TIPE regulates TGFB2 expression and induces extracellular M2 polarization in CRC

Project description:Abstract Background: M2 macrophages (M2) play a crucial role in the development of colorectal cancer (CRC). The elevated expression of TIPE in CRC is closely associated with CRC progression; however, the impact of TIPE on M2 polarization remains unclear. Methods: IHC, WB, flow cytometry, qPCR, and IF were used to verify the promoting effect of TIPE on M2 polarization in specimens and in vitro co-culture models. RNA-seq, ELISA, and WB were used to identify the secreted proteins regulated by TIPE and the underlying mechanism. CCK-8 assay, wound healing assay, and transwell invasion assay were used to verify the effect of M2 on CRC. In vivo experiments were used to identify the promoting effect of TIPE on M2 polarization. Results: In this study, we first demonstrated that the abnormal overexpression of TIPE in CRC promotes M2 polarization and is correlated with a poor prognosis for CRC patients. We then found that TIPE activates the P38 MAPK signaling pathway in CRC to regulate TGFΒ2 secretion and, furthermore, induces M2 macrophage polarization. In subcutaneous and lung xenograft models of CRC in nude mice, we found that TIPE regulates TGFΒ2 secretion from human CRC to promote M2 polarization of murine macrophages. Conclusion: These results indicate that TIPE is a potent oncogene for inducing M2 polarization. It is suggested that all cancers with high TIPE expression need to be alert to the production and infiltration of M2 macrophages, which provides a new perspective for cancer treatment.

FSTL1 Promotes Macrophage M2 Polarization and Its Role in Allergic Rhinitis

Project description:Allergic rhinitis (AR) is a prevalent inflammatory condition characterized by an overactive immune response to allergens. Recent studies have highlighted the role of macrophage polarization in modulating immune responses. This study investigates the role of Follistatin-like 1 (FSTL1) in promoting macrophage M2 polarization and its implications in AR. Using in vitro and in vivo models, we demonstrate that FSTL1 significantly enhances the expression of M2 macrophage markers, including Arg1, CD206, and IL-10, while concurrently reducing M1 markers such as iNOS and TNF-α. Furthermore, FSTL1-treated macrophages exhibited increased anti-inflammatory properties, contributing to the attenuation of allergic inflammation in a murine model of AR. Our findings suggest that FSTL1-mediated M2 polarization plays a crucial role in mitigating allergic responses, providing a potential therapeutic target for AR management.

SIX2-Mediated Microglial M2 Polarization and Exosomal miR-3470b Delivery Protect Dopaminergic Neurons in Parkinson's Disease

Project description:Parkinson's disease (PD) is characterized by the progressive loss of dopaminergic neurons, with neuroinflammation playing a critical role in disease progression. The transcription factor SIX2 has been identified as an anti-inflammatory factor in microglial cells; however, the underlying mechanisms remain poorly understood. In this study, we demonstrated that SIX2 overexpression protects dopaminergic cells by promoting the polarization of microglial cells from the pro-inflammatory M1 phenotype to the anti-inflammatory M2 phenotype. Mechanistically, SIX2 upregulates DDIT4 expression by binding to its promoter region. DDIT4, in turn, facilitates microglial M2 polarization by inhibiting mTOR, thereby activating autophagy. Furthermore, SIX2-induced M2-polarized microglial cells secrete exosomes carrying miR-3470b. These exosomes are taken up by dopaminergic neurons, where miR-3470b inhibits GREM1 expression and enhances TGF-β signaling activity. Consequently, SIX2-mediated mechanisms prevent and restore damaged dopaminergic neurons in PD mice. Our findings reveal a novel regulatory mechanism of microglial M2 polarization and provide new insights into PD immunotherapy. Additionally, the discovery of exosome-mediated miR-3470b communication between microglia and dopaminergic neurons offers a theoretical foundation for developing exosome-based miRNA therapies.

Integrin αM promotes macrophage alternative M2 polarization in hyperuricemia-related chronic kidney disease

Project description:Hyperuricemia is an essential risk factor in chronic kidney disease (CKD), while urate-lowering therapy to prevent or delay CKD is controversial. Alternatively activated macrophages in response to local microenvironment play diverse roles in kidney diseases. Here, we aim to investigate whether and how macrophage integrin αM (ITGAM) contributes to hyperuricemia-related CKD. In vivo, we explored dynamic characteristics of renal tissue in C57BL/6J mice with hyperuricemia-related CKD. By incorporating transcriptomics and phosphoproteomics data, we analyzed gene expression profile, hub genes and potential pathways. In vitro, we validated bioinformatic findings under different conditions with interventions corresponding to core nodes. We found that hyperuricemia-related CKD was characterized by elevated serum uric acid levels, impaired renal function, activation of macrophage alternative (M2) polarization, and kidney fibrosis. Integrated bioinformatic analyses revealed Itgam as the potential core gene and was associated with focal adhesion signaling. Notably, we confirmed the upregulated expression of macrophage ITGAM, activated pathway, and macrophage M2 polarization in injured kidneys. In vitro, through silencing Itgam, inhibiting p-FAK or p-AKT1 phosphorylation, and concurrent inhibiting of p-FAK while activating p-AKT1 all contributed to the modulation of macrophage M2 polarization. Our results indicated targeting macrophage ITGAM might be a promising therapeutic approach for preventing CKD.
