Heavy chain repertoires of 5 uninfected mice, 5 acutely infected mice and 5 chronically infected mice were longitudinally sequenced over 80 days. All samples are named according to the following scheme: dpi#_cohort$_R&.fastq.gz . dpi = days post infection = for all samples following the start of infection. Proteins modulated during persistent lymphocytic choriomeningitis virus infection

Project description:The experimental data indicate that during a persistent infection, lymphocytic choriomeningitis virus (LCMV) may both directly or indirectly modulate the regulatory cellular processes and alter the cellular functions that are not critical for the survival, but are needed for the homeostasis in the organism. Two-dimensional differential in-gel electrophoresis (2D-DIGE) and MALDI-TOF MS/MS analyses were used to determine the cellular proteome response of HeLa cell line to persistent LCMV infection. Quantitative analysis revealed 24 differentially abundant proteins, half of which were up-regulated and the rest down-regulated. Functional categorization showed that LCMV-responsive proteins were mainly involved in metabolism, stress and defense responses. Among identified proteins, significant changes were found for peroxiredoxins, family of antioxidant enzymes. Decreased amount of these antioxidant proteins was accompanied with the elevation of ROS content in infected cells.

Project description:CD47 is a ubiquitous cell surface receptor that limits cell clearance by phagocytes that express its counter-receptor signal-regulatory protein-α and directly regulates T cell immunity by interacting with its inhibitory ligand thrombospondin-1. Murine natural killer (NK) cells express higher levels of CD47 than other lymphocytes, but the role of CD47 in regulating NK cell homeostasis and immune function remains unclear. Cd47-/- mice exhibited depletion of NK precursors in bone marrow, but antisense Cd47 knockdown or gene disruption resulted in a dose dependent accumulation of immature and mature NK cells in spleen. Cd47-/- mice were impaired in controlling chronic Clone-13 lymphocytic choriomeningitis virus (LCMV) infection, which was associated with depletion of splenic NK cells and loss of effector cytokine and interferon response gene expression in Cd47-/- NK cells. These data identify CD47 as a cell-intrinsic and systemic regulator of NK cell homeostasis and NK cell responses to viral infection.

Development of live-attenuated lymphocytic choriomeningitis virus vectors for potent alarmin-driven CD8+ T cell induction in tumor immunotherapy

Project description:Effective cancer immunotherapy requires potent tumor antigen-specific effector CD8+ T cell (CTLeff) responses, culminating in an inflammatory activation of the tumor microenvironment. We describe the artificial reengineering of the lymphocytic choriomeningitis virus (LCMV) genome to serve as replication-competent but stably attenuated immunotherapy platform (artLCMV). artLCMV targeted dendritic cells, thereby delivering tumor-associated antigens for efficient CTL induction. Additionally, owing to in vivo spread, artLCMV infected lymphoid tissue stroma expressing interleukin-33, an alarmin released from necrotic cells. Interleukin-33 signals accounted for artLCMV-induced effector CTL responses of higher magnitude and functionality than those induced by replication-deficient LCMV-, adenovirus 5- or vaccinia virus-based vectors. Accordingly, superior immunotherapeutic efficacy of artLCMV in transplantable tumor models depended on interleukin-33 signaling, and massive CTLeff influx was associated with an inflammatory conversion of the tumor. These properties, together with low seroprevalence but excellent immunogenicity of LCMV in humans, suggest artLCMV holds promise as a cancer immunotherapy platform.

Single-cell profiling of the leukemic and non-leukemic immune cell compartments in CD8+ T-cell Large Granular Lymphocytic Leukemia

Project description:We performed a detailed and precise characterisation of the clonally expanded leukemic cells in CD8+ T-cell Large Granular Lymphocytic Leukemia (T-LGLL). By combining scRNA+TCRαβ-seq with bulk RNA sequencing data we showed evidence of a strong antigen-driven immune response that shapes the entire immune cell repertoire in T-LGLL. Our study highlights how the whole immune cell repertoire including the hyperexpanded CD8+ T-LGLL cells, the non-leukemic CD8+ cells, CD4+ cells, and monocytes contribute to the CD8+ T-LGLL disease phenotype.
