Proteomic identification of organic resideus in the bronze mous excavated from Wushan, Chongqing, China

Project description:The bronze mou as an important bronze artifact was commonly used in Ba-Shu region (now in Chongqing and Sichuan Province) during Spring and Autumn Period and Han Dynasty. Despite of extensive archaeological study of the bronze mou, scientific analysis of bronze mou was less, and no organic residue analysis has been undertaken on the bronze mou. In this study, four bronze mous dated to Han dynasty (206BC-220AD) with organic residues unearthed in three archaeological sites in Wushan, Chongqing were selected to be tested by portable X-ray fluorescent spectrometer (P-XRF) , and then be analyzed by Liquid chromatography Tandem Mass Spectrometry (LC/MS/MS). The alloy compositions results show that all the samples are copper-tin-lead ternary alloy. The proteomic analysis results confirmed that the bronze mou had been used as cooking utensils or containers. At the Tuchengpo Cemetery, soybeans were contained both in bronze mou M32:27 and bronze mou M38:39. There was no useful protein discovered in the bronze mou from the Shennvmiao Cemetery. Proteins of ginkgo and chicken were found in M16:12 at the Gaotangguan Cemetery. It indicates the residents in Wushan, Chongqing had a diversity of food sources around 2000 years ago. Furthermore, this study demonstrates proteomic analysis is highly effective in studying archaeological organic residues.

Mongolia Western HCC

Project description:Mongolia has the worldÃ¢Â€Â™s highest incidence of hepatocellular carcinoma (HCC), with ~100 cases/105 inhabitants. We molecularly characterize the Mongolian (n=192) compared to Western HCCs (n=187) by RNA-seq and WES. Mongolian patients were predominantly younger, female, with HBV-HDV non-cirrhotic livers. Mongolian HCCs had higher rates of protein-coding mutations (121 vs 70 mut /tumor in West), and mutations in known (i.e. APOB) and putative HCC drivers (TSC2). A novel mutational signature (SBS Mongolia) was identified in 25% of cases associated with the carcinogenic DMS genotoxic signature. Two unique molecular classes enriched in HBV-HDV infection, female gender and inflamed tumors are described.

Dairy pastoralism sustained Eastern Eurasian steppe populations for 5000 years

Project description:Through the use of proteomic analysis of 32 dental calculus samples from anceint Mongolia, we show that ruminant dairying was present in Mongolia by at least 3000 B.C.E., over 1500 years prior to previously published work. Excitingly, the earliest site with dairy evidence has been identified as archaeologically Afanasievo, supporting the hypothesis that dairy practices and animals likely entered the Eastern steppe with migrating western steppe populations. Furthermore, at 1200 B.C.E. we detect the first direct evidence for horse milk consumption, demonstrating the Bronze Age origins of equine dairying on the eastern steppe which occurred concomitantly with early evidence for horse bridling and riding in the region. The incorporation of horse milk and riding into early Mongolian subsistence strategies led to dramatic economic and demographic shifts that enabled the formation of the well-known steppe empires.
