Project description:Pituitary belongs to a most important endocrine glands which takes part in the regulation of reproductive functions. The proper functioning of this intermediary between the central nervous system and the target tissues of the reproductive system ensures the proper course of the estrous cycle and affects the female's reproductive potential. It is believed that visfatin, a hormone belonging to the adipokine family, may be responsible for the control of reproductive functions in response to the actual metabolic state of female. Herein we verified hypothesis assuming the modulatory effect of visfatin on the anterior pituitary transcriptome during the mid-luteal phase of the oestrous cycle. We analyzed samples obtained from 5 individuals (n=5). After the enzymatic tissue digestion with the use collagenase V and pancreatin cells were divided for two groups: controls [without treatment] and visfatin (100 ng/mL) treated ones. In vitro cell cultures were conducted for 24 hours.

The main regulatory system responsible for the reproductive functions during both the oestrous cycle and pregnancy is the hypothalamic-pituitary-gonadal (HPG) axis. Project description:BackgroundThe pituitary belongs to the most important endocrine glands involved in regulating reproductive functions. The proper functioning of this gland ensures the undisturbed course of the oestrous cycle and affects the female's reproductive potential. It is believed that visfatin, a hormone belonging to the adipokine family, may regulate reproductive functions in response to the female's metabolic state. Herein we verified the hypothesis that suggests a modulatory effect of visfatin on the anterior pituitary transcriptome during the mid-luteal phase of the oestrous cycle.ResultsRNA-seq analysis of the porcine anterior pituitary cells revealed changes in the expression of 202 genes (95 up-regulated and 107 down-regulated in the presence of visfatin, when compared to the non-treated controls), assigned to 318 gene ontology terms. We revealed changes in the frequency of alternative splicing events (235 cases), as well as long noncoding RNA expression (79 cases) in the presence of the adipokine. The identified genes were associated, among others, with reproductive system development, epithelial cell proliferation, positive regulation of cell development, gland morphogenesis and cell chemotaxis.ConclusionsThe obtained results indicate a modulatory influence of visfatin on the regulation of the porcine transcriptome and, in consequence, pituitary physiology during the mid-luteal phase of the oestrous cycle.

Effect of enhanced early life nutrition on anterior pituitary miRNA transcriptome in bull calves.

Project description:The aim of this study was to measure the impact of contrasting feeding regimes in the first 12 wk of life, on the molecular control of anterior pituitary tissue in bull calves. Holstein bull calves were designated to high (HI; n=15) or moderate (MOD; n=12) dietary groups, with diets designed to provoke growth rates of 1.0 and 0.5 kg/day, respectively. At 12 wk of age, all calves were euthanized, and anterior pituitary tissue harvested. RNA was extracted from the anterior pituitary and used for miRNAseq.

Visfatin facilitates esophageal cancer migration by suppressing miR-3613-5p expression and promoting VEZF1/VCAN production

Project description:Esophageal cancer, one of the most prevalent cancers worldwide, frequently exhibits distant metastases. The adipokine visfatin has been implicated in cancer progression and metastasis. However, the mechanisms by which visfatin regulates motility in esophageal cancer remain unclear. Here, we found that visfatin levels are higher in metastatic esophageal cancer patients than in primary esophageal cancer patients. Visfatin stimulation enhances migration and invasion in esophageal cancer cells. miRNA sequencing results revealed that miR-3613-5p controls visfatin-induced cell motility. Visfatin promotes esophageal cancer cell migration by reducing miR-3613-5p expression and subsequently increasing VEZF1/VCAN production. The visfatin/miR-3613-5p axis exhibits a promising target for inhibiting esophageal cancer metastasis.
