Project description:Germline stem cells are unipotent stem cells dedicated to differentiate into gemmates throughout of life. However, spontaneous reprogramming of spermatogonial stem cells (SSCs) in long-term culture indicates the acquirement of pluripotency of germ cells affected by microenvironment.epidermal growth factor (EGF), leukemia inhibitory factor (LIF) and fresh mouse embryonic fibroblast (MEF) feeder are essential for transformation, and addition of 2i further enhanced the pluripotency. Transcriptome analysis revealed that EGF activated RAS signaling pathway and inhibited p38 to initiate transformation, and synergically cooperated with LIF to promote the transformation propensity. The increased efficiency of SSCs spontaneous reprogramming established new avenues for regenerative medicine, animal cloning, and research model for germline fate determination.

LIF-dependent gene expression in human neural stem cell cultures

Project description:Human cortical neural stem cells, provided by Dr. Clive Svendsen (Waisman Center, University of Wisconsin at Madison), were generated from human fetal cortex (9 weeks post conception) and grown as free-floating neurospheres as previously detailed (Wright, et al 2003. Journal of Neurochemistry 86, 179-195). At 15 weeks of growth, 10 ng/ml LIF was added to the cultures. In cases where LIF was withdrawn, neurospheres were collected, medium removed, and neurospheres washed three times with 1% N2 supplemented with 20 ng/ml EGF, and cultured into the same. Total RNA was prepared from a single human neural progenitor cell grown long-term LIF collected at 3 time points: early passage (GSM51360-GSM51362), mid passage (GSM51363-GSM51365), and late passage (GSM51366-GSM51368). Genomic profiling of human spermatogonial stem cells [WGBS]

Project description:To better understand human spermatogonial stem cells (SSCs), we profiled their transciptome and epigenome, which revealed the mechanism how human SSCs regulates their self-renewal versus differentiation dermination, as well as how latent pluripotency is established in human SSCs. Remarkly, we discovered signaling pathways (e.g. LIF, BMP, WNT) that differentially regulated self-renewal vesus differentiation in SSCs. We also discovered that SSCs repress core pluripotent factors (Sox2, Pou5f1 and Nanog) yet activate ancillary factors (e.g. Klf4, Mbd3, Tcf3, Sall4) transcriptionally and epigenetically.

The requirement of leukemia inhibitory factor or epidermal growth factor for pre-implantation embryogenesis

Project description:In this study, we further verified the role of LIF by looking at gene expressions in morula stage embryos through DNA microarray. Our results showed that LIF knockdown affected 373 gene expressions, and after LIF supplementation we found that the epidermal growth factor (EGF) is most affected by LIF expression. In order to observe the correlation between LIF and EGF, the LIF knockdown embryos were supplemented with various growth factors including LIF, EGF, GM-CSF, TGF, and IGF II. Only LIF and EGF caused the rate of blastocyst development to recover from 52% of control significantly to 83% and 93%, respectively, and all of the parameters, such as the diameter of blastocysts, the numbers of blastomeres, and cells in ICM and TE, to mostly be restored. Moreover, EGF knockdown also impaired blastocyst development which was reversed by LIF or EGF supplementation. These data suggest that the two growth factors EGF and LIF can be compensatory to each other during early embryonic development, and one of them is necessary for sustaining the normal development of preimplantation embryos.

Expression data from aged spermatogonial stem cells

Project description:In vitro and in vivo aging of mouse spermatogonial stem cells alters stem cell function based on quantitative spermatogonial stem cell transplantation analyses. We used microarrays to identify differential gene expression in vitro and in vivo aged spermatogonial stem cells to identify potential causes of observed phenotypic differences in aged spermatogonial stem cell function. Spermatogonial stem cells were isolated from young and serial-transplanted aged mouse donors and cultured for short and long periods. Spermatogonial stem cells were isolated from cultures and subjected to microarray analysis to identify differential gene expression.

Stromal Transcriptomics Uncover LIF as a Key Effector in High Tumor Budding Triple-Negative Breast Cancer

Project description:The overexpressed genes in FAP-positive CAFs from high TB-TNBCs included ZNF235, ANKRD30B, SLC26A2, SPDYC, CDKN1C, LIF, and FAM83A. The secreted LIF was found in 41.1% of TNBC cases with high levels detected in CAFs, while 26.2% of cases showed high LIF production in cancer cells. The recombinant LIF significantly promoted breast cancer cell migration and enhanced PD-L1 expression, effects that were attenuated by EC359. This study highlights the correlation of high TB in TNBC with poor prognosis and upregulation of key signaling pathways in CAFs. FAP-positive CAFs overexpress genes like LIF, which promotes cell migration and PD-L1 expression. LIF inhibition reduces these effects, suggesting LIF as a potential therapeutic target in TB-associated TNBC progression and immunotherapy.
