Effect of human child or adult digestion reproduced in a dynamic in vitro gastrointestinal model on Cryptosporidium parvum transcriptome

Project description:The impact of digestive physicochemical parameters on Cryptosporidium parvum transcriptome was analysed in a human and age-dependent context using a dynamic in vitro gastrointestinal model. The sophisticated TIM-1 in vitro model was used for a comparative analysis of C. parvum gene expression under the digestive conditions encountered in young children (from 6 months to 2 years) or in adults following the simulated ingestion of a glass of water contaminated with C. parvum oocysts. The C. parvum gene expression profile was analysed by RNA-sequencing in the inoculum, as well as the gastric and ileal effluents collected from the TIM-1 system. Results showed that time and compartment of digestion lead to the strongest modification of parasite transcriptome. Accordingly, the highest number of differentially expressed genes was observed in parasite samples collected in child or adult ileal effluents between 120 and 180 min of digestion, where invasive stages (sporozoites) are predominant.

Project description:Vibrio species represent one of the most diverse genera of marine bacteria known for their ubiquitous presence in natural aquatic systems. Several members of this genus including Vibrio harveyi are receiving increasing attention lately because they are becoming a source of health problems, especially for some marine organisms widely used in sea food industry. To learn about adaptation changes triggered by V. harveyi during its long-term persistence at elevated temperatures, we studied adaptation of this marine bacterium in sea water microcosms at 30 oC that closely mimicks the upper limits of sea surface temperatures recorded around the globe.

Project description:For the microarray experiments, 10 g glass wool (Corning Glass Works, Corning, N.Y.) were used to form biofilms (30) in 250 mL in 1 L Erlenmeyer shake flasks which were inoculated with overnight cultures diluted that were 1:100. For EHEC with 7-hydroxyindole and isatin, 1000 mM 7-hydroxyindole in 250 mL DMF, 250 mM isatin in 250 mL DMF, or 250 mL DMF alone were added to cells grown in LB. The cells were shaken at 250 rpm and 30°C for 7 hours to form biofilms on the glass wool, and RNA was isolated from the suspension cells and the biofilm. Keywords: Biofilm study

Daily temperature cycles promote alternative splicing of RNAs encoding SR45a, a splicing regulator in maize

Project description:This was a comparative transcriptome analysis by using high throughput sequencing. To assess the effects of heat stress on maize alternative splicing we used a controlled environment facility called the Enviratron to simulate field conditions. For our experiments, maize plants were subjected to conditions simulating normal diurnal rhythms of light and temperature, with increasing maximal daily temperature (MDT). Maize plants were grown continuously under four different temperature regimes with simulated morning temperatures ramped up over 6 hr to the MDT of 31°C, 33°C, 35°C or 37°C and simulated evening/night time temperatures ramped down over 8 hr to 10°C below the MDT. We tracked the alternative splicing events of maize W22 seedlings grow under different temperatures (MDT of 31°C, 33°C, 35°C or 37°C) to evaluate how different MDTs affect the program of gene alternative splicing in maize. RNA was extracted from small strips of leaf lamina excised from the first fully expanded leaf of V4 and V5 W22 plants (at 20 and 27 DAG, respectively). Plants were sampled in triplicates.
