Immunomodulatory Potential of Dapphitkhai and Chanthaleela Thai Traditional Recipes in the Context of COVID-19 Pathogenesis

Project description:This study aimed to investigate the effects of two Thai traditional recipes including Dapphitkhai and Chanthaleela on various immune parameters and signaling pathways in human white blood cells. The gene expression of whole blood stimulation was analyzed through RNA-sequencing and pathway analysis. Chanthaleela and Dapphitkhai demonstrated a significant decrease in IL-8 secretion and an increase in IFN-β secretion, suggesting their potential to modulate these signaling pathways. Gene expression analysis of Chanthaleela and Dapphitkhai at 500 µg/mL revealed distinct patterns and identified differentially expressed genes. Gene ontology analysis highlighted the impact of the recipes on immune responses and cytokine regulation, while pathway analysis identified key pathways related to cytokine-cytokine receptor interaction, chemokine signaling, and infection-related processes. This study provides valuable insights into the effects of Thai traditional recipes on immune parameters and signaling pathways in human white blood cells. The findings suggest potential therapeutic applications of Thai traditional medicine in immune-related disorders and viral infections like COVID-19.

Venomics analysis of Thai Daboia siamensis using integrated omics data

Project description:Snakebite, classified by World Health Organization (WHO) as a neglected tropical disease, causes more than 100,000 deaths and 400,000 injuries per year. Currently, available antivenoms do not bind with strong specificity to target toxins, which means that severe complications can still occur despite treatment. Moreover, the cost of antivenom is expensive. Knowledge of venom compositions is fundamental for producing a specific antivenom that has high effectiveness, low side effects, and ease of manufacture. With advances in mass spectrometry techniques, venom proteomes can now be analyzed in great depth at high efficiency. However, these techniques require genomic and transcriptomic data for interpreting mass spectrometry data. This study aims to establish and incorporate genomics, transcriptomics, and proteomics data to study venomics of Thai venomous snake, Daboia siamensis. More than a hundred proteins that have not been reported as venom components of this snake such as hyaluronidase-1 and Phospholipase B were discovered. The use of conventional reference databases for mass spectrometry analysis can detect only 46% of proteins identified by integrated omics data. Thus, this multi-omics data is necessary for venomics studies. These findings will be valuable not only for antivenom production but also for the development of novel therapeutics.

Large scale comparison of non-human sequences in human sequencing data.

Project description:Several studies have demonstrated that unmapped reads in next generation sequencing data could be used to identify infectious agents or structural variants, but there has been no intensive effort to analyze and classify all non-human sequences found in individual large data sets. To identify commonality in non-human sequences by infectious agents and putative contamination events, we analyzed non-human sequences in 150 genomic sequencing data files from the 1000 Genomes Project and observed that 0.13% of reads on average showed similarities to non-human genomes. We compared results among different sample groups divided based on ethnicities, sequencing centers and enrichment methods (whole genome sequencing vs. exome sequencing) and found that sequencing centers had specific signatures of contaminating genomes as 'time stamps'. We also observed many unmapped reads that falsely indicated contamination because of the high similarity of human sequences to sequences in non-human genome assemblies such as mouse and Nicotiana.
