Project description:Transcriptional profiling of rat liver after a three days caloric restriction or 3 days starvation. In the present study, we wanted to investigate the expression of multiple genes linked to pathways of energy metabolism. We developed a metabolism dedicated microarray tool: the Mitoligo. This microarray gathers genes linked to the main functions of energy metabolism, such as glycolysis or fatty acid oxidation. This biochip has also been designed to study the expression of genes encoding for all the subunits of proteins constituting the ETC. Eventually, genes coding for protein linked to ROS scavenging systems can also be studied using the Mitoligo. Keywords: Nutritional intervention

Project description:Extracellular vesicles (EVs) are key in intercellular communication, carrying biomolecules like nucleic acids, lipids, and proteins. This study investigated postprandial characteristics and proteomic profiles of plasma-derived large extracellular vesicles (lEVs) in healthy individuals. Twelve participants fasted overnight before baseline assessments. After consuming a controlled isocaloric meal, EVs were isolated for proteomic and flow cytometric analysis. Plasma triacylglyceride (TAG) levels confirmed fasting completion, while protein concentrations in plasma and EVs were monitored for postprandial stability. Proteomic analysis identified upregulated proteins related to transport mechanisms and epithelial/endothelial functions postprandially, indicating potential roles in physiological responses to nutritional intake. Enrichment analyses revealed vesicle-related pathways and immune system processes. Flow cytometry showed increased expression of CD324 on CD9+CD63+CD81+ EVs postprandially, suggesting an epithelial origin. These findings offer insights into postprandial lEV dynamics and their physiological significance, highlighting the need for stringent fasting guidelines in EV studies to account for postprandial effects on EV composition and function.

Project description:Postprandial dyslipidemia is a significant risk factor for atherosclerotic cardiovascular diseases. High-density lipoprotein (HDL)-driven reverse cholesterol transport (RCT) plays a crucial role in clearing postprandial lipids. In this study, we aimed to investigate the role of esculetin, a hypolipidemic substance, in regulating postprandial cholesterol metabolism. We measured postprandial cholesterol levels and conducted metabolomics and transcriptomics studies to understand the effect and underlying mechanism of esculetin on postprandial cholesterol metabolism in mice fed a lipid-rich meal. We found that esculetin significantly elevated the postprandial HDL cholesterol levels and induced alterations in postprandial serum metabolomics and hepatic transcriptomics which involved in the key steps of HDL-driven RCT including lipid metabolism, ABC transporters and bile acid biosynthesis and secretion. Additionally, the ratio and the phagocytic activity of an adipose tissue macrophage subset highly expressing CD36 and Tim4 increased following esculetin administration in mice. CCAAT enhancer-binding protein beta (C/EBPβ), a key transcription factor, was involved in the effect of esculetin on macrophages phagocytosis in ox-LDL-stimulated macrophages cells. Therefore, esculetin appears to elevate postprandial HDL levels and regulate postprandial cholesterol metabolism which are associated with the increased phagocytosis of adipose tissue macrophages and C/EBPβ.

Project description:Dietary interventions can dramatically affect physiological health and organismal lifespan. The degree to which organismal health is improved depends upon genotype and the severity of dietary intervention, but neither the effects of these factors, nor their interaction, have been quantified in an outbred population. Moreover, it is not well understood what physiological changes occur shortly after dietary change and how these may affect the health of an adult population. In this article, we investigated the effect of 6-month exposure of either caloric restriction (CR) or intermittent fasting (IF) on a broad range of physiological traits in 960 1-year old Diversity Outbred mice. We found CR and IF affected distinct aspects of physiology and neither the magnitude nor the direction (beneficial or detrimental) of effects were concordant with the severity of the intervention. In addition to the effects of diet, genetic variation significantly affected 31 of 36 traits (heritabilities ranged from 0.04 to 0.65). We observed significant covariation between many traits that was due to both diet and genetics and quantified these effects with phenotypic and genetic correlations. We genetically mapped 16 diet-independent and 2 diet-dependent significant quantitative trait loci, both of which were associated with cardiac physiology. Collectively, these results demonstrate the degree to which diet and genetics interact to shape the physiological health of adult mice following 6 months of dietary intervention.

Project description:Intermittent fasting has gained increasing popularity globally for its potential benefits for improving health and managing weight, but how it impacts the homeostasis and regeneration of peripheral tissues remains elusive. Here we report that in mice, commonly used intermittent fasting regimens negatively affect hair regeneration by selectively inducing apoptosis in activated hair follicle stem cells (HFSCs). Through manipulating animals' daily feeding schedules and caloric intake, we have found that the impaired hair regeneration is primarily caused by the extended fasting periods within these regimens, rather than a reduction in total caloric intake or alterations in circadian rhythm. As the fasting period lengthens, the severity of the defects worsens. Mechanistically, extended periods of fasting activate the hypothalamic-pituitary-adrenal axis, prompting the release of lipolytic hormones from the adrenal glands into circulation. This triggers rapid lipolysis in dermal adipocytes, elevating levels of free fatty acids within the skin. Consequently, fatty acid oxidation is activated in HFSCs, causing increased cellular oxidative stress and apoptosis. Surgical removal of adrenal glands, genetic inhibition of fatty acid oxidation in HFSCs, or enhancement of HFSCs’ antioxidant capability genetically or pharmaceutically prevents fasting-induced stem cell death. Our study reveals a detrimental effect of intermittent fasting on somatic stem cells and peripheral tissue regeneration, and identifies a close crosstalk between an ancient neuroendocrine mechanism and a critical niche component that functions as a “brake” to halt tissue regeneration when the supply of nutrients becomes unstable.

Project description:In this study, we show that muscle-specific inactivation of FBXW7 elicits striking defects in postprandial glucose metabolism and failure to maintain skeletal muscle mass in adult mice. Further, mice lacking FBXW7 exhibited impaired endurance capacity and exacerbated HFD-induced insulin resistance and postprandial hyperglycemia. At the mechanistic level, RNAseq and quantitative proteomic analysis revealed global effects of FBXW7 deficiency on skeletal muscle transcriptome and proteome. This work illustrates a prominent role of FBXW7 in integrating postprandial nutritional signals to coordinate glucose metabolism and muscle mass maintenance.

Esculetin significantly elevated postprandial HDL cholesterol levels in serum and postprandial bile acid levels in bile, and altered serum metabolomics in mice fed a high-fat meal, indicating esculetin promotes HDL-driven cholesterol excretion after a high-fat meal. Furthermore, esculetin administration in mice led to an increase in the ratio and phagocytic activity of a subset of adipose tissue macrophages (ATMs) expressing high levels of CD36 and Tim4. Inhibition of CD36 by Sulfo-N-succinimidyl oleate (SSO) blocked esculetin-induced elevation of postprandial serum HDL and bile acid levels in bile. Additionally, esculetin demonstrated the ability to increase the uptake of oxidized LDL (ox-LDL) via CD36 in a macrophage cell line, which might involve alteration of the epigenetic landscape controlled by CCAAT enhancer-binding protein beta (C/EBPβ). Esculetin-induced increased uptake of ox-LDL and elevation of CD36 was inhibited in C/EBPβ-deficient cells. A relatively higher expression of C/EBPβ was observed in CD36+ ATMs, and esculetin increased the ratio of C/EBPβ+ CD36+ ATMs in mice fed a lipid-rich meal. Overall, these findings suggest esculetin promotes HDL-mediated postprandial cholesterol excretion by directly binding to C/EBPβ and enhancing CD36-dependent phagocytosis in ATMs.
