Project description:Grapevines cv Sauvignon Blanc were subjected to different stress regimes by simulating the most common extreme weather events occurring within the current climate change scenario. During spring, just before bud break, the vines were either flooded or kept under normal conditions. Both vines were then split in two groups and put in differnt tunnels, either undergoing a simulated heatwave or not. Berry samples were collected from the four groups of plants (control, only flooded, only heatwave, flooded + heatwave) at different timepoints before, during and after the stress. Transcriptomic analyses were carried out on these samples along with some metabolomic assessments to characterize the response to stress in the different samples and the effect of the combined stresses.

Project description:Wheat is one of the most significant crops in terms of human consumption in the world. In a climate change scenario, extreme weather event such as heatwaves will be more frequent especially during the grain-filling (GF) stage and could affect grain weight and quality of crops. Molecular mechanisms underlying the response to short heat stress (HS) have been widely reported for the hexaploid wheat (Triticum aestivum) but the regulatory heat stress mechanisms in tetraploid durum wheat (Triticum turgidum ssp. durum) remain partially understood. In this work, we performed a transcriptomic analysis of durum wheat grains to HS during early GF to identify key HS response genes and their predicted regulatory networks under glasshouse conditions.

Project description:Extreme weather events, such as heatwaves, are becoming increasingly frequent, long-lasting, and severe as global climate change continues, shaping marine biodiversity patterns worldwide. Increased risk of overheating and mortality across major taxa have been recurrently observed, jeopardizing the sustainability of ecosystem services. Molecular responses of species, which scale up to physiological and population responses, are determinant processes that modulate species sensitivity or tolerance to extreme weather events. Here, we analysed the whole-body proteome of the intertidal ragworm Hediste diversicolor (Müller, 1776), a keystone species in estuarine ecosystems and an emergent blue bio-resource, to long-lasting heatwaves (24 ºC vs 30 ºC for 1 month). We hypothesized that this species is phenotypically plastic and thus able to acclimate to heatwaves by inducing cytoprotective pathways and modulating energy metabolism to enhance its thermal tolerance and maintain survival. To test these hypotheses, worms were exposed to control (24 ºC) and a +6 ºC increase in water temperature (30 ºC) for a month, after which they were collected for whole-body proteomic (after 28 days of exposure) analysis. Concomitantly, fatty acid analysis (after 28 days of exposure) and cumulative survival, upper thermal tolerance limits and wet weight (after 30 days of exposure) were compared between control and heatwave exposed worms.

Project description:Pancreatic ductal adenocarcinoma is an extreme lethal cancer with a poor treatment response to all the current chemotherapies. We generated a library of PDAC organoid lines from resected tumors, five from treatment-naive patients and five from patients who received neoadjuvant FOLFIRINOX (eight cycles). We assessed weather residual cancer cells in the tumor lesions of the patients treated with neoadjuvant FOLFIRINOX developed resistance to the treatment. Further we perfromed transcriptome analysis to investigate: 1- weather organoids recapitualting their corresponding tumor tissue 2- To investigate any modifications in the expression of the genes inflicted by FOLFIRINOX treatment.

Project description:Heat stress is one of the extreme weather disasters potently decreasing cereal plant production. Recent studies have demonstrated that protein degradation and rRNA homeostasis as well as transcription factors are involved in thermoresponse in plants. However, how 5-methylcytosine (m5C) RNA modification contribute to heat stress response in plant remains largely unknown. Herein, we identified OsNSUN2 as the mRNA m5C methyltransferase in rice. The osnsun2 mutants specifically displayed severe phenotypes of temperature- and light-dependent lesion mimic and heat stress hypersensitivity. Heat stress enhanced mRNAs m5C methylation involving photosynthesis and detoxification systems, such as β-OsLCY, OsHO2, OsPAL1, and OsGLYI4, in an OsNSUN2-dependent manner, which then increased the protein synthesis. Consistently, the photosystem of the osnsun2 mutants was vulnerable to high ambient temperature and failed to be repaired under tolerable heat stress. Furthermore, OsNSUN2 mutation reduced photosynthesis efficiency and accumulated excessive reactive oxygen species upon heat treatment. Our findings demonstrate an important mechanism of mRNA m5C-dependent heat acclimation in rice.

Project description:Purpose: To identify nucleosome-depleted regions in response to drought in the kidney bean, a genome-wide chromatin profiling strategy was implemented. A better understanding of these OCRs (Open chromatin regions) will be useful in exploring potential genomic regions for crop improvement. Results: Using a comprehensive FAIRE-Seq approach, nucleosome-free chromatin regions were explored in kidney bean (Phaseolus vulgaris. L) in response to both drought and rewatering stress. In all samples, FAIRE peaks (>50%) were enriched in the promoter regions. The drought samples showed peaks very distinct from control and rewatered samples, which showed peaks predominantly at the transcription start site (TSS). Drought and heat-responsive genes and transcription factors were identified within the flanking region of differential peaks. When drought was compared to control, it showed upregulated differential peaks primarily in the promoter and distal intergenic regions. Conclusion: By analyzing FAIRE we are able to identify distinct genomic regions corresponding to droughts and controls. Upstream regions of genes play a significant role in drought stress response. Our data suggest that promoter regions of >2 kb, downstream regions of <= 300, and distant regulatory regions are predominant during the upregulation of drought stress. It would also be helpful to note that genomic regions vary in response to drought, and distal intergenic regions may also be involved in drought responses. Understanding plant genomes and epigenomics could help us develop varieties that can withstand extreme weather conditions.

Project description:Rapid heating events, such as heatwaves, are becoming more frequent and intense as a result of climate change. Importantly, such extreme weather events can be more important drivers of extirpation and selection than changes in annual or seasonal averages and they pose a particularly large threat to poikilothermic organisms. In this study, we evaluate the thermal stress response of a coldwater adapted fish species, the eastern brook trout (Salvelinus fontinalis), to two successive heatwaves during July and August 2022. We sampled brook trout at eight time points from four streams (N=116 fish) that differed in thermal habitat, sequenced mRNA from gill samples using TagSeq, and quantified expression levels of 32,670 unique transcripts. Multivariate analyses found that overall expression patterns in response to water temperature change were similar among streams. These analyses futher detected groups of genes involved in immune response and oxygen carrier activity that were upregulated and downregulated respectively at higher temperatures. We also detected 43 genes that were differentially expressed at different time points and followed the same expression pattern during the two heatwaves. Of these genes, 42 covaried with water temperature and most (27, 62.8%) exhibited responses that varied by stream. Some of the differentially expressed genes, including heat shock proteins and cold-inducible RNA binding proteins, have been widely linked to temperature responses in experimental studies, whereas other genes we identified have functions that have not been well-studied in relationship to temperature (e.g., collagen organization) or have unknown functions. This study shows the utility of landscape transcriptomic approaches to identify important biological processes governing wild organismal responses to short-term stressors. The results of this study can guide future investigations to identify phenotypic and genetic diversity that contribute to adaptive response to heatwaves and improve predictions of how populations will respond to climate change.

Project description:The impact of global warming on weather patterns raises concerns for agriculture management. Using NGS technologies, we identified the miRNA profile that is expressed in rice cultivar grown at two different temperatures. We observed 118 and 100 miRNAs uniquely expressed at 35°C and 25°C respectively. Potential targets of these miRNAs have been identified. An interesting example, would be mir1863b which targets Os07g46670.3; a homologue of the ERD15 gene in Arabidopsis which is involved in dehydration stress response.
