Project description:Strigolactones (SLs), a class of phytohormones, play a pivotal role in governing critical aspects of plant development, including shoot branching and root development. DWARF53 (D53) is a key repressor in the SL signaling. Nevertheless, its involvement and regulatory mechanisms in cotton fiber development—a representative single-cell model, has remained elusive. In this study, we unveil GhD53 represses the transcription of SL-inducible genes GhFAD3-2 and GhFAD3-4, which are involved in linolenic acid biosynthesis—a stimulant of cell elongation, thus inhibiting cotton fiber elongation. We demonstrate that GhD53's inhibitory influence on cotton fiber elongation is relieved in the presence of SLs, as GhD53 undergoes degradation. This alleviation of GhD53 repression triggers the activation of GhFAD3 expression, promoting fiber cell elongation. Remarkably, GhD53 serves as a dual-function transcription repressor, impeding SLs’ role in fiber elongation. Firstly, as a transcription factor, GhD53 directly binds to GhFAD3 promoters, suppressing their transcription. Secondly, GhD53 physically interacts with GhSLRF, a bHLH-type transcription factor, preventing it from binding to GhFAD3 promoters and repressing its transcription activation activity. GhD53 emerges as a pivotal component in the regulatory network governing SL signaling transduction. Overall, our findings provide novel insights into D53’s role in SL signaling and delineate a comprehensive framework for transmitting SL signals to modulate plant cell elongation behavior.

Project description:Legume GRAS-type transcription factors NSP1 and NSP2 are essential for Rhizobium Nod factor-induced nodulation. Both proteins are considered to be Nod factor response factors regulating gene expression upon symbiotic signalling. However, legume NSP1 and NSP2 can be functionally replaced by non-legume orthologs; including rice (Oryza sativa) OsNSP1 and OsNSP2. This shows that both proteins are functionally conserved in higher plants, suggesting an ancient function that was conserved during evolution. Here we show that NSP1 and NSP2 are indispensable for strigolactone biosynthesis in the legume Medicago truncatula as well as rice. Mutant nsp1-nsp2 plants hardly produce strigolactones. The lack of strigolactone biosynthesis coincides with strongly reduced DWARF27 expression in both species. Rice and Medicago represent distinct phylogenetic lineages that split ~150 million years ago. Therefore we conclude that regulation of strigolactone biosynthesis by NSP1 and NSP2 is an ancestral function conserved in higher plants. Since strigolactone biosynthesis is highly regulated by environmental conditions like phosphate starvation, NSP1 and NSP2 will be important tools in future studies on the molecular mechanisms by which environmental sensing is translated into regulation of strigolactone biosynthesis. As NSP1 and NSP2 are single copy genes in legumes, it implies that a single protein complex fulfills a dual regulatory function of different downstream targets; symbiotic and non-symbiotic, respectively. Sunflower response to drought includes root plasticity to adapt to water availability and reach soil water. We identified genotype-specific responses of root architecture to strigolactone application. This experiment aims at identifying strigolactone responsive pathways in 6 genotypes in order to better understand molecular control of root development in sunflower and therefore its response to drought. The ultimate goal will be to improve sunflower breeding through selection of key drought response genes.-The experiment consisted of 2 repeats of four 12-day-old-plantlets of 6 sunflower genotype SF193 (INRA code: XRQ), SF326 (INRA code: PSC8), SF056 (INRA code: FU), SF306 (INRA code: PAZ2), SF302 (INRA code: PAC2), SF268 (INRA code: RHA266), grown in growth chamber conditions and submitted to a 24-hour-treatment of 100 nM strigolactone analogue rac-GR24 (Chiralix, Nijmegen, Netherland) or not. Growth conditions were 14h light at 23°C and 10h night at 20°C under fluorescent bulbs. Plants were grown in hydroponic boxes containing 20 litres of aerated liquid culture medium (as described in Massonneau et al., 2001 Planta).
