Project description:Whole transcriptome sequencing data generated from epidermal samples (suction blister roofs) of healthy female subjects after repetitive irradiation on three consecutive days using a full spectrum solar simulator. RNA was extracted from suction blisters taken from irradiated and control sites on the lower backs of each subject 24 hours after the final irradiation. RNA was cDNA converted and then sequenced on an Illumina HiSeq 2500 (single-end, 50 bp) to an approximate sequencing depth of 100 million reads per sample.

Project description:Purpose: to explore the function and mechanism of skin damage induced by ultraviolet irradiation. The mouse model of UVB irradiation was established. Using miRNA Sequence analysis, the miRNA expression profile of the mouse skin model exposed to UVB radiation and the normal skin mice. GO and Pathway analysis were employed for the prediction of miRNA targets. Results:Compared with normal skin, a total of 23 miRNAs were screened for significantly different expressions. Among them, 7 miRNAs were up-regulated and 16 were down-regulated in the skin wound tissue of mice exposed to UVB irradiation. The differential expression of miRNA is related to a variety of signal transduction pathways, among which mmu-miR-195a-5p and mitogen-activated protein kinase (MAPK) signal pathway is worthy of attention. Conclusion: There was significant difference expression of miRNA in the skin tissue of normal mice and the skin injury induced by UVB irradiation. Differential expression of miRNA can be used in the diagnosis and treatment of UVB-induced acute skin injury.

Project description:The radiation bystander effect has been documented with multiple endpoints in various models. While many of the factors regulating this response are known, the mechanisms are still not well understood, and even less is known about signaling of bystander responses in 3-dimensional (3D) tissues. We have used the EPI-200 3D epidermal tissue model to measure global gene expression changes in bystander tissue in order to gain insight into the molecular pathways involved. We identified 187 genes with significantly altered expression in the region between 250 μm and 1 mm from the site of irradiation four hours after exposure. Gene expression at 250-500 μm and 500-750 μm was similar to that within the first 250 μm, the region that also included the directly irradiated tissue. Gene expression, although still significantly altered, began to return toward control levels in the region between 750 μm and 1 mm, however, suggesting a spatial limit to the bystander signal. The 3D bystander genes were concentrated in gene ontology classifications associated with direct irradiation, such as regulation of cell cycle progression and cell death, as well as categories previously associated with bystander responses, such as extracellular signaling, inflammatory processes, and terminal differentiation.

Project description:Accidents with ionizing radiation (IR) often involve acute high dose exposures that can lead to acute radiation syndrome and late effects. IR can induce genomic lesions, cell death or carcinogenesis. Here, we investigated acute IR-induced cellular genomic signatures at the genome wide level. After exposing the adenocarcinoma cell line A549 to an acute 6 Gy 240 kV X-Ray dose, four surviving clonogenic cells were recovered by minimal dilution and further expanded and analyzed by cytogenetics, chromosome painting and tiling-path array CGH, with the non-irradiated clone0 serving as control. It was found that acute X-ray exposure induced changes in modal chromosome number and specific translocations in the four irradiation surviving clones. Furthermore, clone4 displayed an increased radiosensitivity at D > 5 Gy. Array CGH disclosed unique and recurrent genomic changes, predominantly gains, and disclosed fragile sites FRA3B and FRA16D as preferential regions of genomic alterations in all irradiated clones, which likely relates to irradiation-induced genomic stress. Gene expression analysis revealed a specific profile of 364 genes in clone4, of which p53 pathway genes may contribute to its increased radiosensitivity. IR-induced genomic changes AND fragile site expression highlight the capacity of a single acute radiation exposure to resculpture the genome of tumor cells by inflicting genomic stress. Gene expression in A549 cell line was analysed after exposure to 6Gy X-rays at a dose rate of 1Gy/min.

Project description:Irradiation of the K-rasLA1 mouse model with a fractionated dose of 1.0Gy 56Fe- particles increases the incidence of invasive carcinoma compared to unirradiated controls or those irradiated with an acute dose. Microarray profiling was perfromed on whole lungs from K-rasLA1 mice in order to determine global expression changes in the lung following radiation exposure. RNA was extracted from K-rasLA1 lungs from unirradiated control animals or those irradiated with a fractionated or acute dose of 1.0Gy 56Fe- particles 70 days post-irradiation when lungs are still histologically indistiguishable and only contain benign lesions. 129S2 K-rasLA1 mice were either irradiated with an acute (1.0Gy x1; n = 15) or fractionated (0.2Gy x5; n=13) dose of 1.0Gy 56Fe- particles or left unirradiated (n=13). Animals were euthanized 70 days post-irradiation and RNA was extracted from the whole left lung of each animal.
