Project description:Efficacious stem cell-based therapies for traumatic brain injury (TBI) depend on successful delivery, migration, and engraftment of stem cells to induce neuroprotection. L-myc expressing human neural stem cells (LMNSC008) demonstrate an inherent tropism to injury sites after intranasal (IN) administration. We hypothesize that IN delivered LMNSC008 cells migrate to primary and secondary injury sites and modulate biomarkers associated with neuroprotection and tissue regeneration. To test this, immunocompetent adult female rats received a controlled cortical impact injury (CCI) or sham surgery. LMNSC008 cells or a vehicle (VEH) were administered IN on postoperative days 7, 9, 11, 13, 15, and 17. The distribution and migration of eGFP-expressing LMNSC008 cells were quantified over 1 mm-thick optically cleared (CLARITY) coronal brain sections from TBI and SHAM controls. NSC migration was observed along white matter tracts projecting toward the hippocampus and regions of TBI. ELISA and Nanostring assays revealed a shift in tissue gene expression in LMNSC008 treated rats relative to controls. LMNSC008 treatment reduced expression of genes and pathways involved in inflammatory response, microglial function, and various cytokines and receptors. The data demonstrate a robust proof-of-concept for LMNSC008 therapy for TBI and provides a strong rationale for IN delivery for translation in TBI patients. Efficacy,safety and biodistribution of intranasally delivered adenoviral COVID-19 vaccines in hamsters

Project description:Coronavirus disease 2019 (COVID-19) continues to cause outbreaks around the world despite the vast COVID-19 vaccination programs. Thus, it is important to continue developing better vaccines to protect vulnerable populations. Here, we studied the efficacy, safety, persistence and biodistribution of an adenovirus-based COVID-19 vaccine delivered intranasally. Our results confirmed that mucosal delivery leads to effective protection against infection in hamsters. A comparative study of different adenoviral vectors demonstrated that specifically Ad5-S-3M, expressing the SARS-CoV-2 spike glycoprotein ectodomain with inclusion of three common receptor binding domain mutations, induced a robust humoral immune response and effectively protected hamsters from SARS-CoV-2 infection. In addition, longitudinal analysis of vaccinated hamsters’ tissues demonstrated durable vaccine transgene expression and persistent vector DNA in the nasal turbinates. This persistence may be beneficial in increasing the duration of immunity. Expression of the S-3M did not cause any aberrant changes in the hamster tissues and resulted in very few changes in cellular mRNA expression in target tissues. Safety and biodistribution assessment demonstrated restricted y tissue distribution of intranasally delivered vector and no significant changes in clinical blood chemistry, underlining the safety of the vaccine. These results further support that intranasal delivery is a promising approach for mucosal vaccine administration against respiratory infection pathogens and extend our knowledge of Adeno-vector persistence.

A Molecular Switch for Neuroprotective Astrocyte Reactivity

Project description:The intrinsic mechanisms that regulate neurotoxic versus neuroprotective astrocyte phenotypes and their effects on central nervous system (CNS) degeneration and repair remain poorly understood. Here, we show injured white matter astrocytes differentiate into two distinct C3-positive and C3-negative reactive populations, previously simplified as neurotoxic (A1) and neuroprotective (A2)1,2, which can be further subdivided into unique subpopulations defined by proliferation and differential gene expression signatures. We find the balance of neurotoxic versus neuroprotective astrocytes is regulated by discrete pools of compartmented cAMP derived from soluble adenylyl cyclase (sAC) and show proliferating neuroprotective astrocytes inhibit microglial activation and downstream neurotoxic astrocyte differentiation to promote retinal ganglion cell (RGC) survival. Finally, we report a new, therapeutically tractable viral vector to specifically target optic nerve head astrocytes and show elevating nuclear or depleting cytoplasmic cAMP in reactive astrocytes inhibits deleterious immune cell infiltration and promotes RGC survival after optic nerve injury. Thus, soluble adenylyl cyclase and compartmented, nuclear- and cytoplasmic-localized cAMP in reactive astrocytes act as a molecular switch for neuroprotective astrocyte reactivity that can be targeted to inhibit microglial activation and neurotoxic astrocyte differentiation to therapeutic effect. These data expand upon and define new reactive astrocyte subtypes and represents a novel step towards the development of gliotherapeutics for the treatment of glaucoma and other optic neuropathies.
