Project description:Illumina Infinium EPIC HumanMethylation BeadChip data from saliva DNA samples from a healthy elderly cohort with individuals in the age range 70-95 in Southwest Sweden. The cohort was stratified into study groups based on participants´answers to a questionnaire of different lifestyle factors including vitamin supplementations, smoking and drinking habits, physical activity (per year), sun exposure and eating habits. Vitamin D intake was evaluated from the vitamin D supplementation (alone or in a multivitamin complex), dietary vitamin D intake (fish and seafood frequency) and vitamin D synthesis in the skin (sunlight exposure and use of sunscreen). Differential methylation analysis was performed for all the study groups and the combination of different factors with vitamin D supplementation. Gender, age, smoking and alcohol (SD and frequency) were used as covariates in the analyses. Only the study groups referred to the conclusions of the study are shown.

Project description:Background/objectivesOrthorexia nervosa (ON) is characterized by an obsession with rigid dietary rules, which leads to an emphasis being placed on food purity and health. Exploring the prevalence rates and understanding the potential risk factors associated with ON is essential for developing effective prevention and intervention strategies. This study investigated the prevalence of ON and examined different variables in associations to enhance our knowledge of their impact on ON tendency.MethodsA sample of 500 participants, including 357 women and 143 men, aged between 20 and 60, from an Italian university community was recruited to complete an online survey assessing ON, using the 15-item self-report measure ORTO-15, and Mediterranean diet adherence and lifestyle habits using the Mediterranean Diet Adherence Screener (MEDAS) and the Mediterranean Lifestyle Index (MEDLIFE) questionnaires, respectively. Student's t-test, ANOVA, chi-squared test, and multiple linear regressions were used for analyses.ResultsWe found that MEDAS and MEDLIFE scores were statistically higher in males than in females, while the ORTO-15 score was significantly lower in females than in males. In the total, ON prevalence was 19.8% (women = 16.08% and men = 21.28%). Multiple regression analyses on the ORTO-15 score and different variables showed that in our population sample, ON was associated with female sex (β = -2.98; p = 4 × 10-6) and the body mass index (BMI) (β = -0.41; p = 6.71 × 10-7). When adjusting for sex and the BMI, the resulting ORTO-15 score was associated with health science faculty attendance (β = 1.26, p = 0.003), following a food plan (β = -3.14; p = 1 × 10-7) and carrying out physical activity (β = -1.20; p = 0.03).ConclusionsThis study identified the importance of several factors for ON focusing on lifestyle habits that clinicians should consider when assessing patients at risk for eating disorders. Further studies are warranted to better define the diagnostic criteria of ON and develop effective prevention and intervention strategies to promote a healthy relationship with food.

Project description:BackgroundNumerous studies have demonstrated that lifestyle habits and environmental factors influence the incidence and progression of respiratory diseases. However, there is a paucity of similar research conducted in southwest China.ObjectiveThis study aims to investigate the prevalence and primary influencing factors of respiratory diseases among residents in a specific region of southwest China, and to identify vulnerable populations.MethodFrom February 2024 to May 2024, a multi-stage stratified random sampling method was employed in a specific region of southwest China. Three monitoring points were randomly selected from six jurisdictions within this region, resulting in the collection of relevant information from a total of 4,507 residents through offline interviews. Lasso-logistic regression was conducted using R version 4.3.0 to develop a nomogram for estimating disease probabilities. Interaction analysis was performed with gender and age group serving as grouping variables, while other dimensional factors were utilized as analysis variables.ResultA total of 4,507 respondents participated in this study, of whom 956 (21.21%) were identified as sick. The older adult group (>65 years) exhibited the highest prevalence (30.3%). Results from the Lasso-logistic model indicated that current smoking, alcohol abuse, passive smoking, coupled with poor indoor and outdoor environments were significant risk factors. Additionally, a history of respiratory disease, a family history of respiratory issues, negative emotions, and high stress levels may also contribute to the risk of the disease. Protective factors identified include regular exercise, adequate indoor lighting, frequent ventilation, and regular disinfection practices. The nomogram developed in this study demonstrated good discrimination, calibration, and clinical efficacy. Multiplicative interaction analysis indicated that gender and age group exhibited varying degrees of interaction with factors such as smoking, passive smoking, alcohol abuse, regular exercise, household smoke, house disinfection, dust mites, history of respiratory allergies, use of velvet products, and family history of respiratory conditions. Notably, females, adolescents, and the older adult were identified as particularly susceptible and at-risk groups for these interactions.ConclusionThe prevalence of respiratory diseases is notably higher among the permanent population in southwest China. High-risk lifestyles, coupled with poor indoor and outdoor environments, pose particularly significant threats to women, adolescents, and the older adult. Consequently, improving living habits, renovating aging communities, enhancing the quality of the living environment, and prioritizing vulnerable populations remain central to the objectives of primary health services.

Project description:Twenty-two male volunteers aged 30-70 years with a previous diagnosis of Prostatic Intraepithelial Neoplasia (PIN) were recruited by a consultant urologist at Norfolk and Norwich University Hospital NHS Trust. Ethical approval for the trial was obtained from the Norfolk Research Ethics Committee (reference 05/Q0101/9). All participants gave written, informed consent. Volunteers were excluded if they were undergoing chemopreventive therapy, were receiving testosterone replacement medication, had active infection requiring treatment, a BMI <18.5 or >35 or were diagnosed with diabetes. Subjects were allocated into a 12-month, parallel dietary intervention trial consisting of two dietary intervention groups (i) consuming 400g high glucosinolate broccoli per week or (ii) consuming 400g peas per week. Volunteers avoided foods known to contain glucosinolates for 48 hours prior to each biopsy appointment to avoid acute effects. Needle biopsy samples (4 cores) of prostate tissue were obtained by Trans Rectal Ultrasound (TRUS) guided endoscopy at baseline, six and 12 months after the start of the dietary interventions. Additionally, benign and malignant (Gleason score 7-10) tissue samples obtained by Trans Urethral Resection of the Prostate (TURP) were provided from the Partners in Cancer Research Human Tissue Bank at the Norfolk and Norwich University Hospital NHS Trust.

Project description:BackgroundBenign prostatic hyperplasia (BPH) most often occurs in older men; previous studies and clinical experience suggest a potential link between lifestyle habits such as sleep habits, sedentary behavior, exercise levels, and BPH, but whether they have a clear causal relationship and the direction of that causality is unclear. We aimed to investigate the causal relationship between lifestyle habits and BPH using 2-sample Mendelian randomization (MR) analysis.MethodsInstrumental genetic independent variables strongly associated with the selected exposure factors were filtered from published genome-wide association studies (GWAS) consisting primarily of European ancestry samples. GWAS from BPH was analyzed as an MR outcome with the inverse-variance weighted method, maximum likelihood, weighted median method, MR-Egger regression, and several sensitivity analyses, including Cochran's Q test, intercept of MR-Egger, and MR pleiotropy residual sum and outlier test.ResultsMR analysis showed a significant causal risk relationship between sleep duration and BPH, with an odds ratio of 0.42 (95% confidence interval, 0.25-0.69, p = .001) for BPH when sleep duration was increased by 1 standard deviation, but we did not find a causal relationship between the 2 when we performed a reverse analysis. However, sedentary behavior and different levels of exercise did not significantly affect the risk of BPH.ConclusionsThis study showed a strong causal relationship between sleep levels and BPH, with adequate sleep duration being a protective factor for BPH.

Project description:AimsCardiac conduction disease can lead to syncope, heart failure, and death. The only available therapy is pacemaker implantation, with no established prevention strategies. Research to identify modifiable risk factors has been scant.Methods and resultsData from the Cardiovascular Health Study, a population-based cohort study of adults ≥ 65 years with annual 12-lead electrocardiograms obtained over 10 years, were utilized to examine relationships between baseline characteristics, including lifestyle habits, and conduction disease. Of 5050 participants (mean age 73 ± 6 years; 52% women), prevalent conduction disease included 257 with first-degree atrioventricular block, 99 with left anterior fascicular block, 9 with left posterior fascicular block, 193 with right bundle branch block (BBB), 76 with left BBB, and 102 with intraventricular block at baseline. After multivariable adjustment, older age, male sex, a larger body mass index, hypertension, and coronary heart disease were associated with a higher prevalence of conduction disease, whereas White race and more physical activity were associated with a lower prevalence. Over a median follow-up on 7 (interquartile range 1-9) years, 1036 developed incident conduction disease. Older age, male sex, a larger BMI, and diabetes were each associated with incident conduction disease. Of lifestyle habits, more physical activity (hazard ratio 0.91, 95% confidence interval 0.84-0.98, P = 0.017) was associated with a reduced risk, while smoking and alcohol did not exhibit a significant association.ConclusionWhile some difficult to control comorbidities were associated with conduction disease as expected, a readily modifiable lifestyle factor, physical activity, was associated with a lower risk.

Project description:Sedentary lifestyle, chronic disease or microgravity can cause muscle deconditioning that then has an impact on other physiological systems. An example is the nervous system, which is adversely affected by decreased physical activity resulting in increased incidence of neurological problems such as chronic pain. We sought to better understand how this might occur by conducting RNA sequencing experiments on muscle biopsies from human volunteers in a 5-week bed-rest study with an exercise intervention arm. We also used a computational method for examining ligand-receptor interactions between muscle and human dorsal root ganglion (DRG) neurons, the latter of which play a key role in nociception and are generators of signals responsible for chronic pain. We identified 1352 differentially expressed genes (DEGs) in bed rest subjects without an exercise intervention but only 132 DEGs in subjects with the intervention. Thirty-six genes were shared between the exercise and no intervention groups. Among 591 upregulated muscle genes in the no intervention arm, 26 of these were ligands that have receptors that are expressed by human DRG neurons.
