Metabolomic Profiling of Five Varieties of Wine Lees from Thai Wineries

ABSTRACT: This dataset contains liquid chromatography tandem mass spectrometry data acquired from wine lees of Thai wines produced from Verdelho, Chenin Blanc, Viognier, Cabernet Sauvignon, and Durif grape varieties. The samples were analyzed to investigate their metabolomic composition and to identify bioactive compounds contributing to the antioxidant potential of Thai wine lees.

Grape berry development - System-Level and Granger Network Analysis of Integrated Proteomic and Metabolomic Dynamics Identifies Key Points of Grape Berry Development at the Interface of Primary and Secondary Metabolism

Project description:Grapevine is a popular fruit crop worldwide with essential economic importance. The grape berry undergoes complex biochemical changes from fruit set until ripening. To better understand this dynamic process, we applied mass spectrometry based platforms to analysis the metabolome and proteome of grape berries at 12 developmental stages covering the whole developmental process of grape berries. Primary metabolites involved in central carbon metabolism such as sugars, organic acids and amino acids metabolism together with various bioactive secondary metabolites like flavonols, flavan-3-ols and anthocyanins were annotated and quantified. At the same time, the proteomic analysis revealed the protein dynamics of the developing grape berries. Multivariate statistical analysis of the metabolomic and proteomic data revealed growing trajectories with minor difference indicating that grape berry development is a sequential process resulting in changes in all examined processes. The incorporation of the metabolomic and proteomic results allowed us to schematize representative metabolome and proteome candidates on sugar, glycolysis, TCA cycle, amino acid, phenylpropanoid, flavonoid biosynthetic pathways. The overview of the metabolism dynamics on both protein and metabolite level unveiled the metabolism switch and adjustments during grape berry development.

Identification of antioxidant metabolites from Five Plants of the Polynesian Pharmacopoeia and Cosmetopoeia for skin care

Project description:Oxidative stress contributes to impairment of skin health and aspects in the wound healing process for pathologies such as acne, psoriasis or skin cancer. Five Polynesian medicinal plants, among the most traditionally used for skin care (cosmetopoeia and pharmacopoeia) are studied herein for their antioxidant properties: Calophyllum inophyllum, Gardenia taitensis, Curcuma longa, Cordia subcordata and Ficus prolixa. For this purpose, plant extracts were submitted to in vitro bioassays related to antioxidant properties and their bioactive constituents were identified by metabolomic analytical approach. UHPLC-MS/MS analysis was performed leading to characterize 61 metabolites. Compounds annotated for F. prolixa and C. subcordata were reported for the first time in those indigenous trees. Antioxidant properties were evaluated by TPC, DPPH and FRAP assays. F. prolixa extract was the most active one within antioxidant properties similar to ascorbic acid and showed antioxidant intracellular activity on HaCaT model by AOP1 assay. On-Line-HPLC DPPH allowed the identification of phenolic bioactive compounds. These results highlight the potential of F. prolixa aerial roots as a source of antioxidant for skin care topical applications.

Metabolomic heterogeneity of severe pulmonary arterial hypertension

Project description:Although multiple gene and protein expression have been extensively profiled in human pulmonary arterial hypertension (PAH), the mechanism for the development and progression of pulmonary hypertension remains elusive. Analysis of the global metabolomic heterogeneity within the pulmonary vascular system leads to a better understanding of disease progression. Using a combination of high-throughput liquid-and-gas-chromatography-based mass spectrometry, we showed unbiased metabolomic profiles of disrupted glycolysis, increased TCA cycle, and fatty acid metabolites with altered oxidation pathways in the severe human PAH lung. The results suggest that PAH has specific metabolic pathways contributing to increased ATP synthesis for the vascular remodeling process in severe pulmonary hypertension. These identified metabolites may serve as potential biomarkers for the diagnosis of severe PAH. By profiling metabolomic alterations of the PAH lung, we reveal new pathogenic mechanisms of PAH in its later stage, which may differ from the earlier stage of PAH, opening an avenue of exploration for therapeutics that target metabolic pathway alterations in the progression of PAH. Global profiles were determined in human lung tissue and compared across 11 normal and 12 severe pulmonary arterial hypertension patients. Using a combination of microarray and high-throughput liquid-and-gas-chromatography-based mass spectrometry, we showed unbiased metabolomic profiles of disrupted glycolysis, increased TCA cycle, and fatty acid metabolites with altered oxidation pathways in the severe human PAH lung.

Metabolomic and transcriptomic analysis for rate-limiting metabolic steps in xylose utilization by recombinant Candida utilis

Project description:We previously reported that a recombinant Candida utilis strain expressing a Candida shehatae xylose reductase K275R/N277D, a C. shehatae xylitol dehydrogenase, and xylulokinase from Pichia stipitis produced ethanol from xylose. However, its productivity was low. In this study, metabolomic (CE-TOF MS) and transcriptomic (microarray) analyses were performed to characterize xylose metabolism by the engineered C. utilis and to identify key genetic changes contributing to efficient xylose utilization. Metabolomic analysis revealed that the xylose-fermenting strain accumulated more pentose phosphate pathway intermediates, more NADH, and more glycolytic intermediates upstream of glyceraldehyde 3-phosphate than wild-type. Transcriptomic analysis of the strain grown on xylose indicated a significant increase in expression of genes encoding tricarboxylic acid cycle enzymes, respiratory enzymes, and enzymes involved in ethanol oxidation. To decrease the NADH/NAD+ ratio and increase ethanol yield from the fermentation of xylose, ADH1 encoding NADH-dependent alcohol dehydrogenase was overexpressed. The resultant strain exhibited a 17% increase in ethanol production and a 22% decrease in xylitol accumulation relative to the control.

Investigation of Brazilian grape juice metabolomic profile changes caused by methyl jasmonate pre-harvest treatment

Project description:Grape juice is a major source of potential health-promoting bioactive polyphenols, especially for children and those who do not consume wine. Since the subtropical climate may negatively affect the concentrations of grape polyphenols, especially anthocyanins, elicitors such as methyl jasmonate (MeJa) could be used to promote polyphenol biosynthesis. This work aimed at investigating the impact of MeJa treatment on grape juice produced via a traditional low-cost process from two Vitis labrusca cultivars and in two Brazilian regions. The untargeted LC-MS analytical protocol demonstrated that Isabel Precoce juices strongly benefited from MeJa treatment, especially regarding their anthocyanic profile, regardless of the cultivation region. Known MeJa markers in wine and V. vinifera grapes (flavanols, flavonols and stilbenes) in this experiment had mixed behaviours depending on the region/variety/cultivation. Moreover, it was found that all the detected hydroxycinnamates were influenced by the treatment, especially the concentration of their glucosides, which was increased. Glutathione, 2-S-glutathionyl caftaric acid and indole lactic acid glucoside were identified for the first time as MeJa treatment biomarkers in grape products, indicating a possible positive effect on juice antioxidant properties.
