Impacts of perR on oxygen sensitivity, gene expression, and murine infection in Clostridioides difficile 630∆erm

Project description:Clostridioides difficile infection (CDI), characterized by colitis and diarrhea, afflicts approximately half a million people in the United States every year, burdening both individuals and the healthcare system. C. difficile 630Δerm is an erythromycin-sensitive variant of the clinical isolate C. difficile 630 and is commonly used in the C. difficile research community due to its genetic tractability. 630Δerm possesses a point mutation in perR, an autoregulated transcriptional repressor that regulates oxidative stress resistance genes. This point mutation results in a constitutively de-repressed PerR operon in 630Δerm. To address the impacts of perR on phenotypes relevant for oxygen tolerance and relevant to a murine model of CDI, we corrected the point mutant to restore PerR function in 630∆erm (herein, 630∆erm perRWT). We demonstrate that there is no difference in growth between 630Δerm and a 630Δerm perRWT under anaerobic conditions or when exposed to concentrations of O2 that mimic those found near the surface of the colonic epithelium. However, 630∆erm perRWT is more sensitive to ambient oxygen than 630∆erm, which coincides with alterations in expression of a variety of perR-dependent and perR-independent genes. Finally, we show that 630∆erm and 630∆erm perRWT do not differ in their ability to infect and cause disease in a well-established murine model of CDI. Together, these data support the hypothesis that the perR mutation in 630∆erm arose as a result of exposure to ambient oxygen and that the perR mutation in 630∆erm is unlikely to impact CDI-relevant phenotypes in laboratory studies

Analysis of Germ Stem Cell loss in a murine model of ERM deficiency

Project description:ERM knockout mice were produced. Testis develop a Sertoli cell only syndrome, which appears complete by 10 weeks of age in male ERM knockout mice. Three sets of Affymetrix comparisons were made. #1 and #2, Wt or ERM knockout testis from 4 week old males, totaly RNA. This time preceeds any significant histologic difference between the wt or knockout. The results was the finding that only a very specific set or markers for spermatogonial STEM CELLs were found to be greatly reduced in the ERM knockout. #3 and #4. The sertoli cell line TM4 was transduced with a retroviral ERM. The wild type TM4 cell line was compared to this TM$-ERM cell line, with the finding that only a few genes were increased in the TM4-ERM line. #5, #6 and #7. Primary Sertoli cells were purified from 4 week old wild type or ERM knockout or from 10 week old ERM knockout mice and compared. Keywords: other

A piRNA like small RNA interacts with and modulates p-ERM proteins in human somatic cells

Project description:PIWI-interacting RNAs (piRNAs) are thought to silence transposon and gene expression during development. However, the roles of piRNAs in somatic tissues are largely unknown. Here we report the identification of 555 piRNAs in human lung bronchial epithelial (HBE) and non-small cell lung cancer (NSCLC) cell lines, including 295 that don’t exist in databases termed as piRNA-Like sncRNAs or piRNA-Ls. Distinctive piRNA/piRNA-L expression patterns are observed between HBE and NSCLC cells. piRNA-L-163 (piR-L-163), the top down-regulated piRNA-L in NSCLC cells, binds directly to phosphorylated ERM proteins (p-ERM), which is dependent on the central part of UUNNUUUNNUU motif in piR-L-163 and the RRRKPDT element in ERM, and. The piR-L-163/p-ERM interaction is critical for p-ERM’s binding capability to filamentous actin (F-actin) and ERM-binding phosphoprotein 50 (EBP50). Thus, piRNA/piRNA-L may play a regulatory role through direct interaction with proteins in physiological and pathophysiological conditions.
