Protective efficacy of orally administered, heat-killed Lactobacillus pentosus b240 against influenza A virus

Project description:Influenza A(H1N1)pdm virus caused the first human pandemic of the 21st century. Although various probiotic Lactobacillus species have been shown to have anti-microbial effects against pneumonia-inducing pathogens, the prophylactic efficacy and mechanisms behind their protection remain largely unknown. Here, we evaluated the prophylactic efficacy of heat-killed Lactobacillus pentosus b240 against lethal influenza A(H1N1)pdm virus infection in a mouse model. To further define the protective responses induced by b240, we performed virologic, histopathologic, and transcriptomic analyses on the mouse lungs. Although we did not observe an appreciable effect of b240 on virus growth, cytokine production, or histopathology, gene expressional analysis revealed that oral administration of b240 differentially regulates antiviral gene expression in mouse lungs. Our results unveil the possible mechanisms behind the protection mediated by b240 against influenza virus infection and provide new insights into probiotic therapy.

Metabolic regulation of host defense against influenza virus infection through sensing oxaloacetate

Project description:Metabolic pathways determine cellular fate and function, however, the exact roles of metabolites in host immune responses against virus infection remain undefined. Here, we report that the metabolic pathways of oxaloacetate (OAA) are integrated with antiviral responses and OAA serves as a fundamental “cornerstone” to prime effective host resistance to the infection of influenza virus. MDH1 senses cellular OAA to undergo dimerization, thus functions as a scaffold to recruit the transcription factor ETS2 for phosphorylation by the kinase TAOK1 at serine 313. The phosphorylated ETS2 translocates into the nucleus and sustains the appropriate expression of TBK1 to provide broad-spectrum antiviral activity. OAA deficiency caused by ACLY genetic ablation decreases the antiviral immunity through MDH1–TAOK1–ETS2–TBK1 pathway in vivo, and makes mice more susceptible to lethal H1N1 virus infection. Our results uncover a previously unknown signaling pathway to maintain immune homeostasis by sensing cellular OAA, which links metabolism and antiviral innate immunity.

Ketogenic diet activates protective γδ T cell responses against influenza virus infection

Project description:Influenza A virus (IAV) infection-associated morbidity and mortality are a key global healthcare concern, necessitating the identification of novel therapies capable of reducing the severity of IAV infections. In this study, we show that the consumption of a low-carbohydrate, high-fat ketogenic diet (KD) protects mice from lethal IAV infection and disease. KD feeding resulted in an expansion of γδ T cells in the lung that improved barrier functions, thereby enhancing anti-viral resistance. Expansion of these protective γδ T cells required metabolic adaptation to a ketogenic diet, as neither feeding mice a high-fat high-carbohydrate diet nor providing chemical ketone body substrate that bypasses hepatic ketogenesis protected against infection. Therefore, KD mediated immune-metabolic integration represents a viable avenue towards preventing or alleviating influenza disease.
