Project description:Chronic neuropathic pain is widespread, but individual diagnostic biomarkers are unknown and primate specificity hampers rodent validation tests. Here, we report that microRNA (miR)-sequencing of blood leukocytes from 250 multi-centre pain patients revealed complex regional pain syndrome (CRPS)-related increases in the pain-related microRNA (miR)-21-5p and decreases in the cholinergic-targeted miR-335-5p. Both these miRs were similarly modified in dorsal root ganglia of nerve-injured mice, where long RNA-sequencing detected pain-associated elevation of neurogenesis and inflammation-related transcripts. Moreover, either genomic ablation or intrathecal antisense oligonucleotide neuro-suppression of miR-21a-5p, but not genetic debilitation of B7-H1 lymphocytes alleviated murine pain responses while reducing cellular pathways that are induced in patients’ leukocytes; and a cooperative leukocyte ‘signature’ of 12 miRs including miR-21a-5p and -335-5p predicted 96% of CRPS symptoms severity. Our findings implicate cholinergic-suppressible neuro-inflammation processes in chronic acute pain, suggest pathway-based cooperative validation tests and open unprecedented options for mechanistic studies of chronic pain disorders.

Project description:Complex regional pain syndrome type-I (CRPS-I) is chronic neurological disorder accompanied with devastating pain. Most conventional medical treatments lack effectiveness, making CRPS-I a challenging clinical condition. Electroacupuncture (EA) showed effectiveness in alleviating the pain symptoms of CRPS-I patients. However, the molecular mechanisms underlying EA’s therapeutic effect are still not well understood. Here, we established the rat chronic post-ischemic pain (CPIP) model to mimic CRPS-I and performed repetitive EA on bilateral hind limbs of the CPIP model rats. We then performed RNA-sequencing (RNA-Seq) to study the differences in gene expression, gene networks and molecular pathways in ipsilateral DRGs innervating the hind limb of the CPIP model rats with and without repetitive EA treatment. Our results found that repetitive EA treatment significantly alleviated mechanical allodynia in bilateral hind limbs of CPIP model rats. RNA-Seq analysis indicated that EA modulated the expression of multiple genes and gene networks in the DRGs of CPIP model rats. Further bioinformatics analysis identified the up-regulation of an array of genes involved in biological process such as neutrophil chemotaxis and immune response in the DRGs of CPIP model rats after EA treatment. Thus, these results suggest that EA may alleviate pain response in CPIP model rats via regulating multiple genes. Our work may help to further advance the understandings of the molecular mechanisms underlying EA’s therapeutic effects on CRPS-I and help to identify novel targets for CRPS-I treatment

Project description:Pilocytic astrocytoma (PA) is the most common pediatric brain tumor and driven by aberrant MAPK signaling, typically mediated by BRAF alterations. While five-year overall survival rates exceed 95%, tumor recurrence constitutes a major clinical challenge in incompletely resected tumors despite chemotherapeutic or radiation based therapies. Therefore, we used proteogenomics to discern the biological heterogeneity of PA to improve classification of this tumor entity and identify novel therapeutic targets. Our proteogenomics approach integrates RNA sequencing and LC/MS-based proteomic profiling data from a cohort of 58 confirmed, primary PA samples. An integrative genomics approach was conducted to discern the biological heterogeneity of PA and to identify aberrant pathway activation in these biological subgroups. In summary, pilocytic astrocytomas segregate into two groups where younger patients are significantly associated with Group 1. Importantly, we validate the two distinct biological subgroups in two non-overlapping cohorts. The biological heterogeneity seen here may improve biological classification and reveal novel therapeutic targets specifically useful for non-resectable tumors with high risk of recurrent or progressive disease.

Project description:(Purpose) Biological classification of colorectal cancer (CRC) can help to understand its heterogeneous background. The purpose of this study is to classify CRC based on gene expression profiles using formalin-fixed paraffin-embedded (FFPE) samples and to correlate subgroups of CRC with biological features and clinical outcomes. (Results) CRC was clustered into four subgroups by unsupervised hierarchical clustering method. These subgroups show different biological and clinical features. (Conclusion) Gene expression profiles of CRC using FFPE samples distinguish four subgroups that had different biological features and clinical outcomes. These subgroups may explain heterogeneity of CRC and be useful biomarker for clinical.
