remodeling needs to be precisely controlled because premature ossification in the sutures causes craniosynostosis (CS) to develop. CS has a variable clinical presentation where two frontal bones may be fused together, or a frontal bone may be fused to a parietal bone. While most studies focus on the premature suture ossification, we hypothesized that the process of intramembranous ossification in the frontal and parietal bones contributes to the etiology of CS. By bulk RNASeq we identified 536 unique transcripts between the frontal and parietal compartments.Taken together, we propose that the frontal bone is more active in bone remodeling than the parietal bone, and this control is important for temporal onset of intramembranous ossification in the skull vault.
