GeLC-MS/MS analysis of cereal aphid saliva

Project description:Saliva was collected from c.40,000 diet reared aphids, pooled and concentrated. Samples were coming from 2 aphid species: Metopolophium dirhodum and Sitobion avenae. Proteins were fractionated using SDS PAGE and visible bands were subjected to in gel digestion. Tryptic peptides were analysised on a Thermo LTQ-FT. Protein identification from the MS/MS data was performed using the TurboSEQUEST algorithm in BioWorks v. 3.2 (Thermo Fisher Scientific) to correlate the data against ACYPIproteins v2.1, the official protein set of the pea aphid genome assembly (33291 predicted protein models; accessed November 2011) available at http://www.aphidbase.com/aphidbase/downloads. The following search parameters were used: precursor-ion mass tolerance of 1.5 Da, fragment ion tolerance of 1.0 Da with methionine oxidation and cysteine carboxyamidomethylation specified as differential modifications and a maximum of two missed cleavage sites allowed. Two filters were applied: XCorr vs. charge state (1, 2, 3 and 4 = 1.50, 2.00, 2.50 and 3.00 respectively) and peptide probability (p<0.001). Matches with multiple unique peptides and a cumulative XCorr >20 are reported. PLoS One accepted: Proteomic profiling of cereal aphid saliva reveals ubiquitous and adaptative secreted proteins. Elevated CO2 and virus infection impacts wheat and aphid metabolism

Project description:INTRODUCTION: The aphid Rhopalosiphum padi L. is a vector of Barley yellow dwarf virus (BYDV) in wheat and other economically important cereal crops. Increased atmospheric CO2 has been shown to alter plant growth and metabolism, enhancing BYDV disease in wheat. However, the biochemical influences on aphid metabolism are not known.OBJECTIVES: This work aims to determine whether altered host-plant quality, influenced by virus infection and elevated CO2, impacts aphid weight and metabolism.METHODS: Untargeted 1H NMR metabolomics coupled with multivariate statistics were employed to profile the metabolism of R. padi reared on virus-infected and non-infected (sham-inoculated) wheat grown under ambient CO2 (aCO2, 400 µmol mol−1) and future, predicted elevated CO2 (eCO2, 650 µmol mol−1) concentrations. Un-colonised wheat was also profiled to observe changes to host-plant quality (i.e., amino acids and sugars).RESULTS: The direct impacts of virus or eCO2 were compared. Virus presence increased aphid weight under aCO2 but decreased weight under eCO2; whilst eCO2 increased non-viruliferous (sham) aphid weight but decreased viruliferous aphid weight. Discriminatory metabolites due to eCO2 were succinate and sucrose (in sham wheat), glucose, choline and betaine (in infected wheat), and threonine, lactate, alanine, GABA, glutamine, glutamate and asparagine (in aphids), irrespective of virus presence. Discriminatory metabolites due to virus presence were alanine, GABA, succinate and betaine (in wheat) and threonine and lactate (in aphids), irrespective of CO2 treatment.CONCLUSION: This study confirms that virus and eCO2 alter host-plant quality, and these differences are reflected by aphid weight and metabolism.

Arabidopsis thaliana gene expression changes upon treatment with green peach aphid saliva

Project description:Myzus persicae (green peach aphid) feeding on Arabidopsis thaliana induces a defense response, quantified as reduced aphid progeny production, in infested leaves but not in other parts of the plant. Similarly, infiltration of aphid saliva into Arabidopsis leaves causes only a local increase in aphid resistance. Further characterization of the defense-eliciting salivary components indicates that Arabidopsis recognizes a proteinaceous elicitor with a size between 3 to 10 kD. Genetic analysis using well-characterized Arabidopsis mutant shows that saliva-induced resistance against M. persicae is independent of the known defense signaling pathways involving salicylic acid, jasmonate, and ethylene. Among 78 Arabidopsis genes that were induced by aphid saliva infiltration, 52 had been identified previously as aphid-induced, but few are responsive to the well-known plant defense signaling molecules salicylic acid and jasmonate. Quantitative PCR analysis confirms expression of saliva-induced genes. In particular, expression of a set of O-methyltransferases, which may be involved in the synthesis of aphid-repellent glucosinolates, was significantly up-regulated by both M. persicae feeding and treatment with aphid saliva. However, this did not correlate with increased production of 4-methoxyindol-3-ylmethylglucosinolate, suggesting that aphid salivary components trigger an Arabidopsis defense response that is independent of this aphid-deterrent glucosinolate.
