Artificial Light at Night Disrupts Circadian and Metabolic Gene Expression in the Green Anole Lizard (Anolis carolinensis): A Transcriptomic Analysis

Project description:Artificial light at night (ALAN) disrupts natural light-dark cycles, posing ecological challenges for wildli in urban areas. Here we investigated the efcts of ALAN on gene expression in the brain, liver, skin, and gonads of green anole lizards (Anolis carolinensis) whose urban populations are increasingly exposed to light pollution. To identify genetic pathways impacted by ALAN exposure we analysed expression of genes associated with circadian and metabolic regulation at midday, midnight and at midnight with artificial light. Difrential expression analysis revealed that clock-related genes (PER1, NR1D1, CRY2) were significantly altered in the brain, liver, and skin following ALAN treatment and genes involved in glucagon regulation (GCG) and lipid metabolism (NOCT) were difrentially expressed in the liver, indicating metabolic disruptions. Skin exhibited unique responses to ALAN suggesting that repair responses may be altered as genes related to cellular processes, such as wound healing, were upregulated under normal light and dark conditions. Our findings also show that ALAN disrupts core circadian genes, impacting physiological processes including hormone regulation, glucose homeostasis, and potentially reproductive cycles. This study provides the first transcriptomic evidence of the efcts of light pollution on green anoles, highlighting the need to preserve natural light cycles in urban habitats. An interactive online database developed for this study allows further exploration of gene expression changes, to promote research on artificial light-polluted environments.

Project description:An increasing proportion of the Earth's surface is illuminated at night. In aquatic ecosystems, artificial light at night (ALAN) may influence microbial communities living in the sediments. These communities are highly diverse and play an important role in the global carbon cycle. We combined field and laboratory experiments using sediments from an agricultural drainage system to examine how ALAN affects communities and alters carbon mineralization. Two identical light infrastructures were installed parallel to a drainage ditch before the start of the experiment. DNA metabarcoding indicated that both sediment communities were similar. After one was lit for five months (July-December 2012) we observed an increase in photoautotroph abundance (diatoms, Cyanobacteria) in ALAN-exposed sediments. In laboratory incubations mimicking summer and winter (six weeks each), communities in sediments that were exposed to ALAN for 1 year (July 2012-June 2013) showed less overall seasonal change compared with ALAN-naive sediments. Nocturnal community respiration was reduced in ALAN-exposed sediments. In long-term exposed summer-sediments, we observed a shift from negative to positive net ecosystem production. Our results indicate ALAN may alter sediment microbial communities over time, with implications for ecosystem-level functions. It may thus have the potential to transform inland waters to nocturnal carbon sinks.

Wavelength-dependent effects of artificial light at night on phytoplankton growth and community structure.

Project description:Artificial light at night (ALAN) is a disruptive form of pollution, impacting physiological and behavioural processes that may scale up to population and community levels. Evidence from terrestrial habitats show that the severity and type of impact depend on the wavelength and intensity of ALAN; however, research on marine organisms is still limited. Here, we experimentally investigated the effect of different ALAN colours on marine primary producers. We tested the effect of green (525 nm), red (624 nm) and broad-spectrum white LED ALAN, compared to a dark control, on the green microalgae Tetraselmis suesica and a diatom assemblage. We show that green ALAN boosted chlorophyll production and abundance in T. suesica. All ALAN wavelengths affected assemblage biomass and diversity, with red and green ALAN having the strongest effects, leading to higher overall abundance and selective dominance of specific diatom species, some known to cause harmful algal blooms. Our findings show that green and red ALAN should be used with caution as alternative LED colours in coastal areas, where there might be a need to strike a balance between the effects of green and red light on marine primary producers with the benefit they appear to bring to other organisms.

Carbon fixation (endogenous sugar) and light-dependent gene expression

Project description:Internal sugar and light specific dependent regulation of leaf gene expression was addressed by changing [CO2] to lower than compensation point [CO2] in combination with light or prolonged darkness. Plants were grown on soil in a 12/12 h light/dark rhythm at 20°C day and night and under normal [CO2]. 5 weeks after germination, the above-ground rosettes of the non-flowering plants were harvested, 12 plants per sample. Plants were harvested 4hrs after the end of night (i) under low (< 50 ppm) [CO2] and 150 µE fluorescent light , (ii) under normal [CO2] and light, and, (iii) under low [CO2] and prolonged darkness. Project description:Abnormal circadian rhythms, including exposure to light at night, are associated with a higher cancer risk and a poorer prognosis, which may be one of the reasons that the incidence of cancer is increasing in individuals subjected to these stresses. However, the molecular or systemic mechanisms involved in tumor growth under artificial illumination stress conditions have not been identified. In fact, the question of whether artificial illumination stress promotes tumor growth at all is still controversial. To identify the possible mechanisms underlying tumor progression related to circadian rhythms, we set up nude mouse xenograft models. Eight-week-old male nude mice (BALBc nu/nu) were injected with 100ul (1x106 cells) of Hela cells suspension at two separate dorsal sites. Mice were randomly caged (5/cage) and subdivided into L/L (24-hour light cycle; circadian rhythm disruption model) and L/D (12-hour light/dark cycle; normal circadian rhythm model) groups. 11days after injection, we sacrificed one L/L mouse and one L/D mouse, tumors were immediately preserved using liquid nitrogen. Total RNA from tumors were isolated using QIAshredder and RNeasy-Mini kits (Qiagen).
