Project description:Curcumin is a polyphenolic compound extracted from the turmeric plant (Curcuma longa) and has been extensively studied for its anti-inflammatory and anti-proliferative properties. The safety and effectiveness of curcumin have been thoroughly proven. However, the mechanisms underlying the treatment of osteoarthritis remain unclear. This study aimed to reveal the potential mechanism of curcumin in the treatment of osteoarthritis through metabolomics and transcriptomics. First, we verified the effect of curcumin on inflammatory factors in human articular chondrocytes. Secondly, we used cellular metabolomics to explore the cellular metabolic mechanism of curcumin against osteoarthritis. Third, we evaluated differences in gene expression in human articular chondrocytes by transcriptomics. Finally, to evaluate essential targets and elucidate the underlying mechanisms of curcumin in the treatment of osteoarthritis, we performed a screen for proteins in pathways shared by metabolomics and transcriptomics. Our results showed that curcumin significantly reduced the levels of inflammatory markers, such as IL-β, IL-6, and TNF-α, in human articular chondrocytes. Cellular metabolomics identified 106 differential metabolites, including beta-aminopropionitrile, 3-amino-2-piperidone, pyrrole-2-carboxaldehyde, and various other components. Transcriptome analysis yielded 1050 differential mRNAs. Enrichment analysis showed that differential metabolites and mRNA were significantly enriched in 7 pathways including glycine, serine, and threonine metabolism; interconversion of pentose and glucuronic acid; glycerolipid metabolism; histidine metabolism; mucin type O-glycan biosynthesis; phosphoinositide metabolism; and cysteine and methionine metabolism. A total of 23 key targets were identified to be involved in these pathways. We speculate that curcumin may alleviate osteoarthritis by targeting key proteins involved in glycine, serine, and threonine metabolism, inhibiting pyruvate production, and regulating glycolysis.

Project description:Background: Parkinson’s disease (PD), a severe threat in aging society, is a significant cause of disable. Curcumin, a polyphenol with hydrophobic properties, has been proved to against Parkinson. Our previous study provides an initial understanding of the neuroprotective mechanisms of curcumin in treating Parkinson’s disease. However, further exploration in this area is needed. The present study was designed to investigate the potential mechanism of curcumin for treating PD. Methods: The therapeutic efficacy of curcumin was evaluated using behavioral tests, immunofluorescence of tyrosine hydroxylase (TH). Network pharmacology and transcriptomics predicted the active pathway and bioprocess of curcumin in PD. Activation of the PI3K / AKT signaling pathway was confirmed by quantitative real-time PCR and immunofluorescence. Result: Curcumin restored the dyskinesia and dopaminergic neurons damage of MPTP-induced mice. The results of network pharmacology and transcriptomics showed that curcumin against Parkinson's disease by regulating inflammation, oxidative stress, and aging. The mechanisms of these were associated with activation of PI3K / AKT pathway. Conclusion: In conclusion, the mechanism of curcumin against PD was revealed by our study which lays a foundation for the application of curcumin in treating.

Project description:Curcumin (diferuloylmethane) is the bioactive phenolic compound, and the mechanism by which curcumin exerts its anti-metastatic effects was comprehensive and diverse. Several studies reported the anti-metastasis effect of curcumin by its ability to modulate the epithelial-to-mesenchymal transition (EMT) process in different cancers, and the underlying molecular mechanism is poorly understood. EMT is a highly conserved biological process in which epithelial cells acquire mesenchymal-like characteristics by losing their cell-cell junctions and polarity deviating cellular mechanism towards cancer metastasis triggering cancer cells to escape from a primary site to distant locations causing spread of cancer to the entire system ultimately leading to death. In this perspective, we explored the anti-metastatic potential and mechanism of curcumin on the EMT process by establishing in-vitro EMT model using A549 cells induced by TGF-β1. Our results showed that curcumin inhibited EMT by regulating the expression of crucial mesenchymal markers such as MMP2, vimentin, and N-Cadherin. Besides, the transcriptional analysis revealed that curcumin treatment differentially regulated the expression of 75 genes in the NanoString n-counter platform. Further PPI (Protein-protein interaction) network and clusters analysis of differentially expressed genes (DEGs) revealed their involvement in essential biological processes. Altogether, the analysis gives a comprehensive overview of the anti-metastatic effect of curcumin in inhibiting TGF-β1 induced EMT in A549 cells.

Project description:Expression analysis was performed with human SUP-T1 cell line infected with HIV alone, treated with curcumin alone and infected with HIV in presence of curcumin. The control untreated sample was compared with all the three types of treatments to calculate the fold change. The purpose of the experiment was to identify set of genes that are dys-regulated during HIV infection and shows normal expression when treated with curcumin. This experiment is useful in identifying the mechanism of anti-HIV activity of curcumin at genetic level.

Project description:Botanicals are widely used as dietary supplements and for the prevention and treatment of disease. Despite a long history of use, there is generally little evidence supporting the efficacy and safety of these preparations. Curcumin has been used to treat a myriad of human diseases and is widely advertised and marketed for its ability to improve health, but there is no clear understanding how curcumin interacts with cells and affects cell physiology. D. discoideum is a simple eukaryotic lead system that allows both tractable genetic and biochemical studies. The studies reported here show novel effects of curcumin on cell proliferation and physiology, and a pleiotropic effect on gene transcription. Transcriptome analysis showed that the effect is two-phased with an early transient effect on the transcription of approximately 5% of the genome, and demonstrates that cells respond to curcumin through a variety of previously unknown molecular pathways. This is followed by later unique transcriptional changes and a protein kinase A dependent decrease in catalase A and three superoxide dismutase enzymes. Although this results in an increase in reactive oxygen species (ROS; superoxide and H2O2), the effects of curcumin on transcription do not appear to be the direct result of oxidation. This study opens the door to future explorations of the effect of curcumin on cell physiology.

Project description:In breast cancer (BC) care, radiotherapy is considered an efficient treatment. However, approximately 90% of BC-related deaths are due to the metastatic tumor progression. Then, it is strongly desirable to improve tumors radiosensitivity using molecules with synergistic action. In this study, we developed curcumin-loaded solid nanoparticles (Cur-SLN) in order to increase curcumin bioavailability and evaluated their radiosensitizing ability in comparison to curcumin free (Cur), by using an in vitro approach on BC models. Precisely, here we analyzed the transcriptomic profiles induced by Cur-SLN treatments, in non tumorigenic MCF10A and tumorigenic MCF7 and MDA-MB-231 BC cell lines, highlighting the networks responsible of its radiosensitization ability. Curcumin loaded- SLN can be suggested in future preclinical and clinical studies to test its concomitant use during radiotherapy treatments with the double implications of being a radiosensitizing molecule against cancer cells, with a protective role against IR side effects.

Project description:Curcumin, derived from the rhizome of Curcuma longa is a natural anti-cancer agent and has been shown to inhibit proliferation and survival of tumor cells. Although the anti-cancer effects of curcumin are well established, detailed understanding of the signaling pathways altered by curcumin is still unclear. In this study, we carried out SILAC-based quantitative proteomic analysis of CAL 27 cell line, a HNSCC cell line to investigate tyrosine signaling in response to curcumin. Using high resolution Orbitrap Fusion™ Tribrid™ Fourier transform mass spectrometer, we identified 627 phosphotyrosine sites mapping to 357 proteins. We observed alterations in the level of phosphorylation of 307 sites corresponding to 204 proteins upon curcumin treatment.
