Project description:The goal of this study was to identify mRNAs in spermatozoa that differ between idiopathic infertile men and proven-fertile men. The rationale is that there are many idiopathic infertile or subfertile men, and that semen parameters cannot reliably distinguish idopathic infertile men from fertile men. Spermatozoa contain thousands of RNAs, and the levels of those RNAs may reflect the extent of normal spermatogenesis and sperm maturation. We hypothesized that we could identify differences between two patient populations that would reflect their clinical fertility status.

Project description:EVs are known to contain important cargo of biologically active and regulatory molecules including ncRNAs. EVs in seminal plasma, play an important role in sperm functions , including motility and fertilization. However, less is known about the comprehensive profile of spEV ncRNAs in clinical cohort and if these profiles differ by semen quality status. We assessed sperm-free spEV ncRNA profiles from semen samples of male partners receiving IVF treatment. Men were classified into having normal (n = 59, Status 0) or poor (n = 32, Status 1) semen quality based on WHO semen parameter guidelines.

Maternal ELL3 loss-of-function leads to oocyte aneuploidy and early miscarriage

Project description:Up to one-tenth of women experience miscarriage in their lifetime. Embryonic aneuploidy is a leading cause of infertility, miscarrage and congenital defects. Here, we identify loss-of-function variants of ELL3, a gene encoding a transcription elongation factor, from couples experienced consecutive early spontaneous miscarriages due to embryonic aneuploidy. Maternal ELL3 knockout leads to oocyte aneuploidy. Furthermore, we found that ELL3 localizes to the spindle during meiosis, and that ELL3 depletion in both mouse and human oocytes increases the incidence of meiotic spindle abnormality. Biochemically, ELL3 competes with TPX2 to interact with the microtubule motor KIF11, promoting its ATP utilization. Live imaging analysis shows that ELL3 is essential for promoting spindle elongation rate and driving chromosome movement. Our findings demonstrate that ELL3 loss-of-function variations could lead to oocyte aneuploidy and early miscarriage.

Proteomic analysis of human sperm protamines from men with and without infertility

Project description:The objective of this study was to provide a comprehensive assessment of protamine isoforms and modifications in human sperm with the aim of identifying how protamine modifications/isoforms are altered in men with reduced sperm motility and low sperm count. Sperm samples from 18 men with prior reported pregnancy and normozoospermia 14 men from couples with infertility and asthenozoospermia and 24 men from couples with infertility and oligoasthenoteratozoospermia were included in the study. Proteomic assessment using both top-down and bottom-up LC-MS was performed.

DNA methylation profiling of placental villi from karyotypically normal recurrent miscarriage

Project description:Miscarriage occurs in 15-20% of clinical pregnancies. While chromosomal errors are observed in over 50%, causes of karyotypically normal losses are poorly understood. DNA methylation undergoes reprogramming during development and must be appropriately set to maintain a healthy pregnancy. We hypothesize that aberrant DNA methylation may cause karyotypically normal miscarriage, particularly among women experiencing recurrent miscarriage (RM). DNA methylation in first trimester chorionic villi was assessed in chromosomally normal miscarriages from women with RM (N=33) or isolated miscarriage (M, N=21), and elective terminations (TA, N=16). Differentially methylated candidate loci were identified using the Illumina Infinium HumanMethylation27 BeadChip array by comparing 10 RM to 10 TA samples. Follow up showed a significant increase in methylation in RM and M compared to TA placentae at the CYP1A2 (p=0.002) and AXL (p=0.02) promoters and decrease at the DEFB1 (p=0.008) promoter. Gene function analysis showed an enrichment of imprinted genes (p=9.53E-10) and genes previously associated with RM (p=9.51E-06). DNA methylation was evaluated at 7 imprinted loci using bisulfite pyrosequencing. An increase of outliers at these loci was observed in RM (3.9%) compared to M (0%) and TA (0.9%) (p=0.02), with increased average methylation at the H19/IGF2 ICR1 in M samples (p<0.0001). Altered DNA methylation in the placenta at specific loci, as well as global dysregulation in specific cases, may contribute to or be a consequence of placental insufficiency in karyotypically normal miscarriage. First-trimester placental villi samples from karyotypically normal miscarriages from recurrent miscarriage patients (N, N=10) and chromosomally normal elective terminations (PZET, N=10).

Small noncoding RNA profiles and sperm nuclear basic protein properties in semen of young men living in a high environmental impact area

Project description:We present a pilot study based on a multi-level molecular profiling of semen from 33 healthy young subjects residing in a high (n=18) and low (n=15) polluted area in Italy. Samples were analyzed for spermiogram parameters, protamine/histone ratio, Sperm Nuclear Basic Proteins (SNBP) and DNA interaction, oxidative DNA damage, trace metals in semen, and spermatocyte small-noncoding RNA expression. Although comparable mobility and concentration of sperm cells were observed between samples from the two areas, a higher immature/immune cell concentration characterized the semen of subjects from the high-polluted area. These subjects also showed a lower ratio of protamine to histone and a reduced ability of SNBP to bind and protect DNA from oxidative damage. Trace metal analysis showed an altered ratio of copper to zinc in their semen. In spermatocytes from the high-polluted area, altered levels of 32 microRNAs involved in intraflagellar transport, response to oxidative species, nuclear envelope organization, and lipid metabolism were observed. These miRNA profiles resemble those of spermatocytes undergoing differentiation, including a decrease of miR-23b-3p and let-7f-5p and an increase of miR-34b-3p levels. MicroRNAs expression profiles were coherently related to the sperm concentration in both study groups. In addition, a significant decrease of piRNA levels was also observed. These results support a model that provides new insights into the environmental impact on male germ cells.
